1.Role of stem cells, endothelial cells and cytokines in the development and regression of infantile hemangioma
Chinese Journal of Tissue Engineering Research 2014;(15):2434-2441
BACKGROUND:At present, there are few efficient therapies for infantile hemangioma, and the pathogenesis mechanisms remain unclear.
OBJECTIVE:To review the literatures available on the epidemiology, pathophysiology and pathogenesis of infantile hemangioma, to understand the research progress on the pathogenesis of infantile hemangioma, and to provide a reference for developing new drug therapies.
METHODS:A computer-based online search was performed in the PubMed database for literatures related to the pathogenesis, physiopathological features and epidemiology data of infantile hemangioma published from January 2009 to February 2014. The subject headings are“hemangioma, capil ary, classification, epidemiology, etiology, embryology, cytology, physiopathology, pathology, immunology, genetics, drug therapy, therapy”.
RESULTS AND CONCLUSION:Accumulating evidence has investigated the occurrence, development and regression of infantile hemangioma. However, no large-scale, multi-central epidemiology data are reported, and there is no theory explaining the pathogenesis of infantile hemangioma completely. Moreover, the relationship between al those theories about the pathogenesis remains unclear. The most important obstacle constraining the research is the lack of ideal animal model of infantile hemangioma. Due to the restrictions of nude mice models, it is imminent to develop new animal models for infantile hemangioma research.

Chinese Journal of Ocular Fundus Diseases 2015;31(3):230-234
Objective To observe the effect of micro-pulse laser in the treatment of acute central serous chorioretinopathy (CSC).Methods 105 cases (114 eyes) with clinically diagnosed acute CSC were included in the study, including 78 males (86 eyes) and 27 females (28 eyes) with an average age of (40.40±7.80) years, and mean duration of 26 days. All patients were examined for best corrected visual acuity (BCVA),slit lamp microscope and pre-lens,direct ophthalmoscopy, color fundus photography, fluorescein angiography combined with indocyanine green angiography (ICGA),and micro-perimetry.Patients were randomly divided into micro-pulse laser treatment group (treatment group,61 eyes) and control group (51 eyes). The age (Z =-0.374),gender ratio (x2=0.010),disease duration (Z=-0.525),BCVA (t=1.885),foveal thickness (CFT) (t=-1.754) and macular light sensitivity (t=1.255) were similar between the two groups. The micro-pluse laser treatment was performed with an 810 nm infrared diode laser at the active leakage site on retinal pigment epithelium guided by ICGA.The exposure time was 0.2 s,effective working time was 15%,the laser spot diameter was 100 μm,and the distance between 2 spots was 100 μm.The control group received pseudo-treatment using the same laser parameters. After 2 weeks,1month,3 months and 6 months of treatment, all patients were examined with BCVA,ocular fundus, optical coherence tomography and micro-perimetry.These parameters were compared between the 2 groups. Results The subretinal fluid of 39 eyes in the micro-pulse laser group and that of 3 eyes in the control group were absorbed completely. The cure rates in the micro-pulse laser group (61.9%) was higher than that in the control group (x2 =38.015,P<0.01).In the micro-pulse laser group, the mean BCVA was 67.81±11.70 at baseline, which increased significantly to 75±9.91,76.78± 9.43,78.56 ± 8.57 and 78.52 ± 8.60 at 2 weeks,1 month,3 months and 6 months after treatment respectively. In the control group, that was 63.86±10.35,64.20±11.43,63.90±10.88,64.55±11.04,64.10±11.12 at baseline,2 weeks,1 month,3 months,6 months post-treatment respectively. The mean post treatment BCVA at each time point were significantly higher in the micro-pulse laser group than that in the control group (P< 0.01).The mean CFT was (380.94 ± 50.73) μm at baseline, which reduced to (268.44±44.20),(242.78±41.31),(235.46±38.44),(235.56±38.71) μm at 2 weeks,1 month,3 months and 6 months post-treatment respectively in the nmicro-pulse laser group. In the control group, that was (397.98±52.61),(334.55±59.15),(316.16±55.25),(314.47±53.27),(321.51±55.74) μm at baseline,2 weeks,1 month,3 months,6 months post-treatment respectively. The mean post-treatment CFT at each time point in the micro-pulse laser group were decreased significantly compared to that in the control group (P<0.01).The mean central retinal sensitivity was (15.03 ± 2.00) dB at baseline, which enhanced to (17.06 ± 1.71),(17.37 ± 1.61),(17.56 ± 1.58),(17.48 ± 1.53) dB at 2 weeks,1 month,3 months and 6 months post-treatment respectively in the micro-pulse laser group. In the control group, that was (14.54±2.22),(14.80±2.16),(14.88±2.09),(14.82±2.07),(14.69±2.11) dB at baseline,2 weeks,1 month,3 months and 6 months post-treatment respectively. The mean central retinal sensitivity at each time point of post-treatment in the micro-pulse laser group were enhanced significantly compared to that in the controlled group(P<0.01).Conclusion In the treatment of acute CSC,micro-pulse laser can improve BCVA,reduce the mean CFT and improve the mean central retinal sensitivity. It is an effective and safe method to treat acute CSC.
3. Expression of cyclooxygenase-2 in carcinoma of the gastric cardia and its correlation with angiogenesis
Tumor 2008;28(9):791-794
Objective: To investigate the expression of cyclooxygenase (Cox)-2 in carcinoma of the gastric cardia, and to evaluate the relationship between Cox-2 expression and angiogenesis. Methods: Forty-six resected tumor specimens from patients with carcinoma of the gastric cardia were collected, and 21 corresponding paracancerous normal tissues were randomly selected as controls. Immunohistochemical staining was used for detecting the expression of Cox-2. Monoclonal antiboty against CD34 was used for labeling vascular endothelial cells, and microvascular density (MVD) was determined by counting CD34-positive vascular endothelial cells. The correlations of Cox-2 expression and MVD with clinicopathological parameters of the patients were analyzed. Results: The positive expression rate of Cox-2 and MVD value in the cancerous tissue were 80.4% and 31.95 ± 3.87, respectively, which were significantly higher than those in the pericancerous normal tissues (14.3%, 16.28 ± 1.55; P <0.01). The positive expression level of Cox-2 and MVD value were associated with clinical TNM staging and lymph node metastasis. The Spearman rank correlation test showed that tumor MVD were closely associated with Cox-2 expression. Conclusion: Abnormal expression of Cox-2 and angiogenesis play important roles in tumor invasion and lymph node metastasis of carcinoma of the gastric cardia.

Objective To construct an injectable controlled delivery system of paclitaxel based on thermosensitive PCL1250-PEG1500-PCL1250 hydrogels. Methods A thermosensitive PCL1250-PEG1500-PCL1250 triblock copolymer was synthesized by ring-opening polymerization of e-CL using PEG (Mw=l 500) as the initiator and Sn(Oct)2 as the catalyst. The synthesized PCL1250-PEG1500-PCL1250 copolymers were characterized for their composition, structure, and molecular weight via 1H NMR and GPC techniques. A series of Paclitaxel loaded hydrogels with various predesigned hydrogel concentrations and initial drug loadings were prepared to investigate their gelation ability, in vitro drug release behavior and in vivo biodegradability. Results The results calculated from 1H NMR and GPC indicated that EG/CL ratio(1.55) was consistent with the initial feed ratio(1.6), which offered a strong proof to their composition and molecular structure. The thermosensitive PCL1250-PEG1500-PCL1250 hydrogels exhibited a desirable sol-gel transition ability within the concentration range of 15%-30%. The in vitro release rate of paclitaxel from the paclitaxel/PCL1250-PEG1500-PCL1250 hydrogels was controllable by altering the hydrogel concentrations and initial drug loadings. The PCL1250-PEG1500-PCL1250 hydrogels showed a good in situ gelation ability after subcutaneously injected into mouse back. The in situ formed hydrogels gradually degradated with time and almost disappeared after 45 days in vivo. Conclusion Both the controllable drug release behavior and promising biodegradability of this new thermosensitive PCL1250-PEG1500-PCL1250 hydrogels paved a way to develop a novel delivery system for paclitaxel.
