1.Effects of quickening blood and beautifying skin decoction on female's qi-stagnation and blood-stasis constitution

Objective To expolre the clinical effects of quickening blood and beautifying skin decoction on 20-50-year-old females with physical constitution of qi-stagnation and blood stasis.Methods According to the 2010 enactment of the Chinese Medical Association's Classification and Assessment of Chinese Constitution the standard identification of the physical object and physical scale iden tification,55 cases of 20 50 years old of Qi-stagnation and blood-stasis constitutions were collected and quickening blood and beautifying skin decoction treatment was given.25 g of the decoction were administrated each time,2 times a day,for 2 months.After that,the identification of physical constitution fitness was conducted,to find if this kind of beauty-keeping decoction could improves women's physical constitutions and some beauty-harm related physical symptoms.Results After taking the herbal decoction before and after treatment,the quality of Qi-stangnation and blood-stasis constitutions and factor of the beauty-harmful relevant symptoms were statistically significant differences (P<0.005).The total score of Qi-stangnation and blood-stasis constitutions and beauty-harm relevant symptoms showed that the total effective rate reached 61.82%.Conclusions The result indicates this quickening blood and beautifying skin decoction has a certain effect in improving the quality of qistangnation and blood-stasis situation and treating the beauty-harm relevant symptoms.The traditional Chinese medicine decoction has a certain effect on improving physical constitution which is abnormal.
2.Expression of tyrosine protein phosphatase 1B in adipose tissues of newly diagnosed diabetic patients

Objective:To investigate the role of protein tyrosine phosphatase 1B(PTP-1B) in pathogenesis of type 2 diabetes. Methods: Thirty-four patients were divided into 3 groups: type 2 diabetes with normal BMI group(CDM), obese type 2 diabetes group (ODM) and control group. Fasting blood glucose, fasting insulin and body mass were determined. Expression of PTP-1B in adipose tissues were determined by Western blot. Results: Compared with control group, CDM and ODM groups had insulin resistance, though there was no significant difference between CDM and ODM groups. Expression of PTP-1B in control, CDM and ODM groups were 3.39?0.94,11.57?1.89 and 36.53?14.82, respectively. 3.Treating high angle bimaxillary protrusion with three kinds of extraction method: a clinical study.

OBJECTIVETo compare the effect of three kinds of extraction model on high angle bimaxillary protrusion patients.
METHODSA total of 30 patients with Class I malocclusion and bimaxillary protrusion, aged 14-25 years old, were selected and divided into three groups. Four first premolars were extracted in the first group. The two maxillary first premolars and two mandibular first molars were extracted in the second group. The two maxillary first premolars and two mandibular first molars were extracted in the third group, and two additional micro-implants used as orthodontic anchorage in maxilla. Three groups were all treated with MBT appliance. Cephalometric analysis were carried out before and after treatment, and the results were analyzed with statistics.
RESULTS1) About the hard tissues, compared with the first group, there were statistically significant differences of N-Me, SGo/NMe, ANS-Me, FH/MP, SN/MP, and ODI in the second and the third group after treatment (P<0.01). 2) About the soft tissues, the teeth and the alveolar bone, compared with the first group, there were statistically significant differences of Pg-Pos, Li-SnPos, Si-LiPos, LL-E, L1-NB, L1/NB, U1/L1, L7-MP in the second and the third group after treatment (P<0.01). 3) All patients received consummate orthodontic treatment and obtained fine occlusion. Facial profiles were improved significantly after orthodontic treatment.
CONCLUSION1) After orthodontic treatment with mandibular first molars extraction, FH/MP, SN/MP, N-Me, ANS-Me, L1-NB and L1/NB decreased respectively, and soft tissue profiles were improved significantly. 2) Additional micro-implant used as orthodontic anchorage in maxilla significantly contributed to the maxillary incisor retraction and subsequent soft tissue change. 3) The first molars extraction and additional micro-implant used as orthodontic anchorage are efficient in improving the facial profiles for high angle bimaxillary protrusion patients.

4.The effect of Nifedipine and mechanical strain on matrix metalloproteinase expression in human periodontal ligament fibroblast.

OBJECTIVETo investigate the effect of Nifedipine on matrix metalloproteinase (MMP)-10 and MMP-13 expression in human periodontal ligament fibroblast (HPDLF) induced by mechanical strain in vitro.
METHODSHPDLF was divided into four groups at random by mechanical strain of elongation 0%, 8%, 12% and 16%. Each group was divided into four subgroups again by the concentration of Nifedipine 0, 10, 30, 50 microm. The cells were imposed to state mechanical strain 12 h after incubated with Nifedipine 1 h. And then immunohistochemical staining was used to investigate the expressions of MMP-10 and MMP-13 in intracytoplasm.
RESULTSIn 0% group, there was no significant difference of MMP-10 and MMP-13 expression in all subgroups (P>0.05). The expressions of MMP-10 and MMP-13 were high in group 8%, 12% and 16% without Nifedipine, and increasing significantly with elongation (P<0.001). The expressions of MMP-10 and MMP-13 decreased with dose increasing (P<0.001) in HPDLF after imposed to Nifidepine.
CONCLUSIONThe expressions of MMP-10 and MMP-13 induced by mechanical strain were inhibited by Nifedipine, which suggested calcium ions channels participate in expressions of MMP-10 and MMP-13 induced by loading.

5.Simulation of microenviroment after spinal cord injury in Sprague-Dawley rats

BACKGROUND:We built Sprague-Dawley rat models with mild, moderate, and severe spinal cord injuries to accord with the spinal cord injury types for basic empirical study, and consequently to further understand the microenvironmental change in Sprague-Dawley rats with spinal cord injury, and to provide help for clinical treatment. OBJECTIVE:To observe the changes in nerve function, pathological manifestation and motor sensory evoked potential in Alen’s models and Sprague-Dawley rats with complete spinal cord transection at different time points after spinal cord injury by simulating the microenviroment in Sprague-Dawley rats. METHODS: A total of 125 healthy adult female Sprague-Dawley rats were selected and randomly divided into group sham operation group, 100 gcf hit potential group (20 g×5 cm), 200 gcf hit potential (20 g×10 cm), 300 gcf hit potential group (20 g×15 cm), and spinal cord complete transection group with 25 rats in each group. At 1, 5, 7, 14 and 28 days after model establishment, the degree of spinal cord injury was identified by the BBB scores of motion function, motor evoked potential, and pathological section. RESULTS AND CONCLUSION:(1) Totaly 24 Sprague-Dawley rats died in the experiment. The death rate and the rate of complications were highest in the spinal cord complete transection group. The BBB score of each group was decreased. The BBB scores in every group increased as time went on. There were significant differences between each surgery group and the sham operation group at corresponding time points (P < 0.05). No significant difference was found between the 300 gcf hit potential group and the spinal cord complete transection group at corresponding time points (P > 0.05). (2) In each surgery group, the infiltration of inflammatory cels and obvious sweling of neurons were visible at 1 day after injury. Neural cels reduced with time prolonged. At 28 days after injury, a large number of astrocytes proliferated, scar and spinal cord cavity formed. Above symptoms were worse in the 300 gcf hit potential group and spinal cord complete transection group than in the 100 gcf and 200 gcf hit potential groups. (3) Significant differences in amplitude and latency were detectable between each surgery group and the sham operation group (P < 0.05). No significant difference in amplitude and latency was detected between the 300 gcf hit potential group and the spinal cord complete transection group at corresponding time points (P > 0.05). Results confirmed that hit potential of 20 g×5 cm, 20 g×10 cm and 20 g×15 cm can simulate the microenvironment of Sprague-Dawley rats with mild, moderate and severe spinal cord injury. The rate of complication was lower in modified Alen’s model of different hit potentials than in models of spinal cord complete transection, and was more accorded with basic research.
6.Inhibition of the invasion and migration of hepatocellular carcinoma cells by miR-148a and the mechanisms

Background and purpose: Primary liver cancer is the malignant tumor of liver cells or intrahepatic bile duct epithelium with familiar metastasis and postsurgical recurrence. The purpose of this study was to investigate the effects of miR-148a on the invasion and migration of hepatocellular carcinoma cells and the underlying mechanisms. Methods: The supernatant containing LV-miR-148a lentivirus particles was used to infect SMMC-7721 cells. The expression of miR-148a was determined by RT-PCR. Wound healing assay and transwell assay were performed to detect the effects of miR-148a on the invasion of hepatocellular carcinoma cells. Gelatin zymography assay was used to detect the effects of miR-148a on the enzyme activities of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9). The expression of MMP-2, MMP-9, E-cadherin and vimentin proteins was detected by Western blot assay. Results:RT-PCR showed the expression of miR-148a was upregulated in the infected SMMC-7721 cells. Transwell assay and wound healing assay showed ectopic expression of miR-148a suppressed cell migration and invasion abilities. miR-148a overexpression led to the decrease of the enzyme activities of MMP-2 and MMP-9 (P<0.05). Western blot assay showed that the protein expression of MMP-2, MMP-9 and vimentin proteins was signiifcantly decreased, the expression of E-cadherin had no changes. Conclusion:miR-148a is able to inhibit the migration and invasion of human SMMC-7721 cells in vitro, and the possible mechanisms may be related to decrease the enzyme activities of the MMP-2 and MMP-9 and the down regulation expression of MMP-2, MMP-9 and vimentin.
7.Analysis of clinical features of 27 children with primary immunodeficiency

Objective To investigate the detection and clinical features of primary immunodeficiency disorders (PID) in children for an earlier diagnosis of disease.Methods The clinical data of the 27 children with PID diagnosed in Shengjing Hospital Affiliated to China Medical University from Dec.2003 to Nov.2011 were reviewed,including illness history,birth history,family history,clinical feature,laboratory data,diagnosis,treatment and outcome,etc.Results In 27 children with PID,antibody deficiencies were the most frequent findings (48.15%,13/27 cases),followed by combined immunodeficiency (22.22%,6/27 cases),phagocytic disorders (14.81%,4/27 cases),and immunodeficiency with other major defects accounted for 14.81% (4/27 cases).PID was characterized by recurrent,severe and prolonged infection,but all kinds of PID had their own clinical features.Recurrent infections occurred in 24 cases.Respiratory infections and otitis media were the most common clinical manifestation.Seven patients had a family history.The fatality rate was 37.04% (10/27 cases).Conclusions There are vast varieties of PID in our area and antibody deficiencies are the most common type.All kinds of PID have their own clinical features,which may guide us to choose appropriate lab examination.There are nearly 25% patients with PID who have family history.The fatality rate is high.Patients who suffer from recurrent infections,especially respiratory infections or otitis media,or those with a family history should have early immunology testing so as to be detected and diagnosed of PID earlier.

To detect the effects of pioglitazone on adipose triglyceride lipase (ATGL) in adipose,liver,and muscle of high-fat diet-induced obese rats.Insulin resistance aud decreased expression of adipose triglyceride lipase were found in the adipose,liver,and muscle tissues.Pioglitazone may improve insulin sensitivity and ameliorate ATGL expression in tissues of high-fat diet-induced obese rats.
9.Maternal high-fat diet induces aberrant glucose and lipid metabolism in the early life of offspring and male susceptibility

Objective Maternal dietary modifications determine the susceptibility to metabolic diseases in adult life.However, whether maternal high-fat diet can induce early-onset aberrant glucose and lipid metabolism in offspring mice is less understood .Materials and Methods We examined the effects of feeding the pregnant mice with either a high fat diet or nornal chow diet throughout pregnancy and lactation on the glucose and lipid metabolism in their offsprings . Some glucose and lipid metabolism biomarkers and hepatic histology were analyzed .Results At weaning, for males of dams fed high-fat diet , they had heavier body weight , the blood glucose levels were significantly higher at 30 min ( P<0.001) and 60 min (P<0.01) after intraperitoneal glucose administration , and the AUC was also significantly larger (P=0.0016) and HOMA-IR was higher(P<0.05).For females of dams fed high-fat diet, they had higher blood glucose levels at 30 min ( P<0.01 ) after intraperitoneal glucose administration .No significant differences were observed of AUC and HOMA-IR.Both males and females of dams fed high-fat diet had elevated total cholesterol levels ( P<0.0001 and P=0.0004, respectively).Triglyceride levels showed no significant differences between the two groups of males and females . The offsprings of high-fat diet group indicated hepatic steatosis and there were no differences between males and females . Conclusions Maternal high fat diet during pregnancy and lactation can induce early -onset aberrant glucose and lipid me-tabolism in offspring mice , and the male offsprings are more prone to metabolic changes , such as obesity , glucose intoler-ance and insulin resistance .
10.Clinical characteristics and anti-infective treatment in Listeria monocytogenes meningitis

Objective To describe clinical characteristics and anti-infective treatment of Listeria monocytogenes(L.monocytogenes)meningitis.Methods Clinical manifestations and cerebrospinal fluid(CSF)examination results of 6 patients with L.monocytogenes meningitis in a hospital were analyzed retrospectively.Evaluation methods were developed according to relevant guidelines and literatures,anti-infective treatment and prognosis of patients with L.monocytogenes meningitis were evaluated.Results Two of 4 adults with L.monocytogenes meningitis had triad of fever,stiff neck,and altered mental status,the mean white blood cell count(WBC)of the initial CSF detection was 997×106/L,CSF/blood glucose ratio was 0.32,CSF protein was 1.43g/L;the other 2 neonates had fever,epilepsy,and hyponatremia,WBC were both>1 000×106/L,CSF protein were both>1 g/L,CSF/blood glucose ratio was<0.5.Of 6 patients,none were treated with appropriate initial empiric anti-infection therapy,confirmed by CSF or blood culture,5 cases were treated with ampicillin anti-infective therapy,1 used compound sulfamethoxazole due to penicillin allergy;1 neonate died,1 elderly patient was with moderate disability,the remaining 4 cases were all recovered and discharged from hospital.Conclusion Clinical manifestations and CSF findings of L.monocytogenes meningitis are not different from other purulent meningitis,commonly used antimicrobial agents for the treatment of purulent meningitis are not sensitive to L.monocytogenes,which should be paid attention in clinic.
