1.Transplantation of bone marrow mesenchymal stem cells in the treatment of bone nonunion following limb fractures:experimental results and conversion applications

BACKGROUND:As bone marrow mesenchymal stem cel s can be differentiated into osteoblasts under certain induction conditions, autologous bone marrow mesenchymal stem cel s can be implanted into the bone nonunion site of bone fracture. This new technology garners increasing attention of orthopedic clinicians.
OBJECTIVE:To summarize the clinical efficacy of transplantation of bone marrow mesenchymal stem cel s in the treatment of bone nonunion of limb fractures.
METHODS:A computer-based search of Foreign Medical Journal Ful-Text Service and CNKI databases was performed for articles related to bone marrow mesenchymal stem cel s for treatment of bone nonunion of limb fractures published from 1998 to 2014 using the keywords of“bone marrow stem cel s (BMSCs), stem cel transplantation (SCT), nonunions, tissue engineering”in English and Chinese, respectively. Literatures with repetitive content and lack of originality were excluded. A total of 36 literatures were obtained for further analysis. RESULTS AND CONCLUSION:Bone marrow mesenchymal stem cel s are transplanted into the end of bone nonunion, and then can be induced to differentiate into osteoblasts to repair bone nonunion and bone defects,
laying a theoretical basis for clinical application. Bone marrow mesenchymal stem cel s can repair bone defects, which provides an effective method and material to promote fracture healing. Transplantation of bone marrow mesenchymal stem cel s is safe and effective for treatment of bone nonunion of limb fracture.
2.Mechanisms of heart failure induced by adriamycin

AIM: To investigate the changes of cardiac function, oxidative stress and apoptosis in myocardium of rat model of heart failure induced by adriamycin (ADR). METHODS: At the end of the study, we observed content of malondialdehyde (MDA), activity of superoxide dismutase (SOD) and apoptosis. Expression of P53 protein and p53 mRNA were measured with immunohistochemical method and RT-PCR, respectively. RESULTS: Our data showed that content of MDA increased, activity of SOD decreased and apoptosis in myocardium happened while cardiac function decreased after ADR treatment. p53 gene expression and P53 protein obviously increased in heart failure group. CONCLUSION: There were oxidative stress and apoptosis occurred significantly in a model of heart failure induced by ADR. p53 gene might play an important role in the apoptosis. Correlation analyses suggested that apoptosis in myocardium is related to oxidative stress in ADR-induced heart failure.
3.Effects of the secondary metabolites of rhizotonia leguminicola on immune functions of immunosuppressed mice

Objective:To study the effects of the secondary metabolites of Rhizotonia leguminicola on cyclophosphamide-induced immunoregulation in immunosuppressed mice.Methods:The immunosuppressed mouse model was established by cyclophosphamide(80 mg/kg) injected into abdominal cavity.The diluent of secondary metabolites of Rhizotonia leguminicola(SW) at the dosage of 8,16 and 32 mg/(kg?d) was intragastrically administered for 11 days respectively.Meanwhile,the normal control group and CTX model control group were established.Blood,thymus and spleen of the mice were collected respectively at 24 h after the last intragastric administration,and the immunological detection was also carried out.Results:CTX was injected constantly into the abdominal cavity at 80 mg/kg for 3 d,which leaded to immunosuppression of the mice.The groups which took SW at 8 and 16 mg/(kg?d) could increase the number of WBC of the mice.Cyclophosphamide induced a decrease of phagocytic activity of the peritoneal macrophages in the mice and a boost of hemolysin level of the peripheral blood,lymphocyte transformation of the spleens and the activity of NK cells,which were especially obvious in the group of SW at 16 mg/(kg?d).Conclusion:The secondary metabolites of Rhizotonia leguminicola have effects of recovery and reinforcement in cyclophosphamide-induced immunoregulation of immunosuppressed mice.

To clarify the meaning, role and significance of the new idea of the modern basic theory of traditional chinese medicine, We clarified its origin and evolution starting from the basic concept of idea in this paper, re-vealed the basic connotation and relationship of ideas, scientific ideas, Chinese medicine ideas, and the new ideas of the modern basic theory of traditional Chinese medicine, and tried to give proper definitions; summarized three roles of scientific ideas. According to this basis, we demonstrated the new idea is the scientific ideas with core spirit run-ning through rational questioning, scientific testing, research and conclusion being accepted three-point line cen-tral interlocking; and have three roles changing the theory of traditional chinese medicine, guiding the innovative research on chinese medicine and elevating the scholars' thoughts. We proposed yin and yang to refer to the hypothesis of body's physiology and pathology and prevention and treatment effects, given an empirical analysis a feasible way of medicine and philosophy separation and yin and yang turning to medicine; displayed the guidance of new idea and to show the scientific connotation and scientific value of the theory of traditional Chinese medicine.
5.Liver protection by Sini decoction in hemorrhagic shock and its mechanism relating to oxygen free radical and nitric oxide

AIM: The work was designed to explore protective effects of a traditional Chinese medicine-sini decoction (SD) on liver in hemorrhagic shock and its mechanism relating to oxygen free radical and nitric oxide. METHODS: Anesthetized Wistar rats were subjected to a hemorrhagic shock protocol for 60 min followed by intravenous injection with normal sodium chloride solution or SD solution. Superoxide dismutase (SOD), malondialdehyde (MDA) and nitric oxide (NO) in liver were examined. The inducible nitric oxide synthase (iNOS) was determined immunohistochemically. RT-polymerase chain reaction (RT-PCR)was used to assay the mRNA, which were corresponding to eNOS (endothelial nitric oxide synthase) and iNOS. 6.An experimental study of influence of Qingwen Baidu decoction on serum procalcitonin and C-reactive protein levels in septic rats

Objective To observe the influence of Qingwen Baidu decoction (QBD) on serum procalcitonin (PCT) and C-reactive protein (CRP) levels in septic rats and study the mechanism of heat-clearing and detoxifying method for treatment of sepsis. Methods Fifty male Sprague-Dawley (SD) rats were randomly divided into control group (n=5), model group (n=25), and heat-clearing and detoxifying experimental group (experimental group, n=20). The septic model was reproduced by intra-peritoneal injection of lipopolysaccharide (LPS, 5 mg/kg). In control group, an equal volume of normal saline was given. After modeling for 2 hours, the heat-clearing and detoxifying experimental group received QBD the first time (composition of the decoction: Gypsum Fibrosum Recens 30 g, Rehmanniae Radix 10 g, Bubali Cornu 15 g, Coptidis Rhizoma 4 g, Gardeniae Fructus 5 g, Platycodonis Radix 5 g, Scutellariae Radix 5 g, Anemarrhenae Rhizoma 5 g, Paeoniae Radix Rubra 5 g, Scrophulariae Radix 5 g, Forsythiae Fructus 5 g, Glycyrrhizae Radix 5 g, Moutan Cortex 5 g, Lophatheri Herba 5 g) by gavage (0.01 mL/g); the rest administration time was 08:00 to 09:00, once a day. The rats in model group were given an equal volume of warm water by gavage. At different time points after modeling, the blood of 5 rats in control group, model group, and experimental group was collected from the abdominal aorta. The serum PCT and CRP levels were tested by the enzyme linked immunosorbent assay (ELISA), and the pathological changes in lung and intestinal tissue were observed under a light microscope. Results Compared with the control group, the PCT level of the model group after modeling for 2 hours was significantly increased (ng/L:332.32±22.85 vs. 70.46±3.18, P<0.01), this situation continued until 72 hours after modeling. CRP level in the comparison between the control and model groups did not have statistical significant difference at 2, 8, and 48 hours after modeling (all P > 0.05). The level of CRP in model group was lower than that of control group at 24 hours and 72 hours after modeling (μg/L:281.34±32.81, 237.84±41.42 vs. 350.09±56.67, P<0.05 and P<0.01). There were no statistically significant differences in PCT levels between model group and experimental group after modeling for 8 hours and 24 hours (both P > 0.05). The PCT level of experimental group was significantly lower than that of model group beginning from 48 hours after modeling (ng/L: 321.57±28.00 vs. 358.12±10.14, P < 0.05), and this situation continued until 72 hours after modeling (ng/L: 269.50±49.10 vs. 347.69±26.90, P <0.05). The CRP level of experimental group was significantly lower than that of model group beginning from 8 hours after modeling (μg/L:232.73±13.29 vs. 335.35±53.78, P<0.05), this statistical significant difference between the two groups persisted until 72 hours after modeling (μg/L:177.31±6.70 vs. 237.84±41.42, P<0.05). Compared to those in the model group, the lung tissue inflammatory cell infiltration, the intestinal mucosal inflammation and interstitial edema were milder in the experimental group. Conclusion Heat-clearing and detoxifying therapy can effectively reduce the serum PCT and CRP levels of septic rats induced by LPS, and it can alleviate the infiltration of inflammatory cells in lung tissues so as to play a role in protection of tissue organ.
7.Primary culture and biological characteristics of bone marrow mesenchymal stem cells from a rabbit

BACKGROUND:Rabbit bone marrow mesenchymal stem cels as a kind of adult stem cels with strong proliferation and multilineage differentiation potential exhibit a tremendous application potential in tissue engineering and biological therapy.
OBJECTIVE:To in vitro culture, proliferate and identify rabbit bone marrow mesenchymal stem cels and to observe cel biological characteristics.
MEHTODS:Bone marrow of rabbits was extracted under sterile conditions to separate bone marrow mesenchymal stem cels using the whole bone marrow adherence method and Percol density gradient centrifugation method. Afterwards, the cels were purified and proliferated using differential adherence method. Morphology and growth pattern of cels were observed under microscope, and expression of cel surface antigen markers was detected by flow cytometry.
RESULTS AND CONCLUSION:Rabbit bone marrow mesenchymal stem cels presented with short adherent time and fast growth. After passage and purification, impurities cel counts were decreased. Primary cels presented with triangular, fusiform and spindly shapes. Passage 5 cels with single shape showed the typical polar swirling growth, and could not express CD34 and CD45, but expressed CD29 and CD44. These findings indicate that the cels cultured using the whole bone marrow adherence method and Percol density gradient centrifugation method possess stem cel characteristics in morphology, surface markers and multilineage differentiation, which have been identified as bone marrow mesenchymal stem cels by flow cytometry.
8.Curcumin ameliorates high glucose-induced dysfunction of vasoconstriction via heme oxygenase-1 and GC pathway

AIM: To explore the protective effect of curcumin on high glucose-induced decrease in contraction of isolated rat aortic rings, and to elucidate its underlying mechanism. METHODS: The thoracic aortic rings with endothelium of male Sprague-Dawley rats were mounted on a bath system. Isometric contractions of aortic rings were measured. HO activity was also evaluated. RESULTS: (1)Four hours after incubated with 44 mmol/L of glucose (high glucose),the vascular contraction responses to phenylephrine (PE) decreased compared to control group (containing 11 mmol/L of glucose). (2)Coincubation with curcumin (3×10~(-11)-3×10~(-10) mol/L) and high glucose,the high glucose-induced decrease in contraction responses to PE of arteries was partly inhibited. (3)Four hours after incubation with curcumin,the HO activity in thoracic aorta increased. ZnPP,an inhibitor of HO-1,completely abrogated the protection effect of curcumin. (4)Methylene blue,an inhibitor of guanylate cyclase (GC),partly abolished the protective effect of curcumin. CONCLUSION: Curcumin prevents the high glucose-induced decrease in contraction responses to PE in intact aortic rings. The mechanism might be mainly involved in the activation of HO-1 and GC.
9.Study on the relationship between job burnout and personality characteristics of prison police

Objective To explore the relationship between job burnout and personality characteristics of prison police,and provide a basis for prevention and intervention of prison police burnout.Methods Using Maslach burnout inventory general survey(MBI-GS) and Catteii the sixteen factor questionnaire(16PF) to investigate 217 prison police in Beijing.Results The overall prevalence of burnout syndrome among prison police was 64.7%.The prevalence of emotional exhaustion(11.09±5.86),depersonalization(6.38±4.81) and diminished personal accomplishment(17.96± 2.07) was 46.5%,25.6%,33% respectively.Compared with the national norm of 16PF,the 10 factors of warmth(t=-2.01,P=0.007),reasoning(t=9.22,P=0.000),dominance(t=6.46,P=0.000),liveliness(t=4.01,P=0.000),social boldness(t=-2.00,P=0.047),sensitivity(t=-7.80,P=0.000),abstractedness (t=-2.89,P=0.004),privateness (t=4.35,P=0.000),perfectionism(t=3.19,P=0.002) were significant differences.The emotional exhaustion was significantly related to emotional stability,dominance,liveliness,social boldness and tension.The depersonalization was significantly related to emotional stability,abstractedness and tension.The diminished personal accomplishment was significantly related to self-reliance.Conclusion Prison police burnout situation is more serious,and personality characteristics has a significant impact on job burnout.
10.Epidemiology and distribution of hepatitis C virus genotype among HIV positive former blood donors and transfusion recipients in Hubei province

ObjectiveTo explore the epidemiologic features and distribution pattems of hepatitis C virus (HCV) genotype infection among HIV positive former blood donors (FBDs) and transfusion recipients in Hubei province.Methods597 serum samples from HIV-positive patients in Hubei were collected and examined for anti-HCV by enzyme-linked immunosorbent assay ( ELISA ).Reverse transcription nested polymerase chain reaction (RT-nested PCR) amplification and DNA sequencing were used to evaluate the HCV core regions.ResultsThe prevalence rates of HCV in HIV positive FBDs and transfusion recipients were 76.5% (205/268) and 57.4% (189/329) respectively.HCV genotypes 1b (92.8%,90/97) and 2a (7.2%,7/97 ) were detected.ConclusionsBlood donation and blood transfusion are the major modes of HIV-HCV co-infection in Hubei province.The prevalence of HCV in HIV positive transfusion recipients is lower than that in HIV positive FBDs.HCV genotypes 1b and 2a are the predominant strains among HIV-positive FBDs and transfusion recipients.
