5.Inhibitive effect of photodynamic therapy with different parameters on caries development in rats

ObjectiveTo evaluate the inhibitive effect of Photodynamic therapy (PDT) with different parameters on dental caries formation in rats.MethodsWistar rats were infected with S.mutans to establish the rat caries mode 1.The rat's teeth were treated with 0.9% NS and 0.2% NaF as controls;laser,photosensitizer and laser plus photosensitizer,respectively as treated groups.Hematoporyrin monomethyl ether(HM ME) was used as photosensitizer and laser as the light source.The rats were sacrificed after five weeks.Keyes count was used to measure the inhibitory effect of PDT on dental caries.The surface morphology of the teeth was evaluated by scan electronic microscopy(SEM).Results The PDT-treated group and NaF-treated group showed significantly reduced Keyes scores compared with the negative controls (P<0.05).The laser-treated group,photosensitizer-treated group and NS-treated group showed no significantly reduced Keyes scores compared with the negative controls (P<0.05).The differences of Keyes scores between various parameter PDT treated groups were significant (P<0.05).Additionally,Keyes scores reduced gradually with the increasing of laser power.The outcome of SEM showed that the teeth treated with different laser powers exhibited different changes in morphology.Conclusion HMMEPDT had significant inhibitive efficacy on caries development in rats and its inhibitory effect presented close correlation with the power of laser,which indicates that it is important to apply appropriate power in clinical treatment.
6.Research on the apoptosis of malignant melanoma cell induced by transmembrane form of human blood group A mimotope vaccine

Objective To investigate the apoptotic effect of the transmembrane form vaccine of human blood group A mimotope on malignant melanoma cell line B16. Methods B16 cells were transfected with different recombinant plasmid through Lipofectamine 2000 and incubated with different concentration of monoclonal anti-A antibody at 2.5 μg/ml, 5 μg/ml,10 μg/ml and 20 μg/ml. Apoptosis rate of cells was determined with Annexin Ⅴ/PI double staining by flow cytometry. Results Apoptosis rate to P/F-M-pIRES group B16 cells was 74.74% when anti-A monoclonal antibody concentration was 10 μg/ml; apoptosis rate of plasmids carrying peptide/Fas fusion gene such as P/F-M-pIRES group and P/F-pIRES group were significantly higher than M-pIRES group and pIRES group. The apoptosis rate was statistically significantly different between different recombinated plasmid groups (F=669.707,P<0.01). The apoptosis rate was statistically significantly different between different antibody groups (F=106.596,P<0.01). The interaction between recombinated plasmid groups and antibody groups was statistically significant (F=34.806,P<0.01). Conclusions The transmembrane form vaccine of human blood group A mimotope could induce B16 cell apoptosis in vitro. This vaccine may be a promising candidate for potential malignant melanoma therapy.
7.Problem-based learning in clinical teaching ultrasonic therapeutic application

ObjectiveTo explore the PBL mode in training students' ability,improve the quality of teaching effect,in the teaching of Clinical therapeutic ultrasound and the teaching method based on problems with discussion method.MethodsTo 31 subjects of study who were randomly divided into five groups,each 6 people,a PBL teaching mode was used for 12 periods by proposing and study,group discussion,the whole class exchange and commenting on summary.ResultsThe survey shows that 93.6% students think the teachers' design has gained their ends,71% students are impressed by the knowlege and 96.8% of the students show the good learning attitude,64.5% of the students raised their interest in learning,which shows teaching results are effective.ConclusionPBL mode is superior to LBL teaching mode in training students' ability,improving the quality of teaching effect.
8.Study of signaling pathway of LDL and oxLDL inducing kidney tubular epithelial cells transdifferentiation

Objective To explore whether LDL and oxLDL may induce kidney tubular epithelial-mesenchymal transition (EMT) and its mechanism. Methods The second generation human kidney tubular epithelial cells (TECs) were cultured for 24 hours in different conditions as (1) serum free as control, (2) treated with LDL (50 ?g/ml) , (3) treated with oxLDL(50 ?g/ml), (4) treated with LDL(50 ?g/ml) plus PD98059(5 ?mlo/L) , (5) treated with oxLDL(50 ?g/ml) plus PD98059 (5 ?mol/L). The expression of cytokeratin, E-cadherin, ?-SMA and vimentin was assessed by immunofluorescence and Western-blot. Western-blot was also performed to test the expression of collagen I and phospho-ERKl/2MAPK and phospho-GSK-3?. Results oxLDL was more potently in inducing tubular EMT than LDL at 24 hours as demonstrated by de novo a-SMA expression, increased expression of vimentin, partial loss of cytokeratin and reduction of E-cadherin expression by TECs. The expression of collagen I and phospho-ERKl/2MAPK and phospho-GSK-3? was increased in TECs stimulated by LDL or oxLDL. MAPK inhibitor (PD98059) inhibited the phosphorylation of GSK-3P and almost completely blocked oxLDL-induced tubular EMT. However, PD98059 alone was able to inhibit LDL-induced tubular EMT partially. Conclusions oxLDL is more potently in inducing tubular EMT than LDL. The ERKl/2MAPK-GSK-3? signaling pathway mediates the LDL or oxLDL-induced tubular EMT.
9.Evaluation of fundus changes of pregnancy-induced hypertension syndrome by optical coherence tomography and contrast sensitivity

Background Pregnancy-induced hypertension syndrome (PIHS) appears retinopathy.Optical coherence tomography (OCT) and contrast sensitivity (CS) has been widely used in ophthalmology, but the application of OCT and CS in PIHS fundus lesions is less reported.Objective This study was to assess fundus changes of PHIS by using OCT and CS.Methods Two hundred and thirty-three eyes of 116 PIHS patients with fundus diseases were enrolled from January 2011 to January 2014 in the Cangzhou Central Hospital.One hundred eyes of fifty normal pregnant women were collected at the same period.Best corrected visual acuity (BCVA), fundus examination, OCT, CS were performed on these subjects.The CS values and BCVA between normal pregnant woment and PIHS patients were compared by independent samples t test.The correlation between BCVA grade and CS grade, OCT grade and CS grade, OCT grade and BCVA grade was calculated by trend x2 test and Gamma rank correlation analysis.The study was approved by the Ethics Committee of Cangzhou Central Hospital, all the subjects signed informed consent.Results OCT results showed that 172 eyes were abnormal (65.15%) ,including 94 eyes with neurosensory serous retinal detachment (54.65%) ,40 eyes with pigment epithelium and the ellipsoid zone change (23.26%) and 38 eyes with optic disc edema,retinal hemorrhage and other changes (22.09%).The CS values in each spatial frequency and the BCVA in the PIHS group were significantly lower than those in the normal control group (all at P < 0.05).The OCT examination result was positively correlated with BCVA in the PIHS group (G=0.807,x2=93.418,both at P =0.000).The CS value under the 6.0 c/d had the best correlation with BCVA (G=0.820,x2=97.397, both at P=0.000).The OCT examination result was positively correlated with CS under the 6.0 c/d(G=0.932,x2 =126.376,both at P=0.000).Conclusions Combination of fundus examination,OCT and CS can objectively and comprehensively assess the morphology and function of retinopathy in PIHS patients,which provide the basis for the clinical diagnosis and timely treatment.
10.Association between Angle class II division 2 malocclusion and congenital tooth anomalies

Objective: To study the relationship between Angle Class II division 2 malocclusion and congenital anomalies of dentition, Methods: 100 untreated patients (aged 12~42 years old) with Class II division 2 malocclusion were investigated by history,general examination,radiograph and tooth model. The data of congenital tooth anomalies were analysed.Results: Congenital tooth anomaly was found in 50 of the patients. Upper lateral incisor enamel hypoplasia was found in 7 patients, small size of maxillary lateral incisor in 28, canine impaction in 4, missing of lower incisor in 9, transposition in 2 and anterior tooth Bolton ratio discrepancies in 30 of the patients. Conclusion: Angle Class II division 2 malocclusion is closely related to congenital tooth anomalies. The main factors of anterior tooth Bolton discrepancies may be the small size of maxillary lateral incisor and absence of lower incisor.
