1.Cryptotanshinone effects on the proliferation and apoptosis of renal carcinoma stem cells

Abstract BACKGROUND:Previous studies have found that cryptotanshinone represses multiple tumors, but little is reported on its effect on renal carcinoma. OBJECTIVE:To explore the effect of cryptotanshinone on the proliferation and apoptosis of the renal carcinoma stem cels. METHODS:CD133+ renal carcinoma stem cels were separated from OS-RC-2 cels by immunomagnetic bead separation. Effects of 0, 0.2, 1, 5 mg/L cryptotanshinone on the proliferation and apoptosis of CD133+ renal carcinoma stem cels were detected by MTT and flow cytometry, respectively. Expression levels of Ki67, Bcl-2, Caspase-3 and p-Caspase-3 protein were detected by western blot assay. RESULTS AND CONCLUSION:After magnetic cel sorting, the percentage of CD133+ cels was increased from 6.32% to 82.73%, and there was a significant difference before and after cel sorting (P < 0.001). Cryptotanshinone could repress the proliferation of CD133+ renal carcinoma stem cels and promote cel apoptosis in a dose-dependent manner. The protein expression levels of Ki67 and Bcl-2 in the 5 mg/L cryptotanshinone group were significantly decreased compared with the control group, while the protein expression level of p-Caspase-3 protein was significantly increased. In addition, there was no difference in the protein expression of Caspase-3 between cryptotanshinone and control group. These findings indicate that cryptotanshinone may be a potent anticancer drug for the treatment of renal carcinoma by inhibiting expression of Ki67 and Bcl-2 and promoting protein expression of p-Caspase-3. 2.Pathological study of the skeletal muscle in electrocution

Demonstration of the abnormal distribution of Mb and Fn in electric injuries of skeletal muscles. The electrocution model of rats was established, and the human autopsy samples of electrocution were collected. Immunohistochemical study of myoglobin(Mb) and fibronectin(Fn) were performed on the skeletal muscles in the electric current pathways. The results showed that the muscle fibers showed the bamboo-like appearance and depletion of the Mb as well as the positive reaction of Fn which was called the python-pattern. This experiment proved that the bamboo-like appearance and python-pattern change of the skeletal muscle fibers have the practical value in the medicolegal diagnosis of electrocution without typical current mark.
3.The Effects of rhIL-3, rhIFN-r and DMSO on Proliferation and Differentiation of the HL-60 Cell Line in Vitro

In this experiment, we studied the effects of rhIL-3, rhlFN-r and DMSO on the growth and differentiation of the human leukemia cell line HL-60. The results showed that three agents were capable of suppressing the proliferation of the HL-60 cells and inducing terminal differentiation of the cells, with the exception of early augmentation of the cell growth by the rhIL-3. The differentiation effect was most obvious in DMSO treated cells, then in rhIL-3 and least in rhIFN-r The enhanced effects occurred when the HL-60 cells were treated in combination with DMSO and rhIL-3 or rhIFN-r
4.In Vitro Study of Differentiation of U_(937) Cells Induced by rhTNF-?, rhIL-1-?, PHA-LCM and c-RA

In this study, several physiological factors were studied for their effects on the growth and differentiation of U937 cells in vitro .The results showed that (1)rhTNF-?,PHA-LCM and c-RA exerted a dose and time-dependent suppression of proliferation of U937 cells; (2) the synergistic effect oc-cured when rhTNF-? was combined with c-RA in the cell culture; and (3)rhIL-1-? augmented the differentiation of U937 cells induced by c-RA though it had no effect on the cell line by itself. According to the [3H]-TdR incorporation, it can be concluded that the reduction of DNA synthesis of U917 cells by rhTNF-? ,PHA-LCM,and c-RA is attributed to the inhibition of the growth. The results might have clinical significance in the induction of differentiation of leukemia cells.
5.Application of 320-detector row dynamic volume CT angiography in follow up of coronary artery aneurysms caused by Kawasaki disease

ObjectiveTo assess the values of 320-detector row dynamic volume CT angiography (CTA) and transthoracic echocardiography (TTE) in follow up of coronary artery aneurysm (CAA) caused by Kawasaki disease (KD).Methods320-de-tector row CTA and TTE were applied in long-term follow-up of 8 patients with CAA caused by KD.ResultsIn 8 patients, the mean age at onset was 41.63±22.70 months and the mean follow up time was 43.50±10.99 months. In acute phase, 3 cases of giant coronary artery aneurysms (GCAA) and 5 cases of mid-small CAA were diagnosed by TTE. A total of 16/32 arteries (50%) were involved. At the end of follow-up, 3 cases of GCAA and 2 cases of mid-small CAA were still diagnosed by TTE, and small CAAs were regressed in another 3 cases. A total of 6/32 arteries (18.75%) were involved. Simultaneously at the end of follow-up, a total of 7/32 arteries (21.9%) were involved by 320-detector row CTA. The distribution was consistent with that of TTE. Mean-while, there were one case of left circumlfex artery, one case of GCAA at distal of the right coronary artery, 2 cases of thrombus, 1 case of coronary stenosis and 2 cases of calciifcation.ConclusionsCAA caused by KD may be persistent for a long time. The thrombus, stenosis, and calciifcation of coronary can occurr at late phase in GCAA. TTE is sensitive and reliable to detect proxi-mal and middle segment of coronary lesions, but has limitations in detection of distal segment of coronary arteries. 320-detector row CTA has more comprehensively view of each coronary artery lesions and is especially sensitive and reliable to detect coro-nary thrombosis, calciifcation and narrowing in proximal and distal coronary arteries after acute phase.

8.Quality and Quantity Analysis on Tectoridin of Iris Tectorum Maxim

This study was aimed to establish the quality and quantity analysis methods of tectoridin contented in the Iris tectorum Maxim. Thin layer chromatography (TLC) method was used to give quality analysis on tectoridin in I. tectorum. And the HPLC method was used to give quantity analysis on tectoridin in I. tectorum. The Diamonsil C18 (200 mm í 4.6 mm, 5 μm) column was used as analytical column. The acetonitrile-0.2% phosphoric acid (20:80) was used as mobile phase at the flow rate of 1.0 mL·min-1. The detection wavelength was 265 nm and the column temperature was 25℃. The results showed that the quality analysis of I. Tectorum had specific identification with-out the interference of other ingredients. The amount of inlet tectoridin had a good linearity with the response val-ue of peak area in the range of 0.12~2.22 μg. The average recovery rate was 100.96%. It was concluded that this method was simple and reproducible, which can be used in the quality control of I. tectorum.
9.HPLC Fingerprint of Qian Solidaginis Herba

This study was aimed to establish the fingerprints of Qian Solidaginis Herba by HPLC. The fingerprints of Qian Solidaginis Herba were built by using Diamonsil C18 (200 mm í 4.6 mm, 5 μm) column and acetonitrile -0.2%H3PO4 aqueous solution in gradient as mobile phase. The flow rate was 1.0 mL·min-1. Detecting wavelength was set at 282 nm. The total 10 batches of Qian Solidaginis Herba samples and their adulterants were analyzed. The results showed that 13 peaks were identified as the fingerprints of Qian Solidaginis Herba. Under chromatographic conditions mentioned above. It was concluded that the HPLC fingerprints method was found to have satisfactory accuracy, sta-bility and reproducibility, which was a potential method for the quality control of Qian Solidaginis Herba.
10.HPLC Determination of Chlorogenic Acid, Rutin and Kaempferide in Solidago decurrens Lour

This study was aimed to establish an HPLC method for the determination of chlorogenic acid, rutin and kaempferide in Solidago decurrens Lour. A Diamonsil C18 column (4.6 mm í 200 mm, 5 μm) was adopted with the mobile phase of acetonitrile-0.2% phosphoric acid. The flow rate was 1.0 mL·min-1. The column temperature was at 25℃. The wavelength of detection was set at 282 nm. The results showed that the calibration curve was linear over the range of 63.2~442.4 μg for chlorogenic acid (r = 0.999 3), 8.1~56.8 μg for rutin (r = 0.999 4) and 10.8~75.7μg for kaempferide (r = 0.999 8). The average recovery of chlorogenic acid was 98.6% (RSD = 1.4%), that of rutin was 99.2% (RSD = 0.8%) and that of kaempferide was 100.3% (RSD = 1.0%). It was concluded that the method was simple, economical and accurate with good reproducibility.
