2.Improved methods of establishing rabbit VX2 hepatocarcinoma models

BACKGROUND:Rabbit VX2 hepatocarcinoma models are characterized by expanded and infiltrative growth, abundant blood supply, similar pathological and imaging features to human hepatocelular carcinoma, so this model is ideal for imaging experiments. OBJECTIVE: To establish rabbit VX2 hepatocarcinoma models and compare two different laparotomy methods in establishing rabbit VX2 hepatocarcinoma models. METHODS:Thirty New Zealand rabbits were randomly divided into two groups: traditional laparotomy (tumor particles were directly embedded into the liver with tweezers) and improved laparotomy (tumor particles were injected into the liver through a 15G syringe needle). The growth features of the tumor established in two different ways were compared by ultrasound. RESULTS AND CONCLUSION:The tumor formation rate was 100% in the two groups; the single rate was respectively 50% and 90%; the ectopic implantation rate was 50% and 10%, there were significant differences between two groups (P < 0.05). The mortality rate within 2 weeks was 33% and 0 in the two groups, with significant differences (P < 0.05). The improved laparotomy method is feasible for modeling rabbit VX2 hepatocarcinoma, due to minimal injury, short operation time, high success rate and stable features.
3.Protective effects of total extract of astragalus(TEA) onprimary rats hepatocytes injured by CCl4 or H2O2

Aim To study the protective effect of total extract of astragalus(TEA) on primary rat hepatocytes injured by CCl4 or H2O2 and its mechanism.Methods The primary hepatocytes were isolated by Ⅳ collaganase and injured by CCl4 or H2O2.The contents of malondialdehyde(MDA) and glutathion(GSH),glutathion peroxidase(GSHpx) activity of hepatocytes and the AST and /or ALT level in cultural supernatant were determined by general methods. Results (1) The elevation of MDA content of hepatocytes and AST level in supernatant of cultural hepatocytes,and the loss of GSH content and GSHpx activity induced by CCl4 were restored remarkably by TEA(5~80 mg?L-1);(2)The elevation of ALT level in the supernatant of hepatocytes and MDA content of hepatocytes, and the loss of GSH content and GSHpx activity induced by H2O2 were improved significantly by TEA(5~80 mg?L-1) treatment.Conclusion The results suggest that TEA possess direct protective action on primary hepatocyte in vitro injured by CCl4 or H2O2. These might be associated with its anti-oxidative activity.
4.The expression of ET-1 and ETA/B receptors mRNA in benign prostatic hyperplasia and its significance

Objective To investigate the expression of endothelin 1 (ET 1) and ETA/B receptors mRNA in prostatic tissues and its clinical significance in patients with benign prostatic hyperplasia (BPH). Methods Immunohistochemical staining and RT PCR were used to detect the expression level of ET 1,ETAR mRNA and ETBR mRNA,respectively.The detection results were analyzed in correlation with the clinical parameters of BPH patients. Results The expression level of ET 1 and ETA/BR mRNA in BPH tissue(integral optical densities,0.94?0.08,0.64?0.08,0.97?0.08,respectively)was higher than that in normal prostatic tissue(0.57?0.06,0.37?0.05,0.51?0.04,respectively).In BPH group,the expressed quantities of ET 1 and ETAR mRNA showed positive correlation with IPSS,prostatic volume,prostatic urethral length,prostatic urethral pressure and maximum urethral pressure of BPH patients,but it showed negative correlation with maximum flow rate and average flow rate of them. Conclusions Increased expression of ET 1 and ETAR might play a role in bladder outflow obstruction(BOO) of the patients with BPH.
5.Peripherally inserted central catheter in severely ill patients: A prospective,randomized,controlled study

Objective To compare the clinical application between peripherally inserted central catheterization(PICC) and subclavian central venous catheterization in severely ill patients.Methods A prospective study was carried out in 80 severely ill patients who had been randomly divided into two groups: receiving either PICC(PICC Group,40 patients) or subclavian central venous catheterization(Subclavian Group,40 patients).The success rate,the puncturing time,and the incidence of adverse reactions in the two groups were observed.Results The rate of successful puncture on one session was higher in the PICC Group(92.5%) than in the Subclavian Group(75.0%)(?2=4.501,P=0.034).And as compared with the Subclavian Group,the PICC Group reported a shorter puncturing time(15.7?5.3 min vs 23.9?6.3 min;t=-6.263,P=0.000) and a lower rate of adverse reactions(10.0% vs 27.5%;?2=4.021,P=0.045).Conclusions Use of peripherally inserted central catheterization is simple and safe to perform,superior to subclavian central venous catheterization.
6.Relationship between ambulatory arterial stiffness index and extent of coronary atherosclerosis

Objective To study the relationship between ambulatory arterial stiffness index and extent of coronary atherosclerosis.Methods All subjects were underwent coronary CT angiography.Ambulatory arterial stiffness index(AASI) was measured before coronary CT angiography.AASI value between different periods in each group were calculated and compared.Results AASI between different periods in the normal control group and coronary heart disease group existed statistical difference (P < 0.05).The extent of coronary sclerosis and AASI between different periods in different coronary artery lesions groups were statistically different(P <0.05),AASI was proportional to the degree of coronary sclerosis,24 h AASI,DAASI,NAA SI were related to extent of coronary atherosclerosis lesions,the correlation coefficient between NAASI and extent of coronary atherosclerosis was the largest.Conclusion AASI night is related to coronary atherosclerosis most closely,namely the higher the value of the nighttime AASI,the greater the degree of coronary atherosclerosis.
7.Transvaginal Surgery for Cesarean Scar Pregnancy: Clinical Analysis of 30 Cases

Objective To evaluate the clinical effect and safety of transvaginal lesion resection for cesarean scar pregnancy (CSP).Methods A total of 30 patients with CSP in our hospital received the transvaginal CSP lesion resection from January 2013 to January 2016 and were followed-up to observe the clinical efficacy and safety.Results The operations were successfully completed except one case of conversion to open surgery.The operation time was 30-120 min (53.4±26.0 min), the intraoperative hemorrhage was less than 50 ml, the hospital time was 3-7 d (4.3±1.3 d), and the postoperative time of serum β-hCG decreased to normal value was 14-36 d (24.1±13.2 d).No continuous pregnancy or complications such as incision non-healing, infection, or bladder injury after operation.Conclusion The clinical effect of vaginal CSP lesion resection is good and has fewer complications, being worthy of application.

9.Immortalization and screening of cardiac progenitor cells in mice

BACKGROUND:Transplantation of exogenous stem cells for functional cardiac celldeath or apoptosis easily induced complications after transplantation. Therefore, cardiac progenitor cells of heart itself have been ideal seed cells. OBJECTIVE:To establish stable celllines of cardiac progenitor cells from mouse heart, and to provide ideal cellmodels for studying proliferation and differentiation factors affecting cardiac progenitor cells during adult myocardial cells were damaged. METHODS:(1) Myocardiocytes were isolated from embryonic 15.5 days mice. (2) The cultured myocardiocytes were immortalized using retrovirus SSR69. Immortalized monoclonal myocardial cells were obtained using antibiotic selection and infinite dilution. (3) Monoclonal cellline with the property of cardiac progenitor cells was screened out. RESULTS AND CONCLUSION:(1) Myocardiocytes were successful y isolated and cultured. Partial cells showed obvious beating. (2) Myocardiocytes infected with retrovirus SSR69 were cloned and got 76 clones, then were named as CP15-#, (3) Screening the first 20 clones, the reasonable clones with the characteristics of cardiac progenitor cells were obtained according to myocardial cellmarker genes. The results suggested that immortalized cardiac progenitor cells were established mediated by reversible SV40 T antigen.
10.Expression of HDAC4 in human lung adenocarcinoma and its relationship with chemotherapy resistance

Objective There have been few research on the relationship between expression of HDAC4 and chemotherapy resistance in human lung adenocarcinoma.The present study aims to investigate the expression and clinical significance of HDAC4 in human lung adenocarcinoma tissues.Methods We selected 72 tissues in lung adenocarcinoma patients with docetaxel-resistant from January 2006 to December 2007 in Department of Oncology and Thoracic Surgery, Nanjing General Hospital of Nanjing Military Region, then evaluated the recent efficacy according to the RECIST criteria and divided the tissues into sensitive(n=32, included complete remission and partial remission) and insensitive(n=40, included stability and progress) groups.The expression of HDAC4 in tissues≥the HDAC4 optimal relative expression cut-off value(78.7) was high level HDAC4 group(n=35), otherwise it was low level HDAC4 group(n=37).QRT-PCR analysis was performed to detect the HDAC4 expression levels in sensitive group and insensitive group.Analyzed the progression free survival in high level HDAC4 group and low level HDAC4 group.Results The expression of HDAC4 was significantly higher in the insensitive group compared with the sensitive group [(1.42±0.30) vs (0.60±0.15), P<0.01].The median progression free survival was significantly shortened in the high level HDAC4 group compared with the low level HDAC4 group (10.2 months vs 5.8 months, P<0.05).ConclusionThe expression of HDAC4 increased in docetaxel-resistant lung adenocarcinoma patients, and it is expected to be a predictive indicator of the resistance of docetaxel.
