Objectives To investigate the expressions of miR-101 and COX-2 in colorectal cancer. Methods Thirty-twocolorectal cancer specimens and paired paracancer tissues were collected for assessment of miR-101 and COX-2 expressions by real-time quantitative PCR. Thecorrelation between miR-101 and COX-2 , as well as their correlations with the pathologywere analyzed. Results The expression level of miR-101 and COX-2 mRNA in the cancer tissues was significant difference between the cancer and para-cancer tissues (P < 0. 01). Also, miR-101 expression was negatively correlated with COX-2 mRNA expression (r = -0.373 5, P < 0.01). Furthermore,miR-101 expression was significantlydecreased along with the deterioration of tumor differentiation grade(P < 0.05). Compared with which in Dukes A ~ B stage, COX-2 expression was overexpressed and miR-101 was down-regulatedin colorectal cancer tissue inDukes C ~ Dstage , butnosignificant difference was found (P > 0.05). Conclusion Down-regulated miR-101 expression andparallel COX-2 overexpression hasbeen linked to colorectal cancer. miR-101 and COX-2might be thepotentialdiagnostic markers and therapeutic targets.

Self-assembled gold nanoparticles ( AuNPs) coating was fabricated using an etched stainless steel wire as a support on which AuNPs were first deposited, then after modified with mercaptan, another layer of AuNPs was self-assembled. The AuNPs modified stainless steel wire was used in solid phase microextraction (SPME) coupled with high performance liquid chromatography (HPLC) for the determination of ultraviolet ( UV) filters in environmental water. The best extraction efficiencies were achieved within 30 min at 55℃ and at pH 7 with stirring rate of 800 r/min. Under the optimized conditions, the established AuNPs-SPME-HPLC method for the determination of UV filters benzophnone-3 ( BP-3 ) , 2-ethylhexyl-4-( N, N-dimethylamino ) benzoate ( OD-PABA) , 2-ethylhexyl-4-trimethoxycinnamate ( EHMC) and 2-ethylhexyl salicylate ( EHS) was linear in the range of 0. 004-200 μg/L. The limits of detection of the method were 0. 43-570 ng/L ( S/N=3 ) . The relative standard deviation ( RSD ) of AuNPs-SPME-HPLC was 1 . 9%-4 . 2% ( n=5 ) for spiked water samples of 20 μg/L each UV filter. In the case of real water samples analyses, the recoveries of spiked UV filters were 77 . 9%-108% with RSDs of 3 . 1%-8 . 0% ( n=5 ) .

Objective To discuss the application of longitudinal reduction of Chinese traditional medicine in surgical treatment of distal radius comminuted fractures in the elderly. Methods Before the operation, the 54 elderly patients with the fracture were reduced by longitudinal direction under anesthesia, then they were treated with minimally invasive plate osteosynthesis in the approach upon the reduction condition and fracture types. Results After 12-month follow-up, the fractures were all healed. The mean healing time of the fractures was 8 weeks (6-12 weeks). At the end of follow -up, the mean range of motion of the wrist was at 50°of flexion, 45°of extension, 30° of ulnar deviation, at 20° of radial deviation. According to the criteria of Gartland and Werley, the results were excellent in 35 cases, good in 17 cases, fair in 2 cases and poor in 0 case. Conclusions It is vital to provide a Chinese traditional longitudinal reduction before invasive surgical plate fixation in treatment of distal radius comminuted fractures, to avoid large-area exposure of fracture site, minimize the damage to the soft tissue, maintain reduction of post operation and achieve good wrist function.

BACKGROUND:Shear stress can directly mediate the expression of endothelial cells, especially some cytokine genes whose codes are related to angiogenesis. Otherwise, flow shear stress of blood plays an importantly biological role in regulating vascular structure and function.OBJECTIVE: To observe the effects of laminar flow shear stress on the proliferation of vascular endothelial cells and the expression of protooncogene c-myc in human cerebral arteriovenous malformation.DESIGN: Randomized controlled study.SETTING: Department of Neurosurgery, Shenyang General Hospital of Military Area Command of Chinese PLA.MATERIALS: The experiment was carried out in the Neuromedical Institute, General Hospital of Shenyang Military Area Command of Chinese PLA from November 2006 to February 2007. Fresh samples of human cerebral arteriovenous malformation were derived from 20 patients who were of grade Spetzler Ⅱ -Ⅲ and received resection of human cerebral arteriovenous malformation in the Department of Neurosurgery, General Hospital of Shenyang Military Area Command of Chinese PLA in 2006. All cases were diagnosed with whole-brain angiography before operation. The main reagents and equipments were detailed as follows: M199 culture media (Gilbco BRL), quality fetal bovine serum (HyClone), endothelial cell growth supplement (ECGS; Sigma, USA), CO2 incubator (Forma Scientific, USA), flow cytometry analysis of cell cycle kit (BD Company), flow cytometer (FACS Calibur, BD Company), rat-anti-human c-myc monoclonal antibody (Santa Cruz Company, USA), and reverse transcription polymerase chain reaction (RT-PCR) kit (Promega).METHODS: Tissue explants adherent method was used to culture vascular endothelial cells of human cerebral arteriovenous malformation, and then the cells were classified into 4 groups based on degree of shear stress, including control group, low shear stress group, moderate shear stress group and high shear stress group. Cultured endothelial cells of human cerebral arteriovenous malformation were put in a parallel plate flow chamber. In addition, cells in the low, moderate and high shear stress groups were stressed by low, moderate and high shear stress for 8 hours, respectively. However, shear stress in the control group was 0 Pa. Flow cytometry was used to measure proliferation index, and the expression of c-myc protein and c-myc mRNA were determined by immnocytochemistry and RT-PCR analysis respectively.MAIN OUTCOME MEASURES: Expressions of c-myc protein and c-myc mRNA and proliferation index in endothelial cells under various degrees of shear stress.RESULTS: ① Proliferation index: Proliferation index was higher in the moderate and high shear stress groups than that in the control group (P < 0.05, 0.01). ② Expression of c-myc protein: Immuneposjtjve expression of c-myc protein was gradually increased with the increase of shear stress and there were significant differences in the three shear stress groups as compared with control group (P < 0.05-0.01). ③ Expression of c-myc mRNA: Proliferation index of endothelial cells was higher in the low and moderate shear stress groups than that in the control group (P < 0.05).CONCLUSION: Flow shear stress can induce expression of c-myc and activate expression of c-myc gene based on gene transcription so as to promote the proliferation of vascular endothelial cells in human cerebral arteriovenous malformation
