1.The levels of serum lactate, lactate clearance rate and transaminase in septic rats and their relationship with liver damage

Objective To explore the serum lactic acid,transaminase and their relationship with liver pathological damage in sepsis rats,whether a correlation exists between serum lactate clearance rate and transaminase in sepsis rats,and whether they can be used as indicators for the prediction and evaluation of septic rats liver injury.Methods A total of 150 clean Sprague-Dawley (SD) rats was divided into normal group (n =50),sham operated group (n =50),and the cecal ligation and puncture (CLP) (n =50).Ten rats were killed after successful surgery at the 6th,12th,24th,48th,and 72nd hour in CLP group,respectively.At each time point,10 normal rats and 10 sham-operated rats were taken as a control in the corresponding time point.The liver tissue was obtained for pathological analysis.The levels of lactate and liver transaminase were detected and the lactate clearance rate was calculated.The levels of lactate and transaminase at each time point were statistically compared.The correlation analysis was performed among serum lactate,transaminase,and liver damage pathological changes.Results The rat hepatocytes in CLP group begin to appear at the 6th hour,and the damage was gradually enhanced at the 12th,24th,and 48th,and up to the worst damage at the 48th time point.The levels of lactate and alanine aminotransferase (ALT)in CLP group at the 12th and 24th hour were significantly elevated compared with the normal and sham operation groups (P <0.05); The level of aspartate aminotransferase (AST) in CLP group at the 6th,12th,24th,48th,and 72nd hour was significantly elevated compared with the normal and sham operation groups (P < 0.05).A positive correlation was found between the levels of ALT and lactate (r =0.766,P <0.05),and a negative correlation was found between lactate clearance rate and ALT (r =-0.712,P <0.05).Conclusions In septic rats,both lactate and lactate clearance rate were correlated with ALT,and they were correlated with liver pathological damage.The level of lactate,lactate clearance rate,and ALT could be used as the key indicators to predict liver damage in septic rats.
2.Chemical constituents from Clerodendrum canescens

Objective To investigate the chemical constituents in the whole plant of Clerodendrum canescens. Methods The compounds were isolated by column chromatography over silica gel or Sephadex LH-20 gel and their structures were elucidated by chemical and spectroscopic methods. Results Seven triterpenoids were isolated and identified as: lupeol (Ⅰ), ?-amyrin 3-undecanotate (Ⅱ), lupeol acetate (Ⅲ), lupeol 3-palmitate (Ⅳ), melastomic acid (Ⅴ), ?-amyrin acetate (Ⅵ), betulinic acid (Ⅶ), respectively. Conclusion Among those compounds, compound Ⅱ is a new one, while compounds Ⅳ-Ⅵ are isolated from the plants of Clerodendrum Linn. for the first time.

4.Cryptotanshinone effects on the proliferation and apoptosis of renal carcinoma stem cells

Abstract BACKGROUND:Previous studies have found that cryptotanshinone represses multiple tumors, but little is reported on its effect on renal carcinoma. OBJECTIVE:To explore the effect of cryptotanshinone on the proliferation and apoptosis of the renal carcinoma stem cels. METHODS:CD133+ renal carcinoma stem cels were separated from OS-RC-2 cels by immunomagnetic bead separation. Effects of 0, 0.2, 1, 5 mg/L cryptotanshinone on the proliferation and apoptosis of CD133+ renal carcinoma stem cels were detected by MTT and flow cytometry, respectively. Expression levels of Ki67, Bcl-2, Caspase-3 and p-Caspase-3 protein were detected by western blot assay. RESULTS AND CONCLUSION:After magnetic cel sorting, the percentage of CD133+ cels was increased from 6.32% to 82.73%, and there was a significant difference before and after cel sorting (P < 0.001). Cryptotanshinone could repress the proliferation of CD133+ renal carcinoma stem cels and promote cel apoptosis in a dose-dependent manner. The protein expression levels of Ki67 and Bcl-2 in the 5 mg/L cryptotanshinone group were significantly decreased compared with the control group, while the protein expression level of p-Caspase-3 protein was significantly increased. In addition, there was no difference in the protein expression of Caspase-3 between cryptotanshinone and control group. These findings indicate that cryptotanshinone may be a potent anticancer drug for the treatment of renal carcinoma by inhibiting expression of Ki67 and Bcl-2 and promoting protein expression of p-Caspase-3. 6.Effect of Motor Relearning Program on the Balance Function of Stroke Patients

Objective To observe the effect of motor relearning program (MRP) on balance function of stroke patients.Methods Sixty stroke patients were randomly divided into the MRP group and control group with 30 cases in each group. The patients of the control group were trained with proprioceptive neuromuscular facilitation (PNF) therapy; those of the MRP group were trained with the MRP therapy. The balance function of all patients was assessed with the Fugl-Meyer Balance Function Assessment before and after training.Results After training, the scores of Fugl-Meyer Assessment of all patients increased ( P<0.05), but scores of the patients of the MRP group were significantly higher than that of the control group ( P<0.05).Conclusion The MRP training can improve the recovery of balance function of stroke patients.
7.A neonatal mouse model of AIDS induced by L6565 murine leukemia virus

Aim To establish a mouse model of AIDS and observe the effects of Combivir,an anti-AIDS drug.Methods The model was set up by infecting infant mice with L6565 murine leukemia virus.Quantitative RT-PCR method was used to measure viral load in plasma.The percentage of CD4~+ and CD8~+ lymphacytes was estimated by flowcytometric method.The number of WBC,RBC and marrow karyote was counted.indexes of immune organs were weighed and calculated.Results Compared with control,the viral load in plasma in the model was at high level.The percentage of CD4~+ lymphocyte,the ratio of CD4~+/CD8~+,the number of RBC and marrow karyote,and thymus index were significantly decreased in the model,while the number of WBC and spleen index was remarkably increased.Conclusion AIDS patient symptom is shown in mouse infected with L6565 murine leukemia virus.This suggests that this model is likely to be used as a mice model to evaluate the effect of anti-AIDS drugs.
8.The correlation between patent ductus arteriosus and B-type natriuretic peptide as weel as N-terminalpro-brain natriuretic peptide in preterm infants

Patent ductus arteriosus(PDA) is a common condition that primarily occurs in preterm infants who with low birth weight,which closely related to several co-morbidities(intracranial hemorrhage,necrotizing enterocolitis,pulmonary edema/hemorrhage,bron chopulmonary dysplasia,and retinopathy).Although the color Doppler ultrasound assessment can be used as the gold standard to determine the present of PDA,it is still very hard to fully reflect the hemodynamic change occured in PDA case.In recent years,B-type natriuretic peptide (BNP) and N-terminal pro-brain natriuretic peptide (NT-proBNP),two kind of biological markers,were used in the premature infants to assess the present,shunt volume,progress and durg or surgical intervention indication of PDA.In this paper,the corrections of PDA between BNP and NT-proBNP were reviewed briefly.
9.Relationship between tumor necrosis factor-αgene-308 promoter polymorphism and susceptibility to development of bronchial asthma in Uighur and Kazakh population in Xinjiang

Objective To investigate the relationship between the polymorphism of the tumor necrosis factor-αgene-308 promoter and bronchial asthma (BA)in Uighur and Kazakh population in Xinjiang.Methods PCR product sequencing method was used to detect the polymorphism distribution of TNF-αgene in 60 BA patients and 60 controls among Xinjiang Uighur and Kazakh population.Then we analyzed the association between different TNF-αgenotypes and BA in Uighur and Kazakh population.Results The distribution of GG,GA and AA genotype was significantly different between the two groups.Statistical analysis showed that BA group had a significantly higher TNF-αA allele frequency than that of the control one (P <0.05).After adjustment for sex and age,we found the A allele was a risk factor for BA pathogenesis (P <0.05).Conclusion The polymorphism of TNF-α gene-308 may be associated with the susceptibility to BA in Xinjiang Uighur and Kazakh population,so TNF-α gene-308 may be considered as a genetic marker for early identification of individuals at high risk for BA,which may play an important role in preventing the development of BA in clinical practice.
10.Association between early arterial blood lactate clearance rate and central venous oxygen saturation monitoring and prognosis in patients with severe pneumonia

Objective To explore the value of early arterial blood lactate clearance rate and central venous oxygen saturation (ScvO2 ) monitoring in patients with severe pneumonia .Methods A total of 56 patients with severe pneumonia treated during the period from January 2012 to December 2013 were reviewed retrospectively .The arterial blood lactate concentration and ScvO2 were determined immediately after admission to hospital and 6 hours after admission to calculate the 6‐hour lactate clearance rate .APACHE II score was evaluated at the same time .The patients were divided into three groups according to APACHE II score:10 to <20 (group A) ,20 to <30 (group B) ,≥30 (group C) .The three groups were compared in terms of early arterial blood lactate clearance rate and ScvO2 and estimate their association with the prognosis of patients .Results The initial arterial blood lactate concentration did not show significant difference between the three groups (P>0 .05) .The early lactate clearance rate and ScvO2 at 6 hours in group C were significantly lower than in group B .The values in group B were significantly lower than in group A (P<0 .05) .The mortality rate in group C was significantly higher than in group B ,and the mortality rate in group B was significantly higher than in group A (P<0 .05) .The APACHE II score of the dead patients was higher than that of the suevivors ,but early lactate clearance rate and ScvO2 level were lower than those of the survivors (P<0 .05) .APACHE II score was negatively correlated to early lactate clearance rate (r=0 .661 ,P<0 .01) and early ScvO2 level (r=0 .579 , P< 0 .01) .Conclusions Early lactate clearance rate and ScvO2 level are good indicators to reflect the severity of disease and predict the outcome in the patients with severe pneumonia .
