1.THE EXPRESSION OF LAMININ IN THE DEVELOPING KIDNEY OF MOUSE

Objective To observe whether laminin(LN) is expressed in the developing kidney of mouse and the changes of the expression associated with maturation and polarity. Methods Eight fetal and fourteen postnatal kidneys were examined by immunohistochemistry(Strepavidin Peroxidase conjugated method,SP),using polyclonal rabbit anti-mouse antibody. Results The expression of laminin in the kidneys increased with the gestational weeks of fetus and maturity of the kidney.The expression of laminin was positive in monocell,multicell without space and multicell with space.In the developing kidney,the laminin is expressed in the ureteric bud,glomerular basement membrane(GBM),tubular basement membrane(TBM),renal capsular basement membrane and interstitium.The expression of TBM was prior to the expression of GBM.Conclusion The expression of laminin in the developing kidney of mouse has chronological and spatial sequence.The laminin determines the polarity of cell.Laminin might play a key role during nephrogenesis.
2.The study of association of serum uric acid level with left ventricular mass and QRS duration in patients with chronic heart failure

Objective To investigate the association of serum uric acid (UA) level with ventricular hypertrophy,ventricular remodeling,and ventricular electromechanical activity dyssynchrony in patients with chronic heart failure by studying the correlation among left ventricular mass index (LVMI),QRS duration (QRSd) and UA.Methods Four hundred and fifty-three patients were selected in this study.Each patient received examinations of echocardiography,electrocardiogram (ECG),blood routine,and blood biochemistry.According to the different UA level,the patients were divided into group Ⅰ (UA≤360 μmol/ L),group Ⅱ (360 μmol/L < UA≤420 μmol/L),group Ⅲ (420 μmol/L < UA≤480 μmol/L),group Ⅳ (480 μmol/L < UA ≤540 μmol/L),and group Ⅴ (UA > 540 μmol/L).The differences in LVMI,QRSd,Left ventricular ejection fraction (LVEF),and NYHA class among five groups were compared,and the correlation among LVMI,QRSd and UA were analyzed.Results According to the different UA,from groups Ⅰ to Ⅴ,with increase in UA,LVMI values increased,there was significant difference among five groups [(93.89 ±30.25) g/m2,(94.44 ±32.46) g/m2,(101.04 ± 37.22) g/m2,(124.39 ± 63.22)g/m2,and (133.51 ±41.85) g/m2,respectively] (F =17.501,P < 0.01).UA level correlated positively with LVMI (r =0.329 P <0.01).According to the different UA,from groups Ⅰ to Ⅴ,with increase in UA,QRSd prolonged,there was significant difference among five groups [(93.10 ± 14.56) ms,(92.77 ± 21.12)ms,(94.00±19.95)ms,(97.55 ±20.37)ms,and (101.73 ±25.57)ms,respectively] (F=2.516,P < 0.05).UA level correlated positively with QRSd (r =0.167,P < 0.01).According to the different UA,from groups Ⅰ to Ⅴ,with increase in UA,LVEF was reduced [(54.94 ± 10.26) %,(53.06 ±13.43)%,(51.79 ±14.01)%,(45.82±13.90)%,and (4-3.76±12.01)%,respectively] (F=14.299,P < 0.01),and the proportion of NYHA class Ⅳ increased (3.85%,4.29%,4.00%,21.05%,and 67.11%,respectively) (x2 =326.566,P < 0.01).Conclusions With the increase in UA level in patients with chronic heart failure,LVMI value increased,QRSd prolonged,LVEF decreased,and cardiac function reduced.It indicated that the elevated level of UA might reflect the severity of the ventricular remodeling,and ventricular electromechanical activity in dyssynchrony and development of heart failure.
3.Influence of Nbn gene on the development of granular cells in hippocampus of neonatal mouse

Objective To investigate the influence of neuro-specific DNA repair gene Nbn on the development of granular cells in hippocampus of monatal mouse. Methods The hippocampus development of postnatal mouse was evaluated by comparing the morphological differences of granular cells in hippocampus between the Nbn gene knockout mice (Nbn-CNS-Del) and the controls (Nbn-CNS-Ctr). Serial sections, immunohistochemistry and stereology methods were used to quantitatively analyze the development of granular cells in hippocampus of mice on the postnatal day 7, 14 and 21 (P7d, P14d and P21d). Results In Nbn-CNS-Ctr group, the granular layer in hippocampus manifested "C" in shape, the section area of granular layer was increased, and so was the cell body of granular cells in hippocampus during P7d to P21d; the inner arm granular cells in granular layer of hippocampus emerged on P7d, and most obvious changes occurred during P7d to P14d. While in Nbn-CNS-Del group, the granular layer in "C" shape were seen to form in most of the specimens during P7d to P21d, though they were sparsely distributed. 5.SPERMICIDAL STUDY ON MANDELIC ACID IN VITRO

Mandelic acid(MA). the hydrolysate of a compound extracted from the leaves of prunus Persica B, possesses a strong spermicidal effect in vitro. In this experiment. the International Planned Parenthood Federation screening method for spermicides was used. Dilutions of MA, between 0.8mg/ml and 3.3mg/ml, totally immobilized human spermatozoa in less than 1 min. When the lowest spermicidal concentrotions of MA and boric acid against human spermatozoa were compared, it was found that MA was 40 times more potent than boric acid.
6.Effect of high glucose on the expression of activating transcription factor 4 in cultured retinal Müller glia cells

Objective To observe the effect of high glucose on the expression of activating transcription factor 4 (ATF4) in cultured retinal Müller glia cells.Methods The retinal tissue of Sprague Dawley (SD) rats was collected,and Müller cells were isolated and cultured.The glial fibrillary acidic protein (GFAP) and glutamine synthetase (GS) of Müller cells were identified by streptavidin-biotinperoxidase complex.Cultured rat Müller cells were divided into control group (5.5 mmol/L glucose),group A (20 mmol/L glucose),group B (30 mmol/L glucose) and group C (40 mmol/L glucose).ATF4 protein expressions in Müller cells of four groups were measured by Western blot four days after cultured.Results GFAP and GS expressed in more than 95% of Müller cells.Over 95% of Müller cells of group A,B and C were positive for GFAP and GS.Western blots indicated that ATF4 protein in group A,B and C increased obviously compared with the control group (q=0.293,0.754,0.484; P<0.05).Conclusion High glucose can increase the expression of ATF4 protein and cause endoplasmic reticulum stress in retinal Müller glia cells in vitro.

8.The Expression of Metallothionein-1 in Mouse Kidney

Objective To investigate the expression and cellular localization of metallothionein-1 (MT-1) in mouse kidney. Methods The adult male CD-1 mice were selected as experimental animals. The protein of MT-1 in mouse renal cortex and medulla was detected by Western blot assay. Immunohistochemistry and immunofluorescence methods were ap-plied to observe the location of MT-1 in renal tubules and collecting ducts of mouse kidney. Double-immunofluorescence and confocal laser scanning microscopy were used to observe the subcellular localization of MT-1 in the epithelial cells of re-nal tubules and collecting ducts of mouse kidney. Results The expression of MT-1 was detected in cortex and medulla of mouse kidney. The expression of MT-1was mainly found in proximal convoluted tubules and distal convoluted tubules of cor-tical labyrinth, and proximal straight tubules, distal straight tubules and collecting ducts of ray medullary and medulla of kid-ney. The strongest expression of MT-1 was found in proximal tubules, secondary in distal tubules and last in collecting ducts. The intracellular location of MT-1 was in brush border of proximal tubules and intracytoplasm, in the distal tubule epi-thelial cell cytoplasm and in the basolateral membranes of collecting duct epithelial cells. The subcellular localization was found in the lysosome. Conclusion The different locations of MT-1 expressed in proximal tubules, distal tubules and col-lecting ducts may determine different roles of them in kidney.
