This paper reports that the protective effects of SOD on the experimental gastric ulcers in the mice and erythrocyte membranes, as well as the 125I-SOD traced examination to be studied. The radioactive 125I labelling SOD is using by Iodogen solid phase method. The results of traced examination demonstrated that 125I-SOD could be absorbed into the blood rapidly,and distributed to various organs. The renal radioactivity is highest. The radioactivity in erythrocytes (peak phase) has been accounting for 18. 2% in the whole blood. The radioactivity of bound form in the plasma is only accounted for 12. 5%. It shows that the 125I-SOD was mostly degradation and deiodination. The action of theexogenous O2 free radicals on the erythrocytes showed that the SOD have been markedly inhibited the peroxide damage of O2on the erythrocyte membranes, which inhibitory effect was increased with the concentration of the SOD raised. The effect of SOD on the IM-HCI induced the gastric ulcers, an inhibitory and protective action occur too. In the group of SOD treatment, the gastric mucosal membranes lipid peroxidation was inhibited and the pathological changes remited.
2.The retrospective study of 653 cases of influenza erupting prevalence

Objective To investigate the clinical characteristics and prevalent features of an influenza erupting prevalence.Methods All of 653 patients with an erupting influenza were enrolled in the study from May 29,2003 to July 3,2003. The data included clinic manifestations and etiology. The retrospective study was used to evaluate the clinical and prevalent features. Results All patients consisted of 392 boys and 261 girls aged 5~17 years(mean 11.5 years),who were all students in middle school or primary school. Most of the patients had no typical symptoms including chill(2.45%),headache(30.00%),tiredness(10.87%)and the aching in whole body(4.44%),whereas fever,faucial inflammation and lymphatic follicular hyperplasia of the faucial posterior wall were the most common among these patients,accounting for 100%,60.03%,64.78% respectively. There were 98 patients whose peripheral leukocyte amounts were increased,most happening on the 6 ~7 th day from the onset(75.51%)following the fever on the 5~6 th day(80.6%). Detection of serum double virus-antigen indicated that influenza virus antibody increased more than 4-fold,as well as parainfluenza virus and adenovirus. Even in minority patients,there were several kinds of virus antibody raised 4-fold at the same time. We applied antivirus allopathy associated with the Chinese traditional medicine such as injection of shuanghuanglian or yuxingcao,plus the effectual antibiotics when necessary . All of the patients were cured and no sequelae. Conclusion Sometimes the influenza is prevalent only in some special crowds and has no typical symptoms such as chill,headache,tiredness and the aching in whole body.The antibiotics should be used in time after the synthetic treatment has no effect,when the patient has the onset for more than 5 days and has the fever for more than 4 days.
3.Isolation, culture and identification of adipose-derived stem cells in guinea pigs

AIM: To isolate and culture the adipose-derived stem cells (ADSCs) from guinea pig so as to create bases for the experimental study on treating sensorineural deafness by ADSCs transplantation. METHODS: The experiment was carried out between December 2005 and March 2006 in the Open and Key Laboratory of Clinical Medicine at Henan Universities. The inguinal fat pads were excised from adult male guinea pig weighing 500-750 g. A great quantity of adhesive and natant blood cells were observed under inserted microscope and phosphate buffer was used to wash natant cells repeatedly. Then the cells were cultured in DMEM with 10% fetal bovine serum. Before passage, the cells were washed by a small quantity of phosphate buffer and then cultured in 0.25% trypsin and 0.02% EDTA (2 mL). Then most of the ADSCs presented cytoplasm recovery and round shape, followed by digestion of 2 mL DMEM with feral bovine serum. Cellular suspension was collected to count the cells that were incubated at the density of 2?104/cm2. The morphology of ADSCs was observed constantly. Growth curve of ADSCs at passages 1, 3, 10 were recorded and detected by MTT. Surface markers of ADSCs were detected by flow cytometry. RESULTS: ①The result of primary culture showed that ADSCs at day 2 began to adhesion and more than 90 % of ADSCs at day 7 were approximately fusiform or fibroblast-shaped.②MTT detection showed that ADSCs had the capability to proliferate successively, especially at passage 1, 2, 3. But ADSCs at passage 10 presented decreasing proliferation. After digestion, the cells at passages 1, 3, 10 experienced the latent period (24-48 hours), logarithmic phase (3-5 days) and growth platform phase.③CD105 and CD44 were positive by flow cytometry, whereas CD34 was negative. CONCLUSION: Due to stable growth and rapid proliferation, the ADSCs from guinea pig can be used as the donor cells in the treatment of sensorineural deafness by stem cell transplantation.
4.Effect role of initial planes on the examination of fetal heart by four-dimensional echocardiography with spatiotemporal image correlation

Objective To investigate whether different initial planes have an effect on examination of the fetal heart by four-dimensional ultrasound with spatiotemporal image correlation (STIC).Methods The standard examination planes of fetal heart were rendered using the heart volume data which were acquired in different initial planes with STIC,then these rendered planes were compared and scored.Results① At the four-chamber initial planes,the reconstruction rate of longitudinal planes of the aortic arch (46.27%) and the pulmonary artery (49.25%) were poor,in addition,the reconstruction showed low quality,and the average scores were 1.366 and 1.403,respectively.② At the left outflow-tract initial planes,reconstruction rates of all planes were above 75%,and there was no statistical difference(P=0.169).Each reconstruction plane had good score,even if the scores of longitudinal planes of the aortic arch/pulmonary artery were low,whose scores reached 1.936 and 2.051,respectively.③ At the initial longitudinal planes of the aortic arch,the reconstruction rate (96.67%) and average reconstruction score (2.617) of longitudinal planes of the pulmonary artery were obviously higher than those of other planes.④ Compared with different initial planes,volume data acquired from left outflow-tract initial planes could lead to high-quality reconstructed planes of all standard examination planes.While four-chamber initial planes acquired high-quality standard examination planes except longitudinal planes of the aortic arch and pulmonary artery.Volume data from initial longitudinal plane of the aortic arch acquired low-quality standard examination planes except longitudinal planes of the aortic arch and longitudinal plane of the pulmonary artery.Conclusions With STIC,sonographers can not only save the volume data for off-line analysis and remote consultation but also choose the better initial planes for themselves during the examination of fetal hearts.
5.Study on the relation between the cell cycle regulators and laryngeal carcinogenesis

Objective:To investigate the clinicopathlogical significance of the CyclinE,p21WAF1/CIP1 and p53 expression in laryngeal carcinogenesis.Method:The expression of CyclinE,p21WAF1/CIP1 and p53 was examined by using immunohistochemical method in normal epithlia (20),dysplastic epithlia (40) and laryngeal cancers (60).Result:①In normal epithlium,dysplastic squamous epithlium and in squamous cell carcinoma of the larynx,the positive immunostaining rate of CyclinE protein was 5.0%(1/20),20.0%(8/40) and 45.0%(27/60) respectively (P<0.001),and p21WAF1/CIP1 protein was 95.0%(19/20),75.0%(30/40) and 63.3%(38/60) respectively (P<0.01),while the rate of p53 was 0,30.0%(12/40) and 61.7%(37/60)(P<0.001) respectively.②p21WAF1/CIP1 positive expression in highly、moderately and poorly differtiated carcinomas were 76.2%(16/21)、66.5%(19/29) and 30.0%(3/10) respectively (P<0.05),p21WAF1/CIP1 protein expression was significantly associated with tumour cell differentiation.③The positive expression of CyclinE closely related with the positive expression of p53 (P<0.05).No correlation was found between p21WAF1/CIP1 expression and p53 expression.Conclusion:①CyclinE and p53 could be an early molecular event in the carcinogenesis of larynx.②p21WAF1/CIP1 expression is associated with tumour cell differentiation.③The mutual mechenism of cell cycle regulators (CyclinE、p21WAF1/CIP1 and p53) play an important role in the laryngeal carcinoma.
6.Isolation, culture, identification and labeling of neural stem cells from newborn guinea pigs

BACKGROUND: Transplantation of neural stem cells into inner ear brings hope for treatment of sensorineural deafness, while guinea pig is the preferred animal in developing animal models of aminoglycosides poisoning-induced deafness. Successful animal models have been established after study for several scores of years. How to isolate neural stem cells and label them from hippocampi of guinea pigs is one of the problems to be solved in transplantation of stem cells into inner ear.OBJECTIVE: To isolate and culture neural stem cells from hippocampi of newborn guinea pigs so as to lay basis for transplantation of neural stem cell in the treatment of sensorineural deafness.DESIGN: Single sample observation SETTING: Staff Room of Microbiology and Immunology, Zhengzhou University MATERIALS: This experiment was carried out at the Staff Room of Microbiology and Immunology, Zhengzhou University from March to June 2005. Newbornguinea pigs born within 24 hours of either gender, weighing 50 to 80 g, provided by Experimental Animal Center of Zhengzhou University were used in this experiment.METHODS: Neural stem cells from the hippocampal tissue of newborn guinea pigs were isolated and cultured by serum-free medium containing basic fibroblast growth factor (bFGF) and epidermal growth factor (EGF).Expression of nidogen was detected with immunocytochemical staining.Neural stem cells were labeled by DAPI.MAIN OUTCOME MEASURES: ① In vitro growth of neural stem cells; ② neural stem cells nidogen expression was identified by immunofluorescent staining; ③ labeled stem cells by fluorescence and detected the labeling rate.RESULTS: ①Clone-like cluster of neural stem cells were obtained from hippocampal tissue ② Neural stem cells could express nidogen. ③Labeling rate of DAPI was 93.4% and no attenuation of fluorescence brightness as observed after 8 generations in culture.CONCLUSION: The cells obtained from hippocampal tissues of newborn guinea pigs can be used as donor cells in the treatment of neurosensory deafness by transplanting neural stem cells, for their obvious proliferation ability and high-labeled efficiency with fluorescent dye.
7.Clinical features and prognostic factors of elderly patients with liver failure complicated with fungal infection

Objective:To observe the clinical characteristics of elderly patients with liver failure complicated with fungal infection,and to analyze the factors influencing the prognosis.Method:117 cases of elderly patients with liver failure,who were treated in our hospital from May 2013 to May 2015,were selected as object,according to whether combined with fungal infection,the patients were divided into Fungal infection group (57 cases) and non fungal infection group (60 cases).The differences of liver function index,treatment effective rate and mortality were observed between the two groups;To compare the prognosis of patients with different clinicopathological features,and to explore the factors influencing the prognosis of patients.Results:The scores of ALT,AST and DBIL in patients with fungal infection were significantly higher than those without fungal infection,and the difference was statistically significant (P<0.05);The effective rate of the treatment group was significantly lower than that of the non fungal infection group,the mortality rate was higher than that of the non fungal infection group,the difference was statistically significant (P<0.05);The death rate of patients with age was more than or equal to 70 years of age,with diabetes,with fungal infection were higher (P<0.05);Logistic regression analysis showed that combined with diabetes mellitus,combined with fungal infection was an independent prognostic factor for elderly patients with liver failure (OR=2.982,4.817,P<0.05).Conclusion:Liver function injury is severe in elderly patients with liver failure,the mortality rate is high,and fungal infection is more serious,and diabetes and fungal infection are risk factors for mortality.
8.Analysis on the situation of angiotensin-converting enzyme gene polymorphism in military service officers of Jinan theater

Objective To investigate and analysis the situation of angiotensin-converting enzyme (ACE) gene polymorphism in military service officers of Jinan theater .Methods Roche Cycler480II fluorescence quantitative PCR analyzer was used to detected ACE genotype .Meanwhile ,some samples were randomly analyzed by agarose gel electrophoresis .Results ACE DD genotype ac-counted for 23 .3% in the military service officers of Jinan theater cadres ,ID type accounted for 43 .2% ,II type accounted for 33 .5% ,D and I allele frequencies were 0 .44 and 0 .56 ,respectively ;type II ACE gene frequency was highest in female cadres ,while frequency of ID type was highest in male cadres ,the difference was statistically significant between two groups (P< 0 .05);the difference of allele frequency between men and women was not statistically significant (P>0 .05) .Conclusion ACE gene polymor-phism with type II and Iallele were dominant in the military service officers of Jinan theater cadres ;ACE gene polymorphism survey is important for early detection and timely prevention of certain related diseases .
9.Relationship between potassium channel and ADAR1 in Hep-2 cell line

OBJECTIVE To study the relationship between potassium channel and ADAR1 in Hep-2 cell line. METHODS The potassium current was recorded by the whole-cell recording technique of perforated membrane clamp. Using semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) , the expressions of ADAR1 mRNA in the Hep-2cell line were detected. RESULTS The similar membrane current was observed when cells were held at -40 mV and test potentials ranged from -80 mV to +80 mV. The current exhibited properties of voltage-dependent, outward rectification. It exhibited complete activation after alatency of 25ms and no or little inactivation over the 800ms voltage pulse. It could be blocked by potassium channel blockers TEA. The relative intensities of ADAR1 mRNA of t he Hep-2 cell line were different before and after its potassium channel was blocked. CONCLUSION Delayed rectifier potassium channel exist in human laryngeal carcinoma cell line Hep-2.The potassium channel is voltage-dependent. There is a correlation between the potassium channel and ADAR1 mRNA in Hep-2 cell line.

To improve the quality of postgraduate students,school of medicine of Shanghai Jiao Tong University have offered medical molecular genetics course to postgraduates and obtained good teaching efficiency. More than ten professors gave lectures on the academic foreland,leading students to find and solve academic problems and training practical and innovative ability of students. Accord-ing to the questionnaires of evaluation of the course and suggestions from postgraduates in the recent two years,some recommendations for the further teaching reform were proposed,such as using multi-plex teaching models,strengthening communication for teaching information with the students,em-ploying comprehensive evaluation methods and strengthening coordination and management of the course.
