1.AN ELECTRON MICROSCOPIC HISTOCHEMICAL OBSERVATION ON LYSOSOMAL ACID PHOSPHATASE AND ARYLSULPHATASE IN THE HEPATOCYTE OF GUINEA PIG, PIGEON AND TOAD

By using the cytochemical methods, the activity of lysosomal acid phosphatase (ACP) and arylsulphatase (ArSase) in liver cells of the three kinds of vertebrates, guinea pig, pigeon and toad, was observed. The results show that the distribution of the two enzymes has some difference in the hepatocytes of all three kinds of animals. There are four forms of ArSase localization in the dense bodies of hepatocytes. Furthermore the three kinds of animals have different ArSase localization characteristics. The biological significance of these phenomena was discussed.
2.Estimation of parameters in stratified cluster sampling on randomized response technique for sensitive question survey

Objective:To explore a scientific,complicated sampling method and corresponding formulas for sensitive question survey.Methods:Cochran W.G.'s classic sampling theories,two randomized response technique(RRT) models,total probability formulas,and properties of mean and variance were used in this study.Results:Formulas for the estimation of the population proportion and its variance for dichotomous sensitive questions on Greenberg model in cluster sampling and stratified cluster sampling were deduced.Formulas for the estimation of the population mean and its variance for quantitative sensitive question on the additive constant model in cluster sampling and stratified cluster sampling were deduced.Our survey methods and formulas were successfully applied in surveying the pre-marriage sex and cheating in examination in Soochow University.Conclusion:Our survey methods and formulas on two RRT models for sensitive question survey in cluster sampling and stratified cluster sampling have high reliablility.

This paper reviewed that high-intensity interval training (HIT) is effective on cardiorespiratory fitness and walking ability, but limited in quality of life in stroke paitents. More researches are needed to explore the differences from HIT to low and moderate intensi-ty continuous training .
5.The chemical constituents of Ligularia pleurocaulis

Aim To study the chemical constituents of the underground part of Ligularia pleurocaulis (Franch.) Hand-Mazz.. Methods The dried roots and rhizomes of L.pleurocaulis were extracted with methanol. Isolation and purification were performed by silica gel column chromatography and recrystallization etc. Structures of the pure compounds were established on basis of spectral analysis. Results Twelve compounds were obtained from L.pleurocaulis, they were 6-angeloyloxy-furanoligu-larenone (1), 2-oxo-3-hydroxy-eremophila-1(10),3,7(11),8-tetraen-8,12-olide (2), tiglic acid (3), oleanolic acid (4), lupeol (5), β-sitosterol (6), daucosterol (7), caffeic acid (8), emodin (9), 7-methoxy-coumarin (10), ferulic acid (11) and 4-hydroxy-2,5-dimethoxy-benzaldehyde (12). Conclusion Compound 1 is a new eremophilane and compound 2 is a new natural compound. All above compounds were obtained for the first time from L.pleurocaulis.
6.Effect of parecoxib on hippocampal inflammation following partial hepatectomy in rats

Objective To investigate the effect of parecoxib on hippocompal inflammation following partial hepatectomy in aged rats.The effects of selective COX-2 inhibitor (parecoxib) on hippocampal inflammation were also evaluated.Methods Seventy male rats,aged 20 months,weighing 500-600 g,were randomly divided into three groups: control (n=10),surgery (n=30),and parecoxib (n=30).Control animals received sterile saline to control for the effects of injection stress.Rats in the surgery group received partial hepatectomy under isoflurane anesthesia and sterile saline injection.Rats in the parecoxib group received surgery and anesthesia similar to surgery group rats,and parecoxib treatment.On postanesthetic day 1,3 and 7,animals were euthanized to assess the hippocampal COX-2 expression,PGE2 production and caspase-3 expression.Results Surgery significantly increased the expression of COX-2 mRNA and protein expression,PGE2 production and caspase-3 protein expression on day 1 postoperatively (P<0.01),until day 3 (P<0.05) compared to control group.Parecoxib treatment group significantly suppressed COX-2 mRNA and protein expression,PGE2 production and caspase-3 protein expression on postoperative day 1 and day 3 (P<0.05 or P<0.01),in comparison with the surgery group.There was no significant difference between control group and other groups of the expression of COX-2 mRNA and protein expression,PGE2 production and caspase-3 protein expression on day 7.Conclusion Partial hepatectomy induces a short-term hippocampal inflammatory response in the rats.Parecoxib suppressed the hippocampal inflammation via the down-regulation of COX-2 mRNA and protein,PGE2 and caspase-3 protein in rats following partial hepatectomy.
7.The Effects of Rosiglitazone on Cardiomyocyte Apoptosis and Expression of Bax and Bcl-2 in Type 2 Diabetic Rats

Objective To investigate the effects of rosiglitazone on the cardiomyocyte apoptosis and expression of Bax and Bcl-2 in type 2 diabetic rats. Methods 8 weeks old SD rats were feeded with high calorie for one month, and then were intraperitoneally injected with a low dose of STZ (30mg/kg). The diabetic and control rats were killed at the 12th and 24th weeks after establishing model, respectively. Cardiomyocyte apoptosis was evaluated by TUNEL and flow cytometry. Guinea pig, pigeon and toad are the vertebrates on different evolution level. The acid phosphatase content in the hepatocytes of centrilobular area and periportal area of hepatic lobules were quantitatively measured by microspectrophotometer and image analyser. The results showed that the content of acid phosphatase of hepatocytes was highest in guinea pig, lowest in pigeon, and toad in between the both. The average density of acid phosphatase distribution in the hepatocytes increases according to the evolution order. The significance of this phenomenon was discussed.

10.Abnormal expression of connexin 36 plays a role in the pathogenesis of levodopa induced dyskinesia in rat model of Parkinson' s disease

Objective To explore whether gap junction disturbances are involved in the pathogenesis of levodopa-induced dyskinesia ( LID ). Methods The hemi-parkinsonian ( PD ) rat was treated intraperitoneally with L-dopa methylester (20 mg/kg) and benserazid (10 mg/kg) for 21 days and abnormal involuntary movement was evaluated to establish LID rat model. The experimental animals were divided into three groups: LID group, PD group and normal control group, respectively. The behavior responses of intraperitoneal injection of different doses of carbenoxolon and intracerebroventricular injection of quinine were observed to estimate the effects of gap junctional blockade on the abnormal involuntary movement ( AIM ) in the rat model of LID. Double immunofluorescence labeling was used to analyze the expression of connexin 36 ( Cx36 ) in enkephalin positive medium spiny neurons and parvalbumin ( PV ) positive interneurons in the striatum. Western blottings was used to observe the expression of Cx36 in the striatum and moter cortex. Results Behavioral characteristics indicated that high dose of carbenoxolone ( >60 mg/kg) intraperitoneal injection and intracerebroventricular injection of quinine ( 0.5, 1.0, 2.0 μmol/L, > 2.5 μmol/L ) could decrease the AIM score of LID rats. Western blotting indicated that expression of Cx36 in lesioned striatum and motor cortex of LID rat model was 219.56% ±18.12% and 226.03% ±16.33%, respectively, which induced a significant upregulation in comparison with the normal control group (104.05% ±3.82%, t=15.389, P<0.01;105.27% ±2.82%,t=8.074, P<0.01) and untreated PD group (119.31% ±8.92%, t=13.356, P<0.01; 138.20% ±17.88%, t=5.872, P<0.01). Double immunofluorescence labeling staining revealed that Cx36 expression was increased in Enk-positive striatum neurons in LID model ( 57.59% ±5.36%) compared with that in normal control group (32.67% ±4.22%) and PD group (37.24% ±0.86%, F=78.060, P<0.01). The expression of Cx36 in PV-positive interneurons was also elevated in LID group (68.49% ±11.60%) in comparison with normal control group ( 40.43% ± 2.30%) and PD group ( 31.92% ± 5.68%, F = 39.567, P < 0.01 ).Conclusions The Cx36 expression is generally increased in lesioned striatum and motor cortex of LID rat model. In the striatum, the up-regulation of Cx36 is specifically observed in Enk-positive striatum neurons and in PV-positive interneurons. The dyskinesia behavior of LID rats can be significantly reduced by treatment with gap junction blockade. All these results suggest that gap junction dysfunction may play an important role in the pathogenesis of LID.
