1.Roles of cyclinD1/CDK4 during lovastatin suppressing proliferation of breast cancer xenograft tumor

Objective To establish the animal model bearing BRCA1-overexpressing breast tumor in athymic mouse so as to investigate the effects of lovastatin (LOV) on the growth of the xenograft tumor and understand its mechanism. Methods After amplification and identification, pcDNA3-beta-HA-hsBRCA1 plasmid was transfected into MCF-7 cells by liposome transfection to deliver a high level of BRCA1 gene expression examined with RT-PCR and Western blotting (named MCF-7 BRCA1 ). Twenty athymic mouse models of BRCA1-overexpressing xenograft tumor were established by respectively inoculating 2?10 6 MCF-7 cells and MCF-7 BRCA1 cells to right anterior part of the back. Four weeks later, lovastatin at the dose of 50 mg/kg was injected around the tumor for 10 d. Then the tumor volume was measured, the histopathological changes of xenograft tumor were observed with electron microscope by HE staining, the expressions of cyclinD1, CDK4 mRNA and protein were detected by RT-PCR and Western blotting. Results The athymic mouse model of xenograft breast tumor was successfully established after MCF-7 cells were transfected with pcDNA3-beta-HA-hsBRCA1 plasmids. The incidence rate of xenografted breast tumor in athymic mouse was 100%. Routine HE staining of paraffin-embedded section proved that the tumor was a mammary carcinoma in nature. Treated with lovastatin for 10 d, the tumor volume of mouse implanted with MCF-7 cells was large (0.48 cm 3 ) and there was a slight change in expressions of cyclinD1, CDK4 mRNA and protein, while the tumor volume of mouse inoculating MCF-7 BRCA1 cells was small (0.21 cm 3 ) and the expressions of cyclinD1, CDK4 mRNA and protein obviously descended. The two groups showed diminution in cancer cells and increase of necrosis, more significant in those mice inoculating MCF-7 BRCA1 cells. Conclusion Lovastatin could resist the proliferation of BRCA1-overexpressing breast tumor, which may relate to the decrease of the cyclinD1, CDK4 mRNA and protein expressions.
2.EFFECTS OF EICOSAPENTAENOIC ACID IN COMBINATION WITH RETINOIC ACID ON N-RAS EXPRESSION IN HL-60 CELLS

Objective:To study the effects of eicosapentaenoic acid (EPA) and retinoic acid (RA) on proliferation and differentiation of HL 60 cells and the corresponding mechanisms. Methods:Flow cytometry was used to assay cell proliferation function, NBT reduction experiments to evaluate cell differentiation, and Western blot to analyse pRB expression. Results:The proliferation index (PI) in all experimental groups were all lower than that of the control , and EPA in combination with RA was the lowest one. NBT reduction experiments showed that the OD value of cells treated by EPA and RA was about 5.9 times of the control, but correspondingly by RA was 2.6 times. Moreover, p21 N ras expression of HL 60 cells were all decreased by treatment with EPA or/and RA. Conclusion:The synergistic effects of EPA and RA on proliferation and differentiation of HL 60 cells may be partly due to the change of p21 N ras expression.l proliferation function,NBT reduc
3.Transurethral Retrograde Dissection of the Prostate for Benign Prostatic Hyperplasia

60 g in 15 cases).The operation time ranged from 35 to 85 minutes(mean,45),and the blood loss was 50-100 ml(mean,60 ml).No case needed blood transfusion.No rupture of the prostatic capsule,venous sinuses,or perforation of the bladder-prostate junction occurred in this series.The patients were followed up for 4 to 6 months,during which the Qmax increased to 16.4-23.2 ml/s(mean,19.8 ml/s),RU decreased to 0-15 ml(mean,5 ml),and IPSS score improved to 0-6.8(mean,4.2).No patient developed urinary incontinence.Conclusions Transurethral retrograde dissection of the prostate is effective for the treatment of BPH with a low morbidity rate of urinary incontinence.The procedure is simple and results in low blood loss.
4.Expression and Significance of PKR,p-PKR and p-EIF2α in Human Cervical Lesions

Objective:To identify the relationship between expression of protein kinase R(PKR), phosphating PKR, EIF2α(p-PKR, p-EIF2α) in PKR→EIF2α signal transduction passage and the grades of cervical lesions, the role in generation and progression of cervical tumor and their effects to prognosis of cervical cancer patients. Methods:The expressions of PKR, p-PKR and p-EIF2α in human cervical cancer tissue of 63 cases, cervical intraepithelial neoplasia(CINⅠ-Ⅲ) of 114 cases and normal cervical epithelium of 15 cases were detected by immunohistochemical technique. Results:With the increase in grades of cervical lesions, the positive-expression rate of PKR increased and significantly correlated with the grades of cervical lesions(P < 0.05). With the increase in grades of cervical lesions, the positive-expression rates of p-PKR and p-EIF2α increased firstly, and then decreased. In cervical cancer group, the positive-expression rate of PKR was much higher than that of p-PKR(P < 0.01). The development and progression was quicker in later clinical stages of cervical cancer than that of earlier clinical stages of cervical cancer (P < 0.01). The development and progression of cervical cancer was quicker in patients with negative-expression of p-PKR and p-EIF2α than that in patients with positive-expression of p-PKR and p-EIF2α(P < 0.05). Conclusion:The positive-expression rate of PKR was correlated with the grades of cervical lesions. There are some factors which can impede PKR and EIF2α to be phosphorylated or make p-PKR and p-EIF2α dephosphorylate in high level cervical lesions, which promotes the development and progression of cervical lesions, worsens the prognosis of cervical cancer.
5.Primary study on the signal transduction concerned human papillomavirus 18 E6 oncoprotein

Objective To explore the relationship of signal transduction among human papillomavirus 18 E6 oncoprotein (HPV18E6), signal transducers and activators of transcription 1 (STAT1), protein kinase R( PKR )/α subunit of eukaryotic initiation factor 2 ( eIF2α ), nuclear factor-kappa Bp65 ( NF-κBp65 ), mitogen-activated protein kinase( MAPK)/c-Jun N-terminal kinase(JNK) ,and possible molecular mechanism. Methods Construct two lentiviral vectors which contain shRNA interfering sequence aiming at the targets of HPV18E6 oncogene and NC sequence( HPV18E6-RNAi-LV, NC-GFP-LV), based on the transduction with HPV18E6-RNAi-LV and NC-GFP-LV into HeLa cell to interfere the expression of HPV18E6 oncogene and NC sequence,the expressions of mRNA and protein( including phosphating patem)of HPV18E6, STATI, PKR, eIF2α, NF-κBp65, MAPK, JNK are measured with RT-PCR and Western blot, the difference of proliferation and sensitivity to carboplatin of HeLa cell are determined with Transwell cell methods and MTT among every groups. Results The expression of HPV18E6 oncogene can affect the expression level of mRNA and protein of NF-κBp65 and PKR genes, also affect phosphating levels of phosphating protein p-STAT1, p-PKR and p-eIF2α;the restraining rates of proliferation and sensitivity to carboplatin of HeLa cell are higher in HPV18E6-RNAi-LV group than the other groups( P<0. 05 or P<0.01 ). Conclusion HPV18E6 oncoprotein not only reduces the expression of PKR but dephosphorylates p-STAT1, pPKR and p-eIF2α to restrain activation of PKR/eIF2α signal transduction passage, maintain the proliferation and invading ability of HeLa cell and restrain apoptosis. The signal transduction among HPV18E6, MAPK/JNK are not clear.
6.Effect of two kinds of hemofiltration on vascular endothelial system

Objective To evaluate the effect of single CVVH and HDF on endothelial system. Methods Group CVVH:20 patients with multiple organ dysfunction syndrome ( MODS) underwent CVVH for 12h.Vascular access was through two-lumen catheter in femoral vein;Group HDF:Twenty patients with end stage renal disease (ESRD) on maintenance hemodialysis underwent HDF for 4h.Vascular access was through internal arteriovenous fistula.Group control:Healthy people were examined at our hospital.The se-rum levels of sVCAM-1, sICAM-1, vWF and MMP-9 were measured by enzyme-linked immunosorbent assay ( ELISA) after centrifugation in treatment group before and after treatment.Four cytokines were also meas-ured in fasting healthy subjects in the morning.Results The levels of serum sVCAM-1, sICAM-1, vWF, MMP-9 were significantly higher in Group CVVH and HDF than control group before and after treatment( P<0.05).Group CVVH:The serum levels of sICAM-1 and sVCAM-1 were significantly lower in post-treat-ment than pre-treatment, But the serum levels of vWF and MMP-9 showed no obvious changes after CVVH;Group HDF:the levels of four cytokines showed no obvious changes after single HDF.Conclusions Endo-thelium becomes impaired in MODS and ESRD patients, Endothelium may be protected by CVVH through the partial removal of cytokines harmful to endothelium in MODS patients.Single HDF therapy does not pro-tect endothelial function obviously in MHD patients.
7.The Application of Paries Anterior Vagina Muscularis Mucosal Flap in the Repairat/on of Female Complicated Urethrovaginal Fistula

Objective To explore the application of the paries anterior vagina U shape muscularis mucosal flap for the repairing of female complicated urethrovaginal fistula. Methods Fifteen complicated urethrovaginal fistulas patients resulted from severe pelvic frac-ture and with history of failing repairing urethrovaginal fistulas were analyzed retrospectively. The mean diameter of maximum urethrovagi-nal fistula was approximately 2.3 cm. The parias anterior vagina U shape mucosal flaps were used to repair the urethrovaginal fistulas, and the paries anterior vagina muscularis mucosae were used to cover the fistulas surface again. Results Fifteen patients were followed up for 6 to 80 months, and they gained normal voiding, with no urine leakage, urinary incontinence and urethra] stricture occurring. Conclusion The paries anterior vagina U shape mucosal flap is an effective technique for repairing complicated urethrovaginal fistula.
8.Investigation of risk factors in the liver cancer of Wuwei city

Objective To investigate the widespread of the liver cancer risk factors in Wuwei city,where low-incidence for liver cancer and offer the cause clue to first-level prevention. Methods Investigate the risk factor and family history of the Wuwei city resident who wlth liver cancer by using case-control study.Estimate the liver cancer heredity mode with the method of Penrose and simple segregation analysis,calculate the degree of heritability with the metllod of Falconer,s regression.Detect the content of the resulting and promoting cancer fungi,volatility N-second nitroso compound in the meal,nitrate and nitrite in the drinking water and the vitamin C in the health adults'serum by means of culturing appraisement,experiment examination etc.Results The risk factom of liver cancer in the Wuwei city were history of viral 'hepatitis type B,kinsfolk tumor,the eating salting or mildewed foods,cirrhosis,drinking surface water,well water,or wine.By using segregation analysis and estimation of heredity mode show that the morbidity of liver cancer discrepancy to the monogenic inheritance mode,maybe the polygenic inheritance mode in Wuwei city.The h~2 of index case first degree relative was 58.74%.Picking out 8 kinds of nitrosamine,14 kinds of resulting and promoting cancer fungi from resident meal.The content of nitrate and nitrite was (38.97±3.14) mg/L,(0.086±0.043) mg/L respectively in shallow well water,the vitamin C in the health adults'serum in summer was(5.74±2.79)mg/L.Conclusion It can be seen that the history of viral hepatitis type B,kinsfolk tumor,eating salting or mildewed foods,cirrhosis,addicted to drink;higher content of nitrate and nitrite in the drinking water and strong carcinogens such as volatility N-nitroso compound in the meal,and also lackingprotection factors such as vitamin C are the risk factors of the Wuwei city resident who with liver cancer,Theinherit susceptivity is the internal environment of resulting cancer change.
9.Study and application of surface antigen in tachyzoites of Toxoplasma gondii

Toxoplasma gondii is an intracellular protozoan parasite that infects all warm?blooded animals. The surface anti?gens of T. gondii with the potential for application as antigens of diagnosis and vaccines have been studied extensively in recent years especially for P43 P35 P30 P23 and P22. The studies on the surface antigen in tachyzoites of T. gondii are reviewed in this paper.
10.Influence of Semen Processing on Proportion of Aneuploid Sperm

Objective: To analyze the influence of semen processing methods on the proportion of the aneuploid sperm by detecting the sperm's chromosome X, Y, 18 using fluorescence in situ hybridization. Methods: Ten patients with mild ohgoasthenosperia, who were received ICSI treatment, were included in this study. Five semen samples of the patients were randomly selected to detect using Swim-up method (A group) and 5 using sperm-grad double-density centrifugation method (B group).Another 5 patients with mild oligo-asthenosperia were as control (C group). The CEPX / Y and CEP18 probe was used to detect the sperm of these 15 patients by fluorescence in situ hybridization. The proportion of aneuploid sperm was compared in three groups. Results: The sex chromosome aneuploid rates were (4.21±2.49)%, (3.24~1.49) % and (2.62±0.89) % in control, A and B groups. The rates of aneuploid chromosome 18 were (3.00±1.22)%, (2.00~1.22)% and (2.00±1.22)% in control, A and B groups. There were no significant differences in three groups (P>0.05). Conclusion: The results showed that the methods of Swim-up and Sperm-Grad double-density gradient centrifugation could select sperms in motility potential and teratospermia,but not in normal chromosome sperms.
