1.Cytotoxic action of sulfur mustard on cultured lymphocytes and their caspase-3 expression

Objective To investigate the manner and molecular mechanism of lymphocyte cytotoxic action induced by sulfur mustard. Methods Lymphocytes were separated from the spleen of Wistar rats and were cultured. DNA strand breaks were detected by fluorospectrophotometry. RT PCR and Western blotting were employed to detect the gene and protein expressions of caspase 3 and its activation. Results Sulfur mustard induced DNA strand breaks in the lymphocytes. Gene and protein expressions of caspase 3 were increased in a time dependent manner. Conclusion Sulfur mustard induces lymphocyte cytotoxic action by means of depending on the expression and activation of caspase 3.
2.Interventional effect of tetrapeptide on sulfur mustard-induced lymphocyte apoptosis

Objective To investigate the changes of caspase 3 activity and the interventional effect of tetrapeptide AC DEVD CHO on sulfur mustard induced apoptosis of lymphocyte. Methods The activity of caspase 3 was detected by fluorospectrophotometry. DNA ladder and apoptosis were detected by DNA agarose gel electrophoresis and flow cytometry. Results Sulfur mustard enhanced the activity of caspase 3 and its polypeptide inhibitor (AC DEVD CHO) partially blocked apoptosis, which was mainly presented as an obscure phenomenon of DNA ladder and a decrease in percentage of apoptosis. Conclusion Sulfur mustard can induce the apoptosis of lymphocyte by means of activating caspase 3. The inhibitor may have an interventional effect on lymphocyte apoptosis induced by sulfur mustard.
3.tk gene mutation of L5178Y cells induced by sulfur mustard

Objective To detect the mutagenicity of sulfur mustard in the tk gene of L5178Y3.7.2c tk +/- mouse lymphoma cells. Methods The plating efficiency and the mutant frequency were detected by the microtiter procedure. Results Sulfur mustard induced tk locus mutation with mutation frequency 2-15 times higher than that of spontaneous mutation frequency of L5178Y3.7.2c tk +/- cells. There were two different phenotypes of mutation colonies induced, which were large and small colonies, but most of them were small colonies. Conclusion Sulfur mustard is characterized by marked mutagenic activity and cytotoxicity. The compound may result in large range of DNA damage.
4.Effect of OFR on small intestinal injury in rats exposed to sulfur mustard and burn

Objective To investigate the effect of oxygen free radical (OFR) on the injury of small intestine in rats treated with burn, sulfur mustard or sulfur mustard plus burn. Methods Totally 95 SD rats were randomly divided into four groups: 30 rats subjected to 30 third-degree TBSA scalding (B), 30 injected subcutaneously with 2 ml/kg sulfur mustard (S), 30 subjected to both (SB), 5 as normal controls (C). 5.Chromosomal aberration and micronucleus formation induced by sulfur mustard

Objective To study the effects of sulfur mustard on chromosomal aberration in Chinese hamster fibroblast cell line (CHL cells) and the micronucleus (MN) frequency in the bone marrow cells of mice. Methods CHL cells (with and without S9) treated with different concentrations of sulfur mustard for 24 h were harvested for the preparation of chromosome. Subsequently, the frequency and the type of chromosomal aberration were examined. The KM mice were sacrificed at 24 h after subcutaneous injection of sulfur mustard. The number of micronuclei-containing polychromatic erythrocytes (PCEs) was counted by means of bone marrow smear and Giemsa staining. Results Sulfur mustard might increase the chromosomal aberration frequency of CHL cell in a dose-dependent manner. S9 had no effects on the classificaiton and the frequency of chromosomal aberration. 6.Therapeutic effects of combination of three drugs on healing of rabbit corneal wound caused by mustard gas

Objectives: To observe the therapeutic effects of collagen corneal shields (CCS) combined with fibronectin (FN) and epidermal growth factor (EGF) on healing of rabbit corneal wound caused by mustard gas, and to look for the new methods,new technology and new medicine to treat the injured cornea before eyelid edema observed in field hospital.Methods: After the left cornea was contaminated with mustard gas, 24 rabbits were equally divided into three groups(groups A, B, C) at random. Groups A,B,C were treated with PBS,CCS and CCS+FN+EGF,respectively. The corneas were stained with fluorescein and photographed 2, 8, 16, 24, 36,72 hours after contamination. The rate of corneal epithelial healing and percentage of damage were calculated. Results:The healing rates of groups A,B and C were (0.879±0.139)mm2/h, (1.223±0.166)mm2/h and (1.543±0.224)mm2/h,respectively and had statistically significant difference(P<0.01). The percentage of damage was 71.6%, 57.3% and 44.8%,respectively and also had statistically significant difference(P<0.01).Conclusions: The treatment with CCS can accelerate the corneal epithelial healing injured by mustard gas and the therapeutic effects will be better if CCS is combined with FN and EGF.
7.Bacteriological Comparison Between Shaving and No-Shaving Before Knee Arthroscopy Surgery

OBJECTIVE To study the feasibility of no-hair-shaving before knee arthroscopy surgery.METHODS Totally 160 patients preparing for knee arthroscopy surgery were divided into both test and control groups randomly.The test group was no-hair-shaving before operation,and the control group was hair-shaving the day before their operation.Patients of the test group washed their skin with soap water or bathed at the operation day and patients in control group did this the day before operation.Bacterial culture was done in the operative site both before and after sterilization,and observed whether infection occurred in the incision after operation.RESULTS Bacterial culture in skin before sterilization was statistical significance between the two groups and no statistical significance after sterilization.All the incision healed well and no infection was found.CONCLUSIONS No-hair-shaving preoperatively can decrease patients′ discomfort and reduce colonies on skin surface effectively.The method is easy and safety manipulated.

GAP-43,netrin-1,collapsin-1,and neuropilin-1 have been regarded to play crucial roles in the formation of patterned neural connections.The cerebellum consists of five distinct concentric layers:white matter,internal granule layer (IGL),Purkinje cell layer (PCL),molecular layer (ML),and external granule layer (EGL) in young rodents.Cells in EGL are generated after birth.In contrast Purkinje neurons are born before birth,which receive main innervations of climbing fibers fi'om the inferior olivary nucleus and parallel fibers from the internal granule cells.These innervations are mostly established in the first three postnatal weeks,accompanying the sprouting and maturation of Purkinje cells.The potential roles of GAP-43,netrin-1,collapsin-1 and neuropilin-1 in the postnatal development of cerebellum remain unclear.To get insights into the above issue,the expression of GAP-43,netrin-1,collapsin-1,and neuropilin-1 mRNAs and proteins were examined in the cerebellum of mice at postnatal days (P) 5,P10,P20 and adulthood.The results showed that these four molecules were expressed in different temporal and spatial patterns in the postnatal cerebellum of mice,which was in match with axonal synaptogenesis,elongation and synapse formation during postnatal development and adulthood.By using double immunohistocbemistry,it was found that the Purkinje cells stained for GAP-43 were also positive for either netrin-1 or collapsin-1 at P10,and cells stained for collapsin-1 were also positive for netrin-1 or neuropilin-1.It was suggested that the four molecules are involved in the postnatal development of cerebellum.
9.Analysis of One Case of Irinotecan-induced Delayed Diarrhea

Objective:To analyze one case of delayed diarrhea caused by irinotecan. Methods:The pathogeny, mechanism, ge-netics and treatment of the case were analyzed. Results:Delayed diarrhea was the dose-limited toxicity of irinotecan, which was related with the cytotoxicity of the active metabolite 7-ethyl-10-hydroxycamptothecin (SN-38). Genetic polymorphism was one of important risk factors, especially UGT1A1 polymorphisms could be used as a predictor for the diarrhea. The pharmacotherapy of the diarrhea was ef-fective and rational, and the clinical pharmacist provided rational pharmaceutical care for the patient. Conclusion:It is very important to enhance pharmaceutical care for the patients treated with irinotecan.

OBJECTIVE To evaluate the feasibility and sterile effect of low-temperature plasma sterilizer produced by Chinese Academy of Sciences for the sterilization of arthroscope surgical instruments.METHODS To explore the cleaning,package,arrangement and sterile effect monitoring of instruments in the process of arthroscope sterilization using low-temperature plasma sterilizer.RESULTS In the sterilization process of arthroscope surgical instruments using low-temperature plasma sterilizer,the chemical indicators and indicating patch were discolored,the monitoring of bio-box and the sterilization monitoring of surgical instruments were all negative.CONCLUSIONS Using the low-temperature plasma sterilizer machine for the sterilization of endoscope surgical instruments can ensure the security of sterilization,make less injury of precise instruments,shorten the sterilization time,accelerate the surgical turnover,enhance the frequency of surgical instruments application,and increase the efficiency.
