1. Chemo-sensitivity enhancing effects of Shengmai Injection on various chemotherapeutic drugs

Objective: To investigate the chemo-sensitivity enhancing effect and its major active mechanism of Shengmai Injection (SMI). Methods: MTT assay was used to determine the chemo-sensitivity enhancing activity of SMI combined with various current- used chemotherapeutic drugs. Flow cytometry was conducted to determine the effect of Epirubicin combined with SMI on the cell cycle distribution and apoptotic cell death, and the mRNA expression level of MDR-1 gene was determined by RT-PCR. Results: SMI (30 μL/mL) combined with gemcitabine, cisplatin, paclitaxel, and Epirubicin exihibited the potent antiproliferative effects on human lung carcinoma A549, gastric carcinoma SGC-7901, breast carcinoma MCF-7, and hepatocellular carcinoma HepG-2 cell lines, respectively. SMI showed the most significant enhancing effects on HepG-2 cells in combination with Epirubicin, and the sensitivity of HepG-2 to Epirubicin was increased by 16 fold. Whereas SMI (30 μL/mL) alone did not affect the cell cycle distribution and apoptosis of HepG-2 cells, it enhanced the cellular responses when combined with Epirubincin together with a down-regulated mRNA level of MDR-1 gene. Conclusion: SMI combined with the different chemotherapeutic drugs could enhance the sensitivity of cancer cells significantly via down-regulating the mRNA expression level of MDR-1 and the expression of P-glucoprotein with the excretion of drugs.
2.Cloning and expressing of vacA gene related segment with vacuolating activity of Helicobacter pylori

Objective:To obtain the antigen of recombinant vacuolating segment of Helicobacter pylori vacuolating cytotoxing by gene-engineering.Methods:The objective gene was cloned into PQE-30,sequenced and expressed using PCR and recombinant techniques.After expression,the antigenicity of recombinant protein was analyzed by Western blotting.Results:DNA sequence analysis identified that the sequence was integrated.Compared with standard strain AF361700,1.5% of the cloned gene was mutated,one amino acid radicle was changed.SDS-PAGE revealed the molecular weight of objective recombinant protein was 27 000 as it was predicted;the level of expression product was over 30%.Western blotting results showed the objective protein could be recognized by anti-serum against HP.Conclusion:This recombinant protein has perfect antigenicity,so it may be used as a protective antigen in preventing HP infection.
3.Analysis of flavonoids in seeds of Oroxylum indicum by HPLC-ESI-MS

Objective To establish an HPLC-ESI-MS method for simultaneous analysis and determination of flavone in the seeds of Oroxylum indicum.Methods The separation was performed by Hypersil C_(18) column(250 mm?4.6 mm,5 ?m) with a mobile phase of formic acid aqueous solution(A) and acetonitrile(B) 0.5%.The flow rate was 1.0 mL/min and detection wavelength was at 195-400 nm.The samples were analyzed in positive mode.Results Six flavones in the seeds of O.indicum could be separated in one run.Baicalin,chrysin-7-glucuronide, oroxylin A,and baicalin-7-O-glucoside had been tentatively (identified) by ESI-MS and reference data.Conclusion Besides quantification of the components,this new method surpasses previously published ones in product quality control with accurate and rapid advantage,and provides the HPLC chromatographic fingerprints of biological active components in the seeds of O.indicum.
4.Progress of the prevention and treatment of growth retardation in children with nephrotic syndrome

Nephrotic syndrome(NS) is a common disease in children.Glucocorticoids(GC) is the firstline treatment for NS and can cause growth hormone(GH) secretion and synthesis disorder through various ways which reduces levels of GH.Consequently,the growth and development are affected.For another,kddney disease can intervene in the secretion and regulation of insulin-like growth factor (IGF) and insulin-like growth factor binding protein(IGFBP).GH-IGF axis's change plays a role in children's growth retardation.Other factors,such as bone metabolic disturbance and hypothyroidism aggravates the growth inhibition.This review focuses on the prevention and treatment of growth retardation in children with NS.
5.Treatment of coarctation of aorta

Coarctation of aorta is a discrete narrowness of the aorta and most commonly occurs in the isthmus.It is a common congenital cardiovascular malformation with morbidity of 5% ~ 8% among all congenital heart defects.Coarctation can result in the increase of upper limb blood pressure,the decrease of limb blood pressure,and a decline in cardiac function.Treatments include surgical repair and percutaneous interventions with different complications.Long-term complications and heart functions are important indices for evaluating prognosis and should be closely evaluated postoperatively.This review focuses on progresses of therapy methods,postoperative complications and cardiac function.
6.Preliminary study of the destructive characteristics of the liver induce by focused ultrasound surgical clamp

Objective:To observe the destructive characteristics of the bovine liver induced by focused ultrasound clamp technique.Methods:A focused ultrasound surgical clamp(FUSC) consisted of two transducers fixed on a forceps-like handle.The directions of focused ultrasound beams were opposite,with the same central axes.The thickness of bovine liver in vitro ranged from 4 to 10cm.Ultrasound energy was continuously delivered to ablate the liver with a frequency of 1.6MHz,and spatial peak pulse average acoustic intensity(ISPPA) of 12 000W/cm2.The characteristics of complete coagulation necrosis and exposure time were measured after FUSC exposure,and pathological changes in treated bovine liver tissue were observed.Results:FUSC induced complete coagulation necrosis of predetermined bovine liver targets.The shape of coagulation necrosis was corresponded with focused ultrasound beam,with 2.72~3.20cm in length and 1.44~1.80cm in width.However,under the same exposure energy,exposure time for complete necrosis increased,as the thickness of the liver increased.Conclusion:FUSC can induce complete coagulation necrosis of predetermined bovine liver targets.This study would provide a novel surgical instrument for hepatectomy.
7.Study on the postmortem stability of heart fatty-acid-binding protein for the diagnosis of early myocardial infraction

Objective:To investigate the stability of heart fatty-acid-binding protein(H-FABP)for the postmortem diagnosis of early myocardial infraction.Methods:Rabbit model of myocardial ischemia was induced by ligating the anterior branch of the left coronary artery.The areas of H-FABP depletion in the ischemic myocardial cells were studied by immunohistochemistry,image analysis technique and statistical system.Results:The specimens of normal myocardium kept at 4℃ for 1 to 2 days showed homogenous brown reactions for H-FABP.The depletion of H-FABP became evident when these specimens were kept at 4℃ for 3 days after death,and the depletion area increased with the lapse of postmortem interval.The depletion area of H-FABP in ischemic myocardial tissue also increased with the lapse of postmortem interval.The results of image analysis indicated that the depletion area of H-FABP in ischemic myocardial tissue within 4 d were significantly different from that in the normal control group(P0.05).Conclusion:The H-FABP shows medium postmortem stability,and small influenced by autolysis,which is suitable for detection in corpses at 4℃ 4 days after death.H-FABP as a marker for postmortem diagnosis of AMI has some practical value in forensic medicine.

10.Progress of microRNAs and B-cell receptor signaling pathway in chronic lymphocytic leukemia

Many studies have confirmed that dysregulation of microRNAs (miRNAs) expression can cause disruption of the hematopoietic system and contribute to leukemogenesis by regulating the expression of some oncogenes and anti-oncogenes. Chronic lymphocytic leukemia (CLL) is a malignant proliferative disease of B lymphocytes with significant heterogeneity in the incidence of individuals, disease progression, treatment response and clinical prognosis. The increasing studies have shown that the mutation or abnormal expression of miRNAs is closely related to CLL occurrence, progression, prognosis and curative efficacy. Certain miRNAs are involved in the intricate interplay with B-cell receptor (BCR) signaling pathway. The review discusses the latest progress of miRNAs expression in BCR signaling pathway and CLL onset and progression.
