1.Effect of GRP94 expression down-regulation on proliferation abilities of human lung carcinoma A549 cells

Objective:To investigate the effect of GRP94 gene expression down-regulation on the proliferation in human lung carcinoma A549 cells and possible mechanism.Methods:Expression of GRP94 was examined in 4 lung carcinoma cells and two lung bronchial epithelial cells by qRT-PCR and Western blot.Chemically synthesized siRNA targeting GRP94 gene and transfected into A549 cells used by Lipofectamine2000.The mRNA and protein expression levels of GRP94,c-myc and cyclinD1 were detected by qRT-PCR and Western blot.CCK8 assay and Colony formation assay were used to detect the effect of specific GRP94 siRNA on cell proliferation.Results:The levels of GRP94 expression in the four lung carcinoma cells were significantly higher than that in two lung bronchial epithelial cells (P<0.05).GRP94 siRNA significantly downregulated the levels of GRP94 mRNA and protein in A549 cells, and the downregulation evidently suppressed cell proliferation.However, The c-myc and cyclinD1 expression levels in A549 cells in siRNA-GRP94 group was significantly lower than that in siRNA-NC group (P<0.05).Conclusion:The levels of GRP94 expression in lung carcinoma cells were significantly higher than that in normal lung epithelial cells.Knockdown GRP94 expression significantly inhibit the level of lung carcinoma A549 cells proliferation,which may act via influencing the expression of c-myc,cyclinD1.

miR-200c is a member of miR-200 family,and it plays an important role in tumor diagno-sis,metastasis and chemoresistance.miR-200c is identified as a molecular marker and prognostic indicator in cancer screening.It can inhibit epithelial-mesenchymal transition,tumor invasion and metastasis,and increase the chemosensitivity of tumor cells,but the relevant mechanism is still not fully clear.The studies of miR-200c and some factors in regulatory pathway are beneficial to promote its applications in clinical diagnosis.

The association of serum adiponectin concentration with coronary heart disease (CHD) in Chinese Han population was evaluated. Present evidence demonstrated that the lowered serum adiponectin concentration was a susceptibility risk factor for CHD, while the precise relationship between serum adiponectin concentration and CHD in Chinese population requires further study.
5.CT portal venography manifestations of portal collateral circulation in patients with portal hypertension due to cirrhosis

Objective To analyze CT portal venography (CTPV) manifestations of portal collateral circulation in patients with cirrhosis by using a 16-detector row spiral CT scanner. Methods CTPV was performed in 36 patients with portal hypertension due to cirrhosis, the diagnosis was proved by clinical data, hepatic function findings and imaging signs. By using post-processing reconstruction technique, 3D images of portal venous system and portal collateral circulation were obtained. Results CTPV images displayed the portal venous system and its collateral circulation stereoscopically. Of 36 patients, left gastric varices were seen in 29 (80.6%), lower esophageal varices in 18 (50.0%), short gastric or posterior gastric varices in 15 (41.7%),paraesophageal varices in 9 (25.0%), gastro-renal or splenorenal shunts in 8 (22.2%), sponge-like transformation of portal vein in 7 (19.4%), paraumbilical and abdominal wall varices in 6 (16.7%), congenital cavernous in 6 (16. 7%) and paravertebral venous shunts in 4 (11.1%). Conclusion CTPV can well display the site, extent and severity of the portal collateral circulation in patients with portal hypertension due to cirrhosis, which is of great clinical importance for judging the patient's condition, for selecting therapeutic protocols and for estimating prognosis.
6.Determination of Ochratoxin A in Chinese Herbs by HPLC-MS/MS

Objective:To determine ochratoxin A ( OTA ) in Chinese herbs by high performance liquid chromatography-tandem mass spectrometry method ( HPLC-MS/MS) . Methods: The samples were extracted by 80% methanol water solution and purified by immunoaffinity column. The chromatographic separation was carried out on a C18 column, the mobile phase was methanol-acetonitrile-0. 01 mol·L-1 ammonium acetate, and then OTA was detected by MS/MS in an ESI(-)-MRM mode. Results:The limit of detection was 0. 1 μg·kg-1 , the average recoveries ranged from 84. 8% to 91. 2%, and the relative standard deviations ( RSD) ranged from 3. 6% to 8. 1%(n=9). Conclusion:The method is accurate,sensitive and simple, and suitable for the determination of ochratoxin A in Chinese herbs.

Objective To study the diagnosis and treatment of mesenteric diseases,and to enhance the understanding.Methods The clinical and histopathological data of 114 cases of mesenteric diseases were retrospectively analyzed.Results The percentage of mesenteric diseases was 0.06% of all inpatients during the same period;the causes were as follows,inflammation (53.5%),tumor (33.3%),lesion of vessels (8.8%) and others (4.4%).The lesion sites were mostly in the mesentery of small intestine (64.0%).The confirming diagnostic methods were exploratory laparotomy,laparoscopy,fine needle aspiration biopsy and ultrasonography.The therapeutic strategies were cause-oriented,but the prognosis of those with unknown causes was poor.Conclusion Mesenteric diseases are clinically rare,and the prognosis of some of the types is unfavourable.
10.Application of transthoracic echocardiography in interventional treatment of congenital heart diseases

Objective To study the value of transthoracic echocardiography (TTE) in interventional treatment of congenital heart diseases. Methods TTE was used to identify patients with atrial septal defect (ASD, n=26), ventricular septal defect (VSD, n=23) or ductus arteriosus (PDA, n=16). They received interventional treatment with domestic made nitinol occluder. The TTE was then used to monitor the entire process of the surgery and to assess the effect of the surgery. Results The treatment with indigenous nitinol occluder failed in five patients, including two with ASD, two with VSD, and one with PDA. The other sixty-one patients were freated successfully with the occluder. Once the procedure was completed, those patients were found to have neither abnormal valvular regurgination as shown by TTE, nor any shunt flow around the occluder as shown by both TTE and angiocardiography. One month later, TTE demonstrated that the position of all the occluclers remained stable, and no residual leakage or shunt was detected by TTE in all of the sixty-one patients, and the same results were obtained after three months. However, two days after the surgery, ECG showed that three of the patients with VSD suffered from different degrees of atrioventricular block on the second postoperative day, but they eventually recovered after treatment. The successful rate of the procedure was 93.8% (61/65). Conclusions Transthoracic echocardiography may play an important role at three stages (pre-, during- and after), i.e. to select the most suitable patients before the procedure, accurately identify the location, and release of the occluder at the end of the operation; and finally, it serves as a promising method to evaluate the treatment result in patients during their follow-up period.
