1.Effect of paeonol on adhesive function of rat vascular endothelial cells induced by lipopolysaccharide and co-cultured with smooth muscle cells.

OBJECTIVETo observe the changes in the adhesive function of vascular endothelial cells (VEC) and rat monocytes induced by lipopolysaccharide (LPS) and co-cultured with smooth muscle cells (SMC) and the intervention effect of paeonol (Pae).
METHODPrimary rat vascular endothelial cells (VECs) and rat vascular smooth muscle cells (VSMCs) were cultured by predigesting and adhering tissue blocks. The VEC-VSMC co-culture model was established by Transwell chamber. LPS was used to induce VEC injury. MTT assay and LDH assay were used to determine the VEC activity. ELISA assay was used to detect IL-1beta and TNF-alpha secreted by the VEC. The immunocytochemistry assay was carried out to detect the expression of ICAM-1. The Rose Bengal Staining was used to test adhesive function between VECs and monocytes.
RESULTThe concentration of LPS-induced VEC injury was 100 microg x L(-1), and the time was 7 h. after the intervention on the above cell model for 24 h, Paeonol (15, 30, 60 micromol x L(-1)) could effectively inhibit LPS-induced VEC injury and VEC injury, significantly enhance the survival rate of LPS-injured VECs, decrease IL-1beta and TNF-alpha secreted by the injured VEC, and reduce the expression of ICAM-1, so as to inhibit the adhesion of LPS-induced VECs and monocytes.
CONCLUSIONPaeonol could inhibit IL-1beta and TNF-alpha expression to protect VECs from being injured by LPS, and reduce ICAM-1 expression to inhibit the adhesion between VECs and monocytes.

2.Comparison of lOL Master contact and immersion A - scan ultrasound measurements of anterior chamber depth

AlM:To compare the results of lOL Master, contact and immersion A-scan ultrasound measurements for anterior chamber depth ( ACD ) , and evaluate the difference and consistency.METHODS:Fifty-eight cases (98 eyes) with age-related cataract during July to October in 2013 did the A-scan ultrasound with contact and immersion measurements and lOL Master to get the results of ACD. Difference in measurements between methods was assessed using the variance analysisi. Consistency was assessed using Bland-Altman.RESULTS:The ACD measured by lOL Master was 2. 31~3. 90mm, the mean was 3. 03 ± 0. 38mm. The ACD measured by contact A- scan ultrasound was 1. 51 ~4. 06mm, the mean was 2. 88 ± 0. 56mm. The ACD measured by immersion A-scan ultrasound was 1. 99 ~4. 17mm, the mean was 3. 17±0. 38mm. The results of lOL Master and contact A - scan ultrasound had statistical differences (P=0. 022<0. 05). The results of lOL Master and immersion A - scan ultrasound had statistical differences (P=0. 031<0. 05). The results of contact A-scan ultrasound and immersion A-scan ultrasound had statistical differences (P=0. 000<0. 05). The consistency between three methods was poor. CONCLUSlON: The rank of ACD of patients with cataract is immersion A-scan ultrasound, lOL Master and contact A-scan ultrasound. The consistency is poor, and the three methods can’t be interchanged clinically.

Prostate cancer is one of the most common malignant tumors in the male urinary system as well as the second leading cause of cancer death in men. Prostate specific antigen (PSA) screening is the main method for the early diagnosis of prostate cancer, but has a low specificity for its detection. In recent years, a variety of tumor markers with high sensitivity and specificity have been found. This review focuses on some of the more promising tumor biomarkers such as prostate cancer antigen 3, early prostate cancer antigen, prostate-specific membrane antigen, alpha-methylacyl-CoA racemase, and vascular endothelial growth factor.

5.Nonspecific apoptosis induced by long, double-stranded RNA in human osteosarcoma cells

Objective To construct the plasmid that can produce long,double-stranded RNA and identify nonspecific apoptosis induced by long, double-stranded RNA in human osteosarcoma cells. Methods Plasmid pCI-neo-dsRNA was constructed by two promoters driven DNA sequence based on nontoxic, completely exoteric gene EGFP from the head and the end simultaneously to synthesize sense and antisense RNAs. Human osteosarcoma cells MG63 were transfected with pCI-neo-dsRNA and 48 h later the transfected cells were assessed by Apoptosis Detection System, FCMand MTT method. Results Plasmid pCI-neo-dsRNA was constructed successfully, MTT measurement showed that the absorbance of cells decreased gradually with the increase of plasmid pCI-neo-dsRNA dose(0.05 ?g, 0.10 ?g, 0.15 ?g, 0.20 ?g,0.25 ?g,0.30 ?g). MG3 transfected with pCI-neo-dsEGFP presented a great amount of irregular green fluorescence that indicated cell apoptosis. As the quantity of dsRNA increased, the percent of apoptosis cells also raised. While the dose of pCI-neo-dsEGF was 3.0 ?g, the apoptotic results of pCI-neo-dsEGF in MG3 were similar to that of 5-FU. Conclusion Long, double- stranded RNA could trigger human osteosarcoma cell apoptosis and the further research on using long, double-stranded RNA in osteosarcoma gene therapy can be done based on this theory.
6.Finite element analysis of the maxillary central incisor with crown lengthening surgery and post-core restoration in management of crown-root fracture

Objective:To construct the finite element models of maxillary central incisor and the simu-lations with crown lengthening surgery and post-core restoration in management of different crown-root fracture types, to investigate the stress intensity and distributions of these models mentioned above, and to analyze the indications of crown lengthening from the point of view of mechanics. Methods:An extrac-ted maxillary central incisor and alveolar bone plaster model were scanned by Micro-CT and dental impression scanner ( 3shape D700 ) respectively. Then the 3D finite element models of the maxillary central incisor and 9 simulations with crown lengthening surgery and post-core restoration were construc-ted by Mimics 10. 0, Geomagic studio 9. 0 and ANSYS 14. 0 software. The oblique static force (100 N) was applied to the palatal surface ( the junctional area of the incisal 1/3 and middle 1/3 ) , at 45 degrees to the longitudinal axis, then the von Mises stress of dentin, periodontal ligament, alveolar bone, post and core, as well as the periodontal ligament area, were calculated. Results:A total of 10 high-precision three-dimensional finite element models of maxillary central incisor were established. The von Mises stress of models: post >dentin >alveolar bone >core >periodontal ligament, and the von Mises stress increased linearly with the augmentation of fracture degree ( besides the core) . The periodontal ligament area of the crown lengthening was reduced by 12% to 33%. The von Mises stress of periodontal ligament of the B2L2c, B2L3c, B3L1c, B3L2c, B3L3c models exceeded their threshold limit value, respective-ly. Conclusion: The maxillary central incisors with the labial fracture greater than three-quarter crown length and the palatal fracture deeper than 1 mm below the alveolar crest are not the ideal indications of the crown lengthening surgery.
7.Antitumor Activity of the Conjugates Composed of C1027, an Anticancer Antibiotic, and Monoclonal Antibody

C1027, an anticancer antibiotic, showed extremely potent cytotoxicity against cultured cancer cells, being over 10,000-fold more potent than adriamycin. C1027 acted rapidly; DNA synthesis of hepatoma cells was suppressed within 10 mintus of exposure. C1027 consists of two moieties, a new enediyne chromophore and an apoprotein; the chromophore serves as the active part of the molecule. C1027 molecule can be dissociated and reconstituted. The reconstituted C1027 was as potent as natural C1027. Through a newly developed method, enriched conjugation C1027 was conjugated to monoclonal antibodies. As determined by clonogenic assay, the conjugates displayed highly-potent, specific cytotoxicity to target cancer cells. 8.Correlation between the estrogen replacement therapy and the prevalence of female with Alzheimer's dis-ease:a meta-analysis

Objective To investigate the relationship between estrogen replacement therapy and female patients with Alzheimer's risk. Methods We searched the PubMed, Springer, CBM, CNKI, VIP database for studies on the es-trogen replacement therapy and women with Alzheimer's disease, retrieval time from database to 2012 December. We evaluated the methodological quality of the included studies, used RevMan 5.0 software to analyze the extracted data. Results We found seven case-control studies which included 1392 patients and 2719 controls. The meta analysis re-sults show that the proportion of estrogen replacement therapy in AD group is significantly lower than that of the con-trol group, the difference has statistically significant [OR=0.68, 95%CI(0.50,0.90), P=0.01]. Conclusions estrogen re-placement therapy has effects on female patients with Alzheimer's disease, and is a protective factor.
9.Transnasal operations on the sphenoidal sinus and the middle fossa under nasal endoscope combined with microscope

Objective To investigate the surgical method of transnasal operations on the sphenoidal sinus and the middle fossa under nasal endoscope combined with microscope. Methods The operation was performed under nasal endoscope on the side of larger nasal cavity. The middle nasal turbinate was pushed outwards. Then the Handy's expander was inserted between the middle nasal turbinate and the nasal septum in order to get a wide visualization. The sphenoidal frontal wall was opened directly. The nasal endoscope and microscope were utilized in turn to complete the resection of lesions. Results The symptoms disappeared postoperatively in 10 cases of solitary sphenoidal sinusitis. The lesions of 6 cases of sphenoidal cyst and meningioma were all surgically removed on one session. Among 32 cases of pituitary adenoma, a total resection was carried out in 17 cases, a subtotal resection in 12 cases, and a partial resection in 3. After surgery, a supplemental X-knife radiosurgery was employed. Postoperative follow-up in the 48 cases for 0.5~3.5 years (mean, 2.5 years) showed no recurrence of sphenoidal cyst, sphenoidal sinusitis, or meningioma, and 3 cases of recurrence of pituitary adenoma. No intracranial infection, adhesion of nasal cavity, or nasal bleeding was noted. Conclusions Transnasal operations on the sphenoidal sinus and the middle fossa under nasal endoscope combined with microscope has advantages of mild invasion, little blood loss, short operation time, and good outcomes.

Objective To investigate the histomorphology,cellular biocompatibility in vitro and tensile force resistance property of fibrin-binding amniotic membrane(FBAM)and to explore its feasibility and superiority using as ocular surface graft.Methods After observing the morphology of FBAM under light microscope and scanning electron microscope,we did primary culture of corneal epithelial cells,taking monolayer fibrin-binding amniotic membrane as carrier,then observed cell growth overlay area.The elongation nature of FBAM was tested on electronic universal testing machine by calculating elastic modulus,using bilayer amniotic membrane(BAM)and monolayer amniotic membrane(MAM)as controls.Results The fresh FBAM was integrated as a whole,with structurally complete fibrin and amniotic membrane as well as tight binding of fibrin to amniotic membrane.The reserved FBAM was structurally complete,brittler than the fresh FBAM,while amniotic membrane still bound tightly to the fibrin,but under microscope the fiber reticular formation was fairly fuzzy.Single layer of epithelial cells was formed on FBAM after about seven-day culture in vitro.Disparity of cell growth overlay area was significant(P
