Objective: To analyse the clinical significance of a-L-Fucosidase(AFU) in the diagnosis of primary hepatic carcinoma(PHC).Methods: The activity of serum AFU was measured in 33 patients with PHC,42 patients with cirrhosis,37 patients whit hepatitis,10 patients with secondary liver cancer,37 patients with other malignant tumors and 80 normal persons.The differences of the level of AFU between the groups were compared.Results: The mean serum AFU activity was significantly higher in patients with PHC(41.2?14.1 U/L) than that in normal persons(20.5?7.2U/L),secondary liver cancer(22.2?13.6 U/L),other kinds of malignant tumors(22.0?7.1 U/L),P
2.Treatment of unstable angina with trimetazidine

Objective To evaluate the clinical therapeutic effects oftrimetazidine on the treatment of unstable angina (UA) as well as its effects on endothelin-I level and complications of patients. Methods One hundred and twenty patients with UA were randomized into the trimetazidine group (n =60) and the control group (n =60), the trimetazidine group was subjected to treatment with 60 mg trimetazidine everyday for six months plus conventional treatment, and the clinical symptoms, changes in electrocardiogram, changes in the number of plasma circulating endothelial cells (CEC) and endothelin-1 level of the two groups were observed after treatment for four weeks; and the incidence rates of cardiac arrhythmias, cardiac failure, hospitalization due to angina, myocardial infarction and sudden death were also observed after treatment for six months. Results 1) The total effective rate of integrative clinical therapeutic effects in the trimetazidine group and the control group after treatment for four weeks were 86.7% and 68.3%,respectively (P<0.05), and the excellence rates were 36.7% and 15% (P<0.01)respectively; the total effective rates for the therapeutic effects in electrocardiogram were 66.7% and 46.7%,respectively (P<0.05), and the excellence rates were 30.0% and 11.7%, respectively (P<0.01 ). 2) The number of plasma CEC and endothelin-1 level of the two groups after treatment for four weeks significantly decreased (P<0.05), but the decreases in the trimetazidine group were even significant (P<0.01). 3) The incidence rates for cardiac arrhythmia in the trimetazidine group and the control group after treatment for six months were 10% and 20% (P<0.05), respectively, and the incidence rates for cardiac failure were 8.3% and 18.3%, respectively (P<0.05), and the incidence rates for hospitalization due to angina were 10% and 15%, and the incident rates for myocardial infarction were 3.3% and 13.3% respectively (P<0.05). Conclusion Trimetazidine can significantly improve the symptoms of UA and myocardial ischemia, reduce the damages to blood vessel endothelium and complications, and improve the prognosis.
3.Long-term follow-up for laparoscopic anterior 180° partial fundoplication for gastroesophageal refulx disease

Objective To evaluate long-term follow-up for laparoscopic anterior 180° partial fundoplication for gastroesophageal refulx disease (GERD).Methods A total of 48 patients had undergone a laparoscopic anterior 180° partial fundoplication from July 2004 to October 2007.Patients were followed up at 3 months,12 months,3 years,5 years by using a structured questionnaire via phone or e-mail which evaluated symptoms of reflux,dysphagia,side-effects,and overall satisfaction with the clinical outcome.Results Follow-up data were collected from 43 patients,ranging from 60 to 98 months.Postoperative heartburn significantly improved in 37 patients.Normal belching was preserved in 40 patients,and 38 patients were able to eat normally.Thirty nine (90.7%) patients reported a good or excellent result (minimal or no symptoms) at the late follow-up.Two patients underwent laparoscopic anterior 180° partial fundoplication again due to acid reflux at the 12th and 38th month respectively.Conclusion At minimum 5 years followup,laparoscopic anterior 180° partial fundoplication for GERD is effective and lasting,and most patients are satisfied with the outcome.
4.Observation of Therapeutic Effects of Shenfu Injection on Sick Sinus Syndrome

OBJECTIVE:To study the therapeutic effect of Shenfu injection on sick sinus syndrome. METHODS: 125 patients with sick sinus syndrome were divided into two groups. 63 patients were given Shenfu by intravenous drip for 14 days as treatment group. The other 62 patients were given glucose insulin potassium(GIK) for 14 days as control group. The changes of clinical manifestations, electrocardiogram(ECG and DCG) and esophagus electrophysiological examination were compared between two groups with a randomized single blind design. 5.Embryotoxicity of eugenol based on a model of embryonic stem cell test

BACKGROUND:As the pharmacological effect of eugenol constantly being discovered, its application in medical and food industry becomes wider. However, its toxicity studies have not established a complete database, especialy in the improvement of safety assessment of developmental toxicity and teratogenicity. OBJECTIVE:To establish a model of embryonic stem cel test to evaluate the embryotoxicity of eugenol. METHODS:Mouse fibroblasts (3T3) and mouse embryonic stem cels (E14TG2a) were culturedin vitro, and MTT test was performed to detect the cytotoxicity of 3T3 cels and E14TG2a cels with positive control 5-fluorouracil, negative control penicilin G and tested compound eugenol. The concentration of the tested compounds that inhibiting 50% viability of embryonic stem cels (IC50 E14TG2a) and 3T3 fibroblasts (IC50 3T3) was calculated. The hanging-suspension-adherent culture systems were used to induce embryonic stem cels into cardiomyocytes, and the concentration of tested compounds that caused 50% inhibition of differentiation of E14TG2a cels into cardiomyocytes (ID50 E14TG2a) was calculated. The embryotoxic potential of eugenol was classified by prediction model of the embryonic stem cel test. RESULTS AND CONCLUSION:The proliferations of E14TG2a and 3T3 cels were inhibited by eugenol, of which the IC50 3T3 and IC50 E14TG2a values were (3.613±0.192) and (1.799±0.131) mg/L. The differentiation of E14TG2a was also inhibited by eugenol, of which the ID50 E14TG2a was (3.501±0.158) mg/L. Eugenol was evaluated as a chemical compound with strong embryotoxicity by the model of embryonic stem cel test.
6.Analysis of T cell clonality by using T-cell receptor varible β gene repertoire in T-cell acute lymphoblastic leukemia.

AIM: To analyze T cell clonality in patients with T cell acute lymphoblastic leukemia (T-ALL). METHODS: The complementarity determining region 3 (CDR3) size of 24 T cell antigen receptor variable β (TCR Vβ) region gene was analyzed in peripheral blood mononuclear cell (PBMC) samples from 6 T-ALL cases and 10 normal individuals by using reverse transcriptase-polymerase chain reaction (RT-PCR). The PCR products were further studied by genescan and sequencing analysis. RESULTS: Some TCR Vβ subfamily T cells display mono- or oligoclonal expansions in 3 cases of T-ALL, predominantly in Vβ2, Vβ3, Vβ6, Vβ9, Vβ21 or Vβ24, respectively. Polyclonal expansions of T cells were found in the other three cases, which could also be found in normal samples. CONCLUSION: A part of T cell acute lymphoblastic leukemia cells may arise from a clonal expansion of TCR Vβ subfamily T cell. This method may be useful for the detection of minimal residual disease in clinical study of the disease.
7.ADF/cofilins proteinic family constellation in cytoskeletal protein:How does depolymerization occur through combination of ADF/cofilins with actin?

The actin depolymerizing factor/cofilin (ADF/cofilins) are a family of actin-binding proteins expressed in all eukaryotic cells. The ADF/cofilins appear to have multiple functions, and this is reflected in their very complex association with both monomeric and filamentous actin. Phosphorylation by some kinases and other factors such as LIM kinases 1 and 2, TESK 1 and TESK 2 kinase, Insulin, etc, prevents ADF/cofllins from binding actin. The serial researchs of ADF/cofilins are increasingly becoming study hot spots, especially on the relationship between homo-sapiens disease and mechanism of action of ADF/cofilins.Now in this domain wilderness details are still far from clear, such as the mechanism by which actin filaments are depolymerized by ADF/cofilins has been controversial.

9.Study on delaying immunosenescent by lipoteichoic acid of bifidobacterium

Objective:To explore molecule mechanism about anti-aging of bifidobacterium by investigating the effect of lipoteichoic acid (LTA) of bifidobacterium on thymus of aging mice induced by D-galactose.Methods:Male Kunming mice were divided into different groups at random. These mice were induced aging by subcutaneous injection of D-galactose on the back of mice and LTA was simultaneously administered to them daily for 40 days. Body weight, thymus index ,thymus morphology, thymus c-fos, p16 production and peripheral blood IL-2 concentration were tested by HE staining, immunochemistry method, ELISA and other methods. Results:There were significant differences for body weight ,thymus index, thymus morphology,thymus p16, c-fos production and peripheral blood IL-2 concentration between the young control group and the aging model group. Whereas, LTA can improve thymus morphology ,increase body weight ,thymus index and c-fos production, meamwhile, decrease thymus p16 and peripheral blood IL-2 concentration of aging model mice. Conclusion:The results showed that LTA can remarkably delay thymus aging, and it may be a molecular mechanism of bifidobacterium delaying immunosenescent.

Objective To study the effect of lipoteichoic acid(LTA) of Bifidobacterium on the expression of vascular endothelial growth factor(VEGF) and invasion and metastasis of colon carcinoma cell lines.Methods The effects of LTA on adhesion,invasion and metastasis of human colon carcinoma lines LoVo and HT-29 in vitro were examined by MTT colorimetric and transwell chamber.The mRNA and protein expressions of VEGF in the two colon carcinoma cells treated with LTA were detected by RT-PCR and Western blot.Results The adhesion ability decreased in LoVo cells and HT-29 cells after the treatment of LTA(20,50,80 ?g/ml) for 30,60,90,120 min(P
