Osteocalcin is a bone matrix noncollagen protein which is one of the most abundant secretory products of osteoblasts. Resent studies have demonstrated that osteocalcin is not only a specific biochemical marker of bone turnover, but also involved in the regulation of energy metabolism. Further studies in the pathophysiological functions of osteocalcin might contribute to the treatment of obesity, diabetes, and metabolic syndrome.

5.The Treatment of Tennis Elbow by Radiofrequency under Arthroscopy: A Preliminary Report

Objective To discuss the value and the effect of radiofrequency treatment of tennis elbow under the arthroscopy. Methods From March 2003 to Jan 2004,eight cases of tennis elbow were treated by TOPAZ with local anesthesia under arthroscopy. Results The effects and functions were satisfactory after radiofrequency treatment. The symptoms of pain were completely disappeared in 7 and obviously relieve in 1. No blood vessel and nerve injury or infection occurred. Conclusion The methods of radiofrequency therapy for tennis elbow under arthroscopy has the advantage of minimum invasion, less pain and earlier rehabilitation.
6.Study on the genes for TEM and aminoglycoside-modifying enzymes

The genes for TEM and aminoglycoside-modifying enzymes (AMEs) in Acinetobacter baumannii were studied to provide molecular biological support for monitoring their drug resistance.MethodsTwenty strains of Acinetobacter baumannii were clinically isolated from the period of August, 2002 to Feburary, 2004. The drug resistance of these strains to thirteen kinds of antimicrobial agents was tested by microdilute test. The TEM and AMEs genotypes of these strains were analyzed by polymerase chain reaction and verificated by DNA sequencing .Results All twenty strains were sensitive to Imipenam and Meropenem. The resistant rates of the twenty strains to Piperacillin/ Tazobactam, cefepime and ceftazidime were 45.0%, 65.0% and 70.0%, respectively. The resistant rate to aminoglycosides antibiotics was 75.0%. The resistant rates to other antimicrobial agents were greater than 65.0%. The positive rate of genotype TEM, aac(3)-Ⅰ, aac(3)-Ⅱ aac(6”)-1 and ant(3”)-Ⅰ were 45.0%, 55.0%, 15.0%, 35.0%, and 60.0%, respectively. The genotypes of SHV, AmpC DHA and AmpC MIR were all tested negative.Conlusions The multiple-drug-resistance of Acinetobacter baumannii isolated from Tongde Hospital was a serious issue.There were high percentages of TEM and AMEs genotypes. Because of the imperfection frame of the primer,some familiar genes just like OXA,VER and PER were not experimented. We shall further discuss the drug resistance of the third/fourth generations cephalosporin.
7.Radioprotective potential of genistein on murine bone marrow stromal cells

Objective To investigate the radioprotective effect of genistein (GEN) on murine bone marrow stromal cells (BMSCs). Methods Adult male mice were administered orally with GEN at 24 h prior to 6.0 Gy gamma irradiation. The number of CFU-F, the adherent capability of bone marrow cells (BMCs) to BMSCs adherence layer, and the expressions of cell adhesion molecules including vascular cell adhesion molecule-1 (VCAM-1), fibronectin (Fn), and laminin (Ln) on BMSCs cultured in vitro were detected, respectively. Results The number of CFU-F, the adherent capability, the expressions of VCAM-1, Fn, and Ln on BMSCs cultured in vitro in GEN-pretreated mice were higher and recovered more rapidly than those in mice without GEN-pretreatment. Light microscopy revealed that there was no significant difference in morphology of BMSCs in both groups after irradiation injury. Conclusion These observations suggest that radioprotective properties associated with GEN are largely a consequence of the reduction of damage, the induction of cell proliferation, and the promotion of adherent capability recovery in recipient mice. These changes may ultimately ameliorate bone marrow microenvironment and therefore accelerate the rebuilding of hematopoietic system after irradiation injury.
8.The effect of bFGF on apoptosis of cultured human retinal pigment epithelial cells exposed to visible light

Objective To observe the effect of exogenous basic fibroblast growth factor (bFGF) on apoptosis of cultured human retinal pigment epithelial (RPE) cells exposed to visible light,and determine the role of bFGF, fibroblast growth factor receptor 1 (FGFR1),bcl 2 and caspase 3. Methods (2000? 500) lx cold white light was used. Exogenous bFGF was utilized during culture. Annexin annexin V fluorescein isothiocyanate/propidium iodium (V FITC/PI) labeling,flow cytometry, Immunocytochemical staining, enzyme associated absorb examing and reverse transcriptional polymerase chain reaction (RT PCR) were used to determine the apoptosis, the expression levels of bFGF, FGFR1, bcl 2, as well as the activity of caspase 3. Results No protective effect of bFGF was observed under the concentration 5 ng/ml. 9.CHANGES OF CYTOSKELETAL GENES EXPRESSION IN CORTICOSPINAL NEURONS AFTER SPINAL CORD SEMITRANSECTION INJURY IN RAT

Objective To investigate the pattern and the role of cytoskeletal genes expression during nerve regeneration. Methods We examined the changes of expression of ? tubulin(? Tub)and three neurofilament(NF)subunits mRNA in rat corticospinal neurons of sensorimotor cortex on the 1st,3rd,5th,7th,10th,14th,21st,28th and 56th days after spinal cord semitransection injury by using in situ hybridization. Results Levels of ? Tub,NF L,NF M and NF H mRNA were dramatically downregulated within large sized pyramidal neurons in layer V of the sensorimotor cortex of side lateral after spinal cord injury.The level of ? Tub mRNA was decreased as early as 1 day postinjury,whereas the levels of NF subunits mRNA were not decreased until later time(on the 3rd days postinjury),and both of them did not return to control levels on the 56th days following the lesion. Conclusion ? Tub gene expression if inhibited in CNS neurons postinjury. The axotomy signeal and the continuance downregulation of NF mRNA expressioon following spinal cord injury may contribute to the ineffective regeneration response of CNS neurons.
10.ALTERED EXPRESSION OF NEUROFILAMENT GENES IN RAT SENSORY NEURONS OF THE DORSAL ROOT GANGLION AFTER SCIATIC NERVE CRUSH

Objective In order to explore the mechanisms that regulate the expression of genes coding for cytoskeletal proteins during nerve regeneration. Methods In the present study, in situ hybridization was used to examine the changes of light(NF L),medium(NF M)and heavy (NF H)neurofilament protein subunits mRNA in L 4-6 dosal root ganglion (DRG) sensory neurons during nerve regeneration following a unilateral crush of the sciatic nerve. Results The hybridization signals of each neurofilament subunit mRNA were dramatic decrees in DRG sensory neurons post axotomy by light microscope.The signals of NF\|L and NF\|M mRNA were located in cytoplasm of neurons,whereas NF H mRNA was found in both nucleus and cytoplasm of neurons. Conclusion There are the different mechanisms of regulation of neurofilament subunit genes expression,the reduced neurofilament gene expression may represent a general response to axonal injury and plays an important role in effective nerve regeneration.
