2.Ocular manifestations of acquired immunodeficiency syndrome

Objective To observe ocular manifestations of acquired immunodeficiency syndrome (AIDS). Methods Fourty-two AIDS patients (66 eyes)with ocular complaints received examinations of visual acuity, slit-lamp microscope, ophthalmoscope and fundus fluorescence angiography (FFA). The results were retrospectively analyzed. ResultsThere are five types of ocular findings, including cytomegalovirus (CMV) retinitis (37 eyes, 56.0%), retinal microvasculopathy of human immunodeficiency virus (21 eyes, 32.0% ), optic nerve diseases (three eyes, 4.5 % ), retinal neuroepithelial layer detachment (two eyes, 3.0%) and uveitis (three eyes, 4.5%). Conclusions The common ocular manifestations showed progressive necrotic retinitis, retinal hemorrhage and retinal vasculitis and attenuated, cotton-wool spots in AIDS patients.
3.Analysis of neo-adjuvant chemotherapy in the treatment of 32 cases of cervical cancer

Objective To evaluate the clinical efficacy of neo-adjuvant chemotherapy in the treatment of cervical cancer. Methods A total of 32 patients,of which 12 cases of Ⅰ b2,8 cases of Ⅱ a,5 case of Ⅱ b and 7 cases of Ⅲ, were all treated with 2 - 3 cycles of PF chemotherapy before treatment with cisplatin 100 mg/m2, d 1;5-fluorouracil 1000 mg/m2 ,d1-5. The chemotherapy was performed for 3 weeks followed by three weeks rest as one cycle. Results Short-term effect was 81.3% (26/32), with 3 cases of Ⅰ b2 and 1 cases of Ⅱ a had been completely relieved after 2 - 3 cycles of chemotherapies. Simultaneously, chemotherapy improved the removal rate of surgery. Among these 32 patients, there were 4 cases achieved complete response (CR) ,22 cases achieved partial response (PR) ,5 case of stable disease ( SD), 1 cases of progressive disease(PD). The overall response rate( CR +PR) was 81.3% (26/32) ,and the control rate ( CR + PR + SD) was 96. 9% (31/32). The main adverse events included myelosuppression and gastrointestinal response, which were all tolerable. Conclusions The neo-adjuvant chemotherapy reduced the size of cancer,which can improve removal rate of surgery and the quality of life.

6.Effects of interleukin-1 ,interleukin-6 and tumor necrosis factor-α on migration of human lens epithelial cells

Background Lens epithelial cells (LECs)play an important role in maintaining the lens transparency.Inflammatory cytokines have been clarified to participate in the formation of traumatic and after cataracts.However,the influence of inflammatory cytokines on the migration of LECs is still studying.Objective This study was to investigate the effects of interleukin-1 alpha (IL-1α),interleukin-1 beta (IL-lβ),IL-6,tumor necrosis factor alpha(TNF-α) and their mixture on the migration of human LECs in vitro.Methods HLE-B3 cells were cultured in DMEM containing 0.5% fetal bovine serum.10 μg/L of IL-1α,IL-1β,IL-6,TNF-α or the mixture was added in the medium for 24 hours,respectively.Wound-scratch assay and transwell monolayer permeability assay were used to evaluate the effects of different cytokines on LECs migration.The migrating cell numbers at the scratch zone and the transmembrane cell numbers at 12:00,3:00,6:00,9:00 and the center areas on the transwell chamber were calculated under the inverted microscope.In the control group,LECs were cultured in DMEM with 0.5% fetal bovine serum only.Results Wound-scratch assay revealed that 24 hours after cytokine incubation,the LECs numbers in the scratch zone were significantly less in the control group compared with the IL-1β group and TNF-αgroup (P =0.000,0.000),and those in the mixture group were higher than the IL-1β group and TNF-α group (P=O.000,0.000).However,no significant change was found in migrating LECs number between IL-1o group or IL-6 group and the control group (P =0.600,0.098).Transwell assay displayed that after 24-hour culture with cytokines,the largest density of transmembrane LECs was obtained in the mixture group,with a significant difference in comparison with the IL-1β group or TNF-α group (P=0.000,0.000).However,the transmembrane LECs were much more in the IL-1 β group or TNF-α group compared with the control group (P =0.000,0.000).Only a few transmembrane LECs were seen in the IL-1α group and IL-6 group in comparison with the control group(P =0.056,0.327).Conclusions IL-lα and IL-6 can not stimulate the migration of human LECs,but TNF-α and IL-1β can enhance their migration markedly.Mixture of these cytokines may play a much powerful influence on LECs migration.

10.Overview on method and strategy of therapeutic material basis in traditional Chinese medicine by multidisciplinary approach.

Traditional Chinese medicine (TCM) has a good reputation for preventing or healing diseases in clinic due to its higher efficacy, minor toxicity and abundant resources. Screening bioactive components in TCMs is not only crucial for clarifying their action mechanisms, but also the basis of their safety and quality control. TCM is characterized by multiple components, multiple targets and multiple mechanisms, however the complex composition of TCM makes it difficult to study the therapeutic material basis which has become the bottleneck in the process of its modernization and internationalization. Recently, with the rapid development of modern technologies and the unceasing progress of various disciplines, multidisciplinary approach, such as analytical chemistry, chemistry of TCM, pharmacology, cell biology, systems biology and bioinformatics has been successfully applied to the study of TCM. Multidisciplinary approach realizes the communication and interaction of multi-discipline, and accelerates the research and development of TCM. This review summarizes the application of multidisciplinary approach which may have certain potential of bringing new thoughts to TCM research and provide references for screening and identification of therapeutic material basis of TCMs.
