1.Wnt signal transduction and human tumor
Academic Journal of Second Military Medical University 1999;0(12):-
Wnt signal transduction pathway plays a very important role in embryonic development, and its abnormality may lead to developmental defects and cellular malignant transformation. It has been shown that disturbances of the Wnt signal transduction pathway were significantly related to human neoplastic transformation. The core mechanism of the pathway abnormality was the accumulation of free ?-catenin protein in the cytoplasm and nucleus resulting in the expressions of specific target genes by downstream pathway. This paper reviews the composition and structural character, transduction and regulation of the Wnt signal pathway and its relation with human tumorigenesis.

Objective To investigate the possible association between HLA-B51 alleles and Behcet's disease (BD). Methods Totally, 61 Chinese patients of Han nationality, who were diagnosed with BD according to the International Study Group (ISG) criteria, were recruited. The control cohort consisted of 100 healthy individuals. Blood samples were obtained from all the subjects. PCR-sequenee specific primers (SSPs) were used to for the genotyping of HLA-B51 alleles (HLA-B5101-HLA-B5109). Results Com- pared with the control group (11 positive, 11% ), the frequency of HLA-B51 (18 positive, 29.5% ) was sig- nificantly increased in BD patients (χ2=8.79, P<0.01, RR=3.39). The HLA-B51-positive patients and controls consistently carried HLA-B5101 allele with no other alleles observed. There were 15 males and 3 females in HLA-B51 positive patients, 22 males and 21 females in HLA-B51-negative patients, and signifi- cant differences in gender distribution was observed between HLA-B51-positive and negative patients (P<0.05 ). Moreover, the average age of onset in HLA-B51-positive patients significantly differed from that in HLA-B51-negative patients (28.4±10 years vs 37.3±12 years, P<0.05). However, no significant differ- ences were noticed in the clinical types, course, skin lesions, prevalence of genital ulcer, eye damage, joint involvement, or pathergy reaction between HLA-B51-positive and -negative patients (P0.05). Conclu- sions This study supports that HLA-B5101 allele is associated not only with the development of BD, but also with the gender and onset age of patients with BD of Chinese Han nationality.

China Oncology 1998;0(04):-
PURPOSE Anti-cancer drug treatment induces cell death by apoptosis, whether the gene p53 undergoes mutant changes is found to have a close relationship with anti-cancer drugs. The aim of this work is to investigate the role that p53 plays in the 5-FU-induced apoptosis of colorectal cancer cells in vitro. METHODS Thirty-nine col-orectal cancer samples from patients were treated with 5-FU (10?g/ml) and folinic acid (5?g/ml. 10?g/ml) in vitro, using the in situ Terminal Deoxynucleotidyl Transferase assay (TDT) to detect the chemosensitivity. p53 mutations from tumor DNA were detected, after amplication by PCR of exons 5 to 8. by non-radioactive single-strand conformation polymorphism(PCR-SSCP). RESULTS p53-gene mutations were observed in 43. 6% (17/39) of colorectal carcinomas. When TDT assay was used to detect the tumor apoptotic rate, cells with mutated p53 had less chemosensitivity than those without (16. 4% ?4. 89% vs 26. 6%? 6. 80% P

Chinese Journal of Practical Internal Medicine 2001;0(05):-
Objective To analyze the etiology and the clinical characteristis of fever in older patients in Wenzhou. Methods Totolly 87 cases admitted to the First Affiliated Hospital of Wenzhou Medical College from January 2001 to April 2004 were retrospectively analyzed;all cases displayed fever as the major clinical manifestation and met the criteria of fever well. Results The etiology of fever which was infectious disease was in 44 of the 87 cases, accounting for 50.6%,collagen-vascular diseases 4.6%,neoplasms 10.3% and miscellaneous diseases 5.7%.In 28.7% of the cases the etiology could not be found. And in the infectious disease the tuberculosis accounted for 13.6%(6/44). Ultrasonography ,X-rays,CT scan,MRI,the marrow puncture and biopsy contributed to the final diagnosis in 21.1%,16.9%,63%,0,16.7% and 57.1% of the cases, respectively. Conclusion Because the symptoms of the older patients'febrile diseases are atypical, the diagnosis is more difficult.Infectious disease still remains a major cause of the fever in the elderly.A thorough history of disease, full physical examination of the patient and routine laboratory studies, especially erythrocyte sedimentation rate, are very important in determining the etiology of febrile disease.Early and reasonable use of non-invasive imaging techniques and the essential invasive methods are helpful to diagnosis.

Journal of Medical Research 2006;0(01):-
Objective To evaluate the effectiveness of PGE1 therapy for survival and incidence of complications in patients with liver failure. Methods MEDLINE,EMBASE.com,Cochrane Controlled Register and CBMdisc were searched.Randomized controlled trials of PGE1 therapy for liver failure were collected and reviewed. The quality of articles was evaluated and data was extracted from it.The odds ratio for death and complications among the patients with PGE1 therapy as well as the blank controls was calculated at the end of therapy. The results of meta-analysis were assessed according to sensitivity and bias.RevMan4.2 was applied to process the data. Results The results of meta-analysis showed that PGE1 reduced the case-fatality ratio of liver failure [OR=0.30,95%CI(0.23~0.37)] and the risk difference was decreased 29% in PGE1 patients compared with the blank controls. And there was statistical significance in PGE1 reducing the incidence of hepatorenal syndrome, but occurance of the complieations such as hepatic encephalopathy, bleeding, infection etc. These results were not reversed in the analysis of sensitivity, and the analysis of bias showed little bias of them. Conclusions PGE1 can reduce the case-fatality ratio of liver failure and the incidence of hepatorenal syndrome. The accuracy of the results are low due to the poor quality of the included trials. Thus, the effect of PGE1 on liver failure needs to be further evaluated through randomized controlled trials of high quality to be carried out in multicenter and large samples.

ObjectiveTo evaluate the significance of CK19 mRNA monitoring in peripheral blood of breast cancer patients.MethodsPeripheral blood samples were collected from 137 breast cancer patients preoperatively, one day post-operation,7 days,1,3,6,12,18,and 24 months after operation.RTPCR was used to detect CK19 mRNA expression. The relationships were analyzed between CK19 mRNA expression and treatment result, between CK19 mRNA expression and clinicopathological parameters.ResultsThere was no significant difference between the level of CK19 mRNA as tested preoperatively, and 1,7 days postoperatively(P > 0.05 ).From one month and thereafter,CK 19 mRNA decreased significantly when compared with that immediate perioperatives ( P < 0.05 ). Postoperative peripheral CK19 mRNA expression increased in those found with recurrent or metastasized tumors ( P < 0.01 ). CK19 mRNA expression does not correlate with patient's menstrual status, estrogen receptor expression, progesterone receptor expression,HER-2 expression and Ki67 proliferation expression (P > 0.05 ). But there was statistically significant correlation between CK19 mRNA expression and tumour size, histology grading, pathological type, lymph node metastasis ( P < 0.01 ).ConclusionsIn breast cancer patients peripheral blood CK19 mRNA expression is correlated with risk clinicopathological parameters, and increases in patients with postoperative recurrence and tumor metastasis.

China Oncology 2016;26(5):388-393
Background and purpose:Icariin (ICA) is the important active flavonoids extracted from Berberidaceaeepimedium. It has been shown to be effective in suppressing cancers including lung cancer and gastric cancer. Thus, it is expected to be developed for cancer treatment. However, there were few studies on icariin as a promising anticancer drug for the treatment of thyroid cancer. The mechanisms underlying anticancer effects of ICA in thyroid cancer are rarely reported. This study was to explore the proliferation and apoptosis, intracellular ROS and antioxidant enzyme systems of the thyroid carcinoma cell line B-CPAP treated with different concentrations of ICA. It aimed to explore the mechanism underlying anticancer effects of ICA, and to determine whether it is concentration- or time-dependent manner. Methods:The proliferation of B-CPAP cell line treated with different concentrations of ICA was detected by cell counting kit-8 (CCK-8). The cell apoptosis and intracellular ROS were observed by flow cytometry. The expression of intracellular superoxide dismutase and intracellular malondialdehyde were measured by SOD detection kit and MDA assay kit, respectively. Bcl-2 and γ-HA2X were detected by Western blot. Results:ICA reduced B-CPAP cell activity, increased the rate of apoptosis in a dose- and time-dependent manner after 48 h (P<0.01). The ROS of ICA 50 mg/L and 200 mg/L groups were (2.12±0.14)-fold and (2.41±0.12)-fold of the control group, respectively. ICA promoted accumulation of malondialdehyde, and reduced antioxidant enzyme SOD activity. The SOD activity was decreased by (9.35±1.45)% (ICA 50 mg/L group) and (21.5±1.52)% (ICA 200 mg/L group) compared with the control group, respectively. The anti-apoptotic protein Bcl-2 in ICA 200 mg/L group was decreased by (13.64±1.71)%compared with the control group. Conclusion:Icariin inhibited activity of thyroid cancer B-CPAP cells in a dose- and time-dependent manner. It plays an important role in promoting intracellular ROS expression, inhibiting superoxide dismutase expression and decreasing Bcl-2, which leads to irreversible damage to the cell, thereby inducing apoptosis.

Objective To observe the clinical efficacy of thunder-fire moxibustion at Dazhui (GV14) and Shenshu (BL23) in treating mild cognitive impairment (MCI) due to ischemic cerebral stroke. Method Sixty patients with MCI due to ischemic cerebral stroke were randomized into a treatment group and a control group, 30 cases each. The control group was intervened by joint treatment plus cognitive training, while the treatment group was by thunder-fire moxibustion in addition to that given to the control group. The two groups were evaluated by the Montreal Cognitive Assessment (MoCA), Mini-Mental State Examination (MMSE) and Wechsler Memory Scale (WMS), the major symptoms were observed, and the clinical efficacies were compared between the two groups. The two groups were both treated for 8 weeks. Result The total effective rate was 80.0%in the treatment group, versus 70.0% in the control group, and the difference was statistically significant(P<0.05). Conclusion The selected acupoints can improve the cognition and activities of daily living (ADL) in MCI patients;thunder-fire moxibustion at Dazhui and Shenshu plus cognitive training can produce a better clinical efficacy than dry cognitive training. Therefore, we can combine thunder-fire moxibustion with modern rehabilitation to enhance the therapeutic efficacy in preventing and treating MCI.
