1.Research progress in metabolic targets for cancer therapy
Chinese Journal of Pharmacology and Toxicology 2017;31(3):269-278
Cancer cells can change metabolic pathways, including glycolysis and glutamine metabolism, and produce the raw materials needed for rapid proliferation and survival. Therefore, research on metabolic pathways of cancer cells might help find the targets of cancer therapy. In this review, we outlined the metabolic features of aerobic glycolysis, glutamine metabolism and (tricarboxylic acid) TCA cycle in cancer. We also described metabolic targets for cancer therapy and therapeutic agents for the corresponding targets in these metabolic pathways, and finally discussed some of the challenges related to tumor metabolism as a therapeutic target in cancer therapy.

Objective To summarize the experience of humane care on our voluntary blood donors. Methods From March 1,2017 to March 31,2017,281 blood donors were enrolled, called the intervened group, from the fixed blood donation street-spot to investigate their satisfaction and their willingness to donate blood again or not.Meanwhile,168 donors who were not intervened were also observed, called the control group. Results In the intervention group,182 were former blood donors, and 64.5% of the respondents believed that the staff was more trustworthy than before. And 78.2% of the respondents expressed that the communication ability with blood donors was better than before. The incidence of adverse effect of blood donation in the 281 donors was only 0.71%,and all of them were willing to donate again, which was better than the control group. Conclusion Humanistic care during voluntary blood donation is beneficial to improve the quality of service, the satisfaction of blood donation, and help retaining the blood donors.

Objective:To study the influence of Rhizoma Corydalis before and after vinegar processing on the microelement content in the eye tissues of rats to verify the vinegar processing leading the herb to liver meridian. Methods: The rats were divided into the crude Corydalis group, vinegar processing group and negative group. The content of microelement Cu, Zn and Mn in rat ocular tissues was determined and compared after the administration. Results:Corydalis could significantly increase the content of Cu and Zn in fun-dus blood and that of Zn and Mn in the eyeball vitreous, and Rhizoma Corydalis after vinegar processing showed more obvious effect. Conclusion:Rhizoma Corydalis with vinegar processing can improve the distribution of microelements in eye tissues, and because liver opens at eyes, the results provide evidence for Corydalis after vinegar processing leading to liver meridian.

Objective To evaluate the effect of positive experience intervention on anxiety, depression and quality of life among rural empty nesters. Methods A total of 115 rural empty nesters were divided into the intervention group (n=58) and the control group (n=57). Both groups received routine community activities. In addition, participants in the intervention group received positive experience intervention. Geriatric Depression Scale, Self-Rating Anxiety Scale and 12-item Short-Form Health Survey were adopted to measure the related items before and after the intervention. Results The intervention group showed a significant decrease in anxiety (1.54±0.07) and depression (7.28±3.82), compared with the control group (1.64±0.09, 11.81±6.38), the difference was significant (t=-5.960,-4.613, P<0.01). The Physical Component Summary (PCS) and Mental Component Summary (MCS) was 47.01±6.47 and 52.13±6.22 in the intervention group, which were significantly higher than those in the control group (41.37±8.71, 48.71±6.13), the differences were significant (t=3.937, 2.970, P < 0.01). Conclusions Positive experience intervention can decrease anxiety and depression, and improve quality of life of rural empty nesters.

Objective To establish a simple and reliable method for isolation and cultivation of epidermal stem cells from neo-natal rat skin basal layer .Methods The single cells were dissociated with twice trypsinization form neonatal rat skin .Thereafter we purified the basal layer stem cells with differential velocity adherent technique with collagen Ⅳ ,and the slow adherent cells were cultured as negative control cells .Both basal layer stem cells and control cells were cultivated with keratinocyte serum-free medium (K-sfm) .Stem cells were identified with β1-integrin and Keratin 19 by co-immunofluorescence assay ,and colony forming assay was executed to evaluate the proliferation potential of stem cells .Results The polygonal cells grew like flagstones ,with doubling time of approximately 24 hours .Both the morphology and growth properties of cells were in accordance with the character of basal layer stem cells .Co-immunofluorescence identification showed the cells were positive for the expression of β1-integrin and Keratin 19 . Basal layer stem cells had stronger clone forming ability in vitro compare with control group .Conclusion The results indicate that two-procedure trypsinization plus differential velocity adhesion is an ideal method for basal layer stem cells separation followed with vigorous vitality and reliable phenotype .

Objective To explore the feasibility of evaluating cardiac structure, coronary artery, pulmonary artery and cardiac function in one single scan by 320-row CT ECG-gated double phase cardiac function scan mode. MethodsForty patients underwent the 320-detector row CT double phase cardiovascular angiography. The pulmonary phase and aortic phase were reconstructed in order to evaluate the pulmonary and coronary artery. MPR reconstructions of both pulmonary and aortic phase were used to analyze the function of the two ventricles. And the results of the cardiac function were compared with those of transthoracic echocardiography. Thirty-five cases could be analyzed and diagnosed, while the other 5 cases had to be given up because of the poor imaging quality. The mean heart rate was (71.2 ± 11.2) beat per min (bpm). No arrhythmia case included. Results ( 1 ) Pulmonary embolism were diagnosed in 11 cases, coronary artery disease (CAD) were found in 5 cases, while post-stent implantation were observed in 7 cases. Six cases of congenital heart disease were diagnosed with 3 ASD and 3 primary pulmonary hypertension. Another one was diagnosed with left atrial myxoma, and 5 cases were pulmonary embolism associated with CAD. All of above cases were verified by final clinical diagnosis. (2) The heart function parameters including LVEDd , RVEDd, LVESd, RVESd and LVEF were (36.7 ±3.3), (43.3 ± 3.4) mm,(31.6±5.1), (41.3 ±5.1) mm and (47.1 ±15.1) for CT, while those were (40.3 ±3.1), (47.3 ±4.2) mm,(37.3 ±5.6), (45.3 ±3.3) mm,and (46.0 ± 14.8) for ultrasound, respectively. The CT results were correlated with the ultrasound ( n = 35, r = 0.886-0.988, P < 0.01 ). (3) The average radiation exposure was ( 5.4 ± 0.5 ) mSv. Conclusions 320-row CT ECG-gated double phase cardiac function scan mode is feasible for the "one-stop-shop" examination of the cardiovascular disease. This noninvasive method is recommended for the diagnosis, differential diagnosis, treatment and prognosis of cardiovascular disease.

OBJECTIVETo analyze the styryllactone components in Goniothalamus cheliensis Hu (Annonaceae).
METHODUPLC-Q-TOF-MS was used to identify the main styryllactone components in G. cheliensis. The chromatographic separation was performed on ACQU ITY UPLC BEH C18 column and eluted by actonitrile and 0.1% acetic acid in water gradiently. The mass spectrometer equipped with electrospray ionization souce was used as detector under the positive ion modes.
RESULTTwelve styryllactons were identified based on their MS data and published literatures, and the MS fragmentation regularity of the styryllactones was also proposed.
CONCLUSIONIt is an accurate and effective method to obtain the structural information of styryllactones.
