1.Transfer of interferon-induced viral resistance from human lymphocytes to amnion FL cells
Chinese Journal of Immunology 1985;0(02):-
Transfer of interferon-induced viral resistance from human lympnocytes to numau amnion FL cells was studied by using dye uptake method.Lymphocytes pretreated with Human IFN-?(500u/ml)and then cocultured with FL cells at a 5:1 ratio for 4 hours can efficiently transfer the IFN-induced viral resistance to FL cells. This process depended on the close contact and sutable ratio of donor lymphocytes to recipient FL cells at a given density.Antiviral activity in the cocultured homologous cells was not blocked by sheep antisera to Namalva-IFN,but could be prevented by rotatingcocultures.Specific ~(51)Cr release and staining with vital dyes showed little effect ofcell-mediated cytotoxicity on FL cells, which expressed the transfered antiviral state. These findings suggest that the IFN-induced signal molecules on membrances of lympho-cytes might have served as an important mediator in induced transfer action. Thus thepathway of transfered antiviral activity may be a new and efficient host defense aga-inst viral ections.
2.Kinetic analysis on production of IL-1, IL-6 and TNF? by human monocytes
Chinese Journal of Immunology 1985;0(06):-
Kinetics of production of IL-1, IL-6 and TNFa induced by endotoxin from human monocytes as well as their relationships were studied using cytokine bioassays and immu nocytochemical staining methods. The results showed that accu mulation of IL-1 ?, IL-6 and TNFa molecules in cytoplasma and their release into supernatants had different kinetic patterns from each others, reflecting the difference in gene activation, transcription, translation and release of these three cytokines. In addition, only part of monocytes synthesize IL-1 ?, IL-6 and TNF?, suggesting the presence of different cytokineproducing monocyte subsets.

Objective To evaluate the effects of continuous pulmonary artery Perfusion with hypothermic oxygenated blood on lung function in children with congenital heart defects (CHD) and pulmonary hypertension(PH). Methods 20 patients diagnosed with CHD and PH were divided into group 1( perfused with oxygenated blood) and group 2 (conventional CPB). A canula was inserted into the main pulmonary artery for the perfusion with a rate of 15~30 ml?kg -1 ?min -1 . CPB was established as usual. The data of two groups including the oxygen concentration, airway pressure, blood gas analysis were recorded. At fixed times, blood sample was taken out for the measurement of the sICAM-1 and MDA. Results The concentration of sICAM-1 and MDA in group 1 was much lower than that in group 2 on 12 and 24 hours after surgery. Issues of the lung function including OI?A-aDOa_2 and airway pressure was significantly different between the two groups. Conclusion The study showed that the continuous pulmonary perfusion with oxygenated blood could preserve lung function in patients with CHD and PH.

Objective To investigate the frequency of micresatellite instability (MSI) and loss of heterozygosity (LOH) and select sensitive loci for studying microsatellite DNA imbalance in thymic squamous cell carcinoma. Methods 5 microsat-ellitc polymorphism markers and extrated DNA were selected from 9 specmiens of paired thymie squamous cell carcinoma/nor-real tissues. MSI and LOH in the specmiens of thymic carcinoma and relevant pericancerou tissues were detected by polymerase chain reaction (PCR) followed by 6% polyacrylamide gel electrophoresis(PAGE) with silver staining. Results MSI or LOH was detected in 9 thymic carcinoma tissues. The frequency of MSI or LOH was 66.7% (6/9) at loci of D6S1708, 33.3% (3/9) at TP53, 33.3% (3/9)at DM, 33.3% (3/9)at D11S988 and 0% (0/9)at D8S136, LOH at D6S1708 (5/6) was a common genetic alteration. DI1S988 had only LOH alteration. Conclusion D6S1708, TP53, DM, and D11S988 are sensi-tive loci for studying microsatellite DNA imbalance in thymic squamous cell carcinoma. Microsatellite DNA imbalance may play a certain role in occurrence and development of thymic squamous cell carcinoma, and the relationship between MSi or LOH.The linicopathological features of thymic squamous cell carcinoma needs further investigation.

Objective To explore the mechanism of gene Nelin on the differentiation of vascular smooth muscle cells(VSMC).Methods Human VSMCs were stably infected with over-express lentiviral vector [Nelin-VSMC] and interference lentiviral vector [LV-Nelin-SiRNA-VSMC] by Western blotting and RT-PCR.The effect of Nelin expression on phenotypic transition of VSMCs was observed and the mechanism involved was explored. Results The Nelin-VSMC cells take on the contractile phenotype of long slender cells possessing hill-valley characteristics.Overexpression of Nelin can enhance SMoα-actin expression with the increase of total RhoA and nuclear serum response factor(SRF) expression.Treated with a selective Rho kinase inhibitor Y-27632,the expression of SMα-actin and nuclear SRF protein induced by Nelin was effectively reduced.Meanwhile, the opposite results were observed in the Nelin-depression VSMC.Conclusions Nelin can enhance the expression of SMα-actin and carry a pivotal role in the process of VSMCs differentiation and phenotype modulation by activating Rho and SRF.
