1.Expression of 5-lipoxygenase in colon mucosa of ulcerative colitis

Objective To investigate the expressions of 5-lipoxygenase (5-LOX) protein and mRNA in colon mucosa of ulcerative colitis (UC) and to analyze its correlation with endoscopic and histological grading. Methods The biopsies from 32 UC patients and 26 healthy controls were collected, and the expressions of 5-LOX mRNA and protein in colonic mucosa was determined by real-time fluorescent quantitative reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry assay, respectively. The endoscopic and histological grading of 32 UC patients were also recorded. Results According to endoscopic grading, there were 10 cases of grade Ⅰ , 19 of grade Ⅱ and 3 of grade Ⅲ in 32 UC patients, and there were 19 cases of grade Ⅰ , 9 of grade Ⅱ and 4 of grade Ⅲ according to histological grading. The expressions of 5-LOX mRNA and protein in UC patients were significantly higher than those in healthy controls (P <0. 05 ). The rate of 5-LOX protein expression increased with endoscopic and histological grades, which was also positively correlated with endoscopic grades ( P < 0. 05 ), but not with histological grades ( P >0. 05 ). Conclusion The expressions of 5-LOX mRNA and protein in colon mucosa from UC patients might be a marker for disease activity. 5-LOX may play a pivotal role in inflammation of UC and be a target site for treatment.
2.Practice and reflection on the performance evaluation index system of scientific research in a general hospital

Objective Implementing the scientific research performance evaluation indicator system constructed to evaluate the situation of scientific research performance conducted by two basic Departments during the 12th Five-Year period in a general hospital.To understand the practical applicability of the scientific evaluation index system,and also propose comments and suggestions based on the comparison between assessment score and actual research situation in the departments.Methods Based on the database of scientific research management information platform,collect related research data from two basic departments of the hospital in latest five years.Using SPSS software to conduct statistical analysis on the total score of the first and two indicators,as well as the specific indicators of the three indicators,according to the performance evaluation index system and the weight built by the hospital.Results The index system can basically reflect the general information of scientific research input and output in the department,however,some indicators cannot truly reflect the index importance and the differences between the index weight is not significant,so it is necessary to make amendment based on demonstration.Conclusions Hospital should dynamically update the index system to make them match the developing strategy of the hospital,also important to combine them with research stimulation to guarantee more scientific management and better service to the research development of hospitals.
3.Clinical value of endoscopy and histopathology in differential diagnosis of intestinal tuberculosis and Crohn's disease

Objective Clinical values of endoscopy and histopathology in differential diagnosis of Crohn's disease(CD) and intestinal tuberculosis were evaluated by characterising the distinctive diagnostic features of CD and intestinal tuberculosis in surgically resected specimens.Methods To review retrospectively the surgical enterocolonic specimens and to compare the clinical,endoscopic and morphologic features of CD or intestinal tuberculosis.Selected histological parameters were evaluated retrospectively in both 30 cases of CD and intestinal tuberculosis.Results The sensitivity,specificity and accuracy of endoscopy in diagnosis of CD and intestinal tuberculosis were 80.0%,25.6% 49.3% and 86.7%,46.2% 63.8% respectively.The histological characteristic parameters of intestinal tuberculosis were confluent granulomas often with caseous necrosis and thinning submucosa;the features of CD were granulomas without caseous necrosis ,submucosal hyperplasia fissure like ulceration and prominent lymphoid aggregates.Some histopathology similarities of CD and intestinal tuberculosis were found,such as chronic non-specific inflammation of intestinal wall,ulceration,collections of epithelioid histocytes,microgranulomas and transmural inflammation.Conclusions Although distinctive diagnostic features of CD and intestinal tuberculosis were found in their clinical,endoscopical and pathological study.The final differential diagnosis depends largely upon pathology.
4.Intravitreal Conbercept for treatment of choroidal neovascularization secondary to pathologic myopia

Objective To evaluate the efficacy of intravitreal injections of conbercept in choroidal neovascularization (CNV) secondary to pathologic myopia (PM).Methods A retrospective case series of 37 eyes of 37 patients affected with CNV secondary to PM treated by intravitreal injections of Conbercept.All the patients were examined with best-corrected visual acuity (BCVA) of Early Treatment Diabetic Retinopathy Study (ETDRS) chart,central macular thickness (CMT) of optical coherence tomography (OCT) at baseline.The initial average letters of ETDRS BCVA was 49.86±8.87,CMT was (306.38±31.01) μm.All eyes were treated with intravitreal Conbercept 0.05 ml (10 mg/ml).Follow up visits were performed monthly after injection.The mean follow-up time was 16.8 months.Injections were repeated according to the situation of BCVA,CMT,subretinal fluid and CNV leakage in follow-up.All eyes received an average of 4.8 injections.BCVA,CMT and ophthalmoscope examination were assessed monthly.The relationship of BCVA improvement and CMT reduction with the data at baseline and number of treatments were analyzed by Spearman Rho correlation respectively.Results During the 1,3,6 and 12 months after treatment,the mean BCVA were all improved with statistically significant difference (t=17.629,P<0.01).At 12 months,15 eyes (45.9%) had improvement of 15 letters or more,32 eyes (86.5%) had improvement of 5 letters or more,and 1 eye (2.7%) had decreased more than 5 letters.During the 1,3,6 and 12 months after treatment,the mean CMT were all decreased with statistically significant difference (F=43.726,P<0.01).At 12 months,the retinal fluid of 34 eyes (91.9%) were absorbed completely;33 eyes (89.2%) had angiographic closure at that time.There was no significant relevance between BCVA improvement and sex,age and course of the disease (P>0.05),but a significant negative correlation was found between the BCVA improvement and BCVA at baseline,injection times (P<0.05).There was no significant relevance between CMT reduction and sex,age,course of the disease and injection times (P>0.05),but a significant positive correlation was found between CMT reduction and CMT at baseline (P<0.05).There were no systemic or ocular serious side effects during the follow up.Conclusions Intravitreal injections of Conbercept showed BCVA improvement and CMT reduction.It appeared to be effective and safe for choroidal neovascularization secondary to pathologic myopia.Intravitreal Conbercept for CNV secondary to PM showed BCVA improvement,CMT reduction and safety.
5.TRACE ELEMENTS AND EXTRACELLULAR MATRIX

Objective To investigate the effects of trace elements on the metabolism of extracellular matrix and explore the physiological and pathological mechanism of trauma. Methods Based on the experimental and clinical data, it was studied that the action of trace elements in the metabolism of extracellular matrix in trauma repairing. Results During wound healing, the trace elements were the components of many kinds of enzymes, carriers and proteins. They took part in the synthesis of hormones and vitamins as well as the transmission of information system. They activated many different kinds of enzymes and regulate the levels of free radicals. The trace elements had the complicated effects on the synthesis, decompose, deposition and reconstruction of collagen and other extracellular matrix. Conclusion The trace elements play an important role in regulating the metabolism of extracellular matrix.
6.Expressions of cyclooxygenase-2 and 5-1ipoxygenase in colonic mucosa of ulcerative colitis and the relationship between them

Objective To investigate the expressions of cyclooxygenase-2 (COX-2) and 5-lipoxygenase (5-LOX) in colonic mucosa of patients with ulcerative colitis (UC)and to analyze the relationship between them. Methods The specimens of colonic mucosa from 32 UC patients were graded according to endoscopic and histological grading standards, and specimens of colonic mucosa from 26 healthy controls were also collected. The expressions of COX-2 and 5-LOX mRNA and protein in colonic mucosa were determined using real-time fluorescent quantitative reverse transcription polymerase chain reaction(RT-PCR) and immunohistochemistry. The correlation between COX-2 and 5-LOX was analyzed. Results For UC patients,10 were class 1, 19 class 2 and 3 class 3 according to endoscopic grading, whereas 19 were class Ⅰ , 9 class Ⅱ and 4 class Ⅲ according to histological grading. The expressions of COX-2 and 5-LOX mRNA in active UC patients were 81. 25% and 53.13%, respectively, and were 11.54% and 19. 23% in healthy controls, respectively, with significant differences between the two groups (all P values<0.01). The positive expressions of COX2 and 5-LOX increased in accordance with increasing of endoscopic grading and histological grading.The levels of COX-2 and 5-LOX were 20. 08±1.17 and 37.83 ±1.48 in colonic mucosa tissues of UC patients, respectively, and 48.42 ± 1.69 and 11.28 ± 1.62 in healthy controls, respectively. There was significant difference between the two groups (all P values<0.05). A good positive correlation was found between COX-2 and 5-LOX. Conclusions The expressions of COX-2 is closely related to 5-LOX in UC patients. Both may play a pivotal role in inflammation of UC.
7.Establishment of a multidrug resistant human salivary gland adenoid cystic carcinoma cell line

Objective:To establish a multidrug resistant human salivary gland adenoid cystic carcinoma cell line.Methods:Human salivary gland adenoid cystic carcinoma cell line ACC-2 was treated by 24-hour-exposure to high dose of Bleomycin(BLM)(20 ?g/ml).Drug sensitivity was evaluated by MTT assay.Cell counting was employed to make the growth curve and to calculate the cell doubling time.Flow cytometry(FCM) was used to study the cell cycle distribution and apoptosis.The colony formation ability was also observed.Results:Multidrug resistant cell line of human salivary gland adenoid cystic carcinoma was established and named ACC-2/BLM.After 10 times repeated exposure to BLM,the resistance index(RI) to BLM,5-Fluorouracil(5-FU),Cisplatin(CDDP),Cyclophosphamide(CTX),Vincristine(VCR) were 7.299,1.03,2.15,1.114 and 5.96 respectively.Compared with ACC-2,the proliferation potential of ACC-2/BLM cells was decreased.The ACC-2 apoptosis cells were much more than ACC-2/BLM cells after 9-day-treatment by BLM at 60 ?g/ml.Conclusion:ACC-2/BLM cell line has multidrug resistant characteristics.

AIM:To construct hybrid nucleic acid vaccines which contain Plasmodium falciparum merozoite surface protein 1 (MSP1 ) block 1 7gene and gene fragment coding for several T cell epitopes. METHODS:MSP1 block 1 7gene of FUP strain was connected with a polyvalent gene fragment which contains several T cell epitopes from MSA1 ,MSA2 ,RESA,IL- 1 and Tetanus toxin,the connected gene fragment (hybrid gene,HG) was cloned into eukaryotic expression plasmid p CDNA3.1 (- ) ,VR1 0 1 2 for intracellular expression,VR1 0 1 2 /TPA for secretion,the recombinants were identified by PCR and restriction enzyme digestion. RE- SUL TS &CONCLUSION:The hybrid nucieic acid vaccines:VR1 0 1 2 /HG,VR1 0 1 2 /TPA/ HG and p CDNA3.1 /HG were successfully constructed.
9.Staphylococcus aureus Infection in Lower Respiratory Tract Patients in Clean Intensive Care Unit

OBJECTIVE To find the cause of lower respiratory tract infection as soon as possible and take measures to control the infection.METHODS The records of hospitalized patients in the intensive care unit(ICU) form Nov 28 to Dec 6 in 2006 were analyzed;and the disinfection efficiency,sterilization effect and the environment were monitored.RESULTS The incidence rate of lower respiratory tract infection was 54.55%.The qualified rate of disinfection and sterilization monitoring was 35.71%.The respiratory tubing,bed handle,bottle openner,computer mouse,the surface of the file cabinet and air vents were heavily infected.The Staphylococcus aureus was isolated from the quilt cover and the surface of the file cabinet.CONCLUSIONS We should put more emphasis on the management of the infective disease in the ICU,on hand washing and the cleanliness and maintenance of the devices.Disinfection and isolation guidelines should be strictly followed to reduce the incident rate of infection in the ICU.
10.SLOW DIGESTIBILITY OF CEREAL STARCH AND POSTPRANDIAL GLYCEMIC RESPONSE

Objective To analyze slowly the digestibility and postprandial glycemic response after consum of different cereal starch. Method Starch was quantified into different nutritional fractions using the in vitro Englyst test. Ten healthy subjects consumed 7 kinds of carbohydrate foods with 50 g normal maize starch, waxy maize starch, wheat starch, sticky rice starch, rice starch, potato starch or glucose. Blood samples from postprandial zero to 120 min after consumption of test materials were collected for measurement of glucose, glycemic index and extended glycemic index. Results Native cereal starch was ideal slowly digestible starch (SDS) and the proportion of SDS was about 50% based on Englyst test. The GI value of cereal starch was more than 90% and belonged to high glycemic index foods, but EGI was positive, regarding glucose powder as 100%. Conclusion Cereal starch with slow digestibility and similar glycemic response was a better carbohydrate material to provide a slow and prolonged release of blood glucose and maintenance of glucose homeostasis, which was potentially beneficial to health.
