Objective Using a bioreactor to culture reendothelializing homograft bioprosthetic valve (HBV) in order to ultimately form viable tissue in vitro, so as to provide basic material for animal experiment and clinical application. Methods The reendothelializign HBV was placed in a pulse duplicator system (“bioreactor”), designed and fabricated by us. These grown under gradually increasing nutrient media flow and pressure for additional 14 days: 125 ml/min at 30 mm Hg (days 1-4), 250 ml/min at 40 mm?Hg (days 5-7), 500 ml/min at 50 mm Hg (days 8-10), 750 ml/min at 75 mm Hg (days 11 to 14). Throughout the studies, the flow device was placed in an incubator with 95% humidity and 5% carbon dioxide. The morphologic structure was observed and photographed by stereomicroscope with 0.5% silver nitrate (AgNO_3)staining and SEM on the 7th, 10th and 14th day. The retention rate of the ECs overlaid onto the HBV was evaluated with the percentage (%). Results The reendothelization level on the surface of the leaflets was 92% before pulsatile-flow-cultivation. But the rate of ECs retention on the scaffold exposured to pulsatile flow was only 36%, 23% and 11% on the 7th, 10th and 14th day, respectively. Conclusion The retention rate of ECs on the scaffold was low for pulsatile-flow-cultivation in vitro. Problems, including deficiency of mature stress fibers in the endothelial cells from MSCs deficiency of the adherence and growth factors still need improvement.

Objective To use metabonomics method to study the change of the basic materials of month rhythm of wei qi deficiency syndrome; To find the potential markers so as to provides a new way for the essence of the wei qi deficiency syndrome research. Methods Based on the autumnal equinox in lunar calendar month, the beginning of a month (the first day of lunar August), the middle of a month (the 15th day of lunar August), and the end of a month (the 30th day of lunar August) were set as the three days to draw experimental materials. Two weeks before drawing materials, 20 rats were randomly divided into control group and model group, 10 rats in each group. The model rats were modeled by the stimulus of fatigue combined with coldness and hotness. Control group rats received conventional breeding. The rats in the both groups during the three experiments received decollation and the blood was taken at the 12 o’clock at noon. HPLC-MS was used to detect plasma metabolites, and partial least squares were used to make statistical analysis on the data for comparing plasma metabonomics original data of control group and model group. Possible metabolic markers of wei qi deficiency syndrome were explored, and the potential makers of month rhythm change of wei qi deficiency syndrome were deduced. Results Oleamide, phosphatidyl glycerol, cortisol, proline, dimethyl fumarate, and eicosapentaenoic acid may be potential markers of wei qi deficiency syndrome in the beginning of a month. Sphingosine-1-phosphate, malic acid, cortisol, oleamide, carnitine, eicosapentaenoic acid and dimethyl fumarate may be potential markers of wei qi deficiency syndrome in the middle of a month. Cholesteryl acetate, threonine, cortisol, dimethyl fumarate, oleamide, eicosapentaenoic acid and pyroglutamate may be potential markers of wei qi deficiency syndrome in the end of a month. Conclusion Month rhythm change of wei qi deficiency syndrome may be influenced by oleamide, cortisol, eicosapentaenoic acid, dimethyl fumarate, and aconitic acid, and may be closely related to energy metabolism, meanwhile accompanied by regulation of cell, hormone and nerves.

A plane, three-dimensional chamber of pulsatile-flow-cultivation and the liquid store chamber connected by medical silica gel tube were designed and constructed by ourselves according to the design principle. The rotator pump of cardiopulmonary bypass unit was acted as the power source. The mixed gas containing 5% CO2 and 95% air was supplied through the ventilation orifice of the liquid store chamber. The temperature of these components was stabilized by thermostatic waterbath. The test of biomechanics and biological compatibility for the system was carried out by cultures experiment during two weeks. The results of the experiment showed that there was no leak in the pulsatile-flow-cultivation components in which the concentration of CO2 was controlled about 5%+1%, the temperature at 37 +/- 1 degree C, and the value of pH between 6.8 and 7.5. The flow rate of the system could be adjusted exactly between 0.125 L/min and 6.0 L/min. The endothelial cells on the viable homograft valve increased about 10 times after being cultured for 2 weeks. The cultures of cell and mould taken from the leaf and Dacron cloth of homograft valve were reported to be negative. The results of the experiment demonstrated that there was satisfactory homeostasis of these components in effective modeling pulsatile-flow-field for the implantation cells cultured, proliferated, and remodeled under the condition inferior or superior to physiological level in vitro. The system can meet the need for study of pulsatile-flow-cultivation and tissue engineering heart valve constructed in vitro.
