1.MicroRNA biology technology and the progress research of miR-125 in disease

MicroRNA,which is found in recent years,is a class of small noncoding RNAs of about 18-24 nucleotides in length,that can regulate gene expression.It induces mRNA degradation or inhibit translation of tagert genes mainly by complete or incomplete base pairing to the 3' untranslated region of tagert messenger RNAs.The application of microRNA biology technology provides new methods to the research of multiple diseases,and can further elucidate its occurrence,development mechanism.In this paper,microRNA biology-related researches are introduced and miR-125 in a variety of diseases is reviewed.
2.The comparative study on clinical effects of PFNA and DHS on treating intertrochanteric femur fractures in the elderly

Objective To comparetheproximalfemoralnailantirotation(PFNA)anddynamichipscrew(DHS)treatmentonin-tertrochanteric femur fractures in the elderly .Methods Retrospective analysis was conducted on 60 patients with intertrochanteric femur fracture collected from December 2009 to December 2012 ,which underwent these two methods ,including 32 cases of PFNA group ,28 cases of DHS group .The 5 indicators including operation time ,intraoperative blood loss ,complications ,fracture healing time and hip function recovery were analyzed .Results Sixty cases were followed up for 10 to 25 months ,average 17 months ,com-pared with DHS group ,PFNA group was with short operation time ,less bleeding ,less complications ,short time of fracture healing , according to standard of Harris score ,the hip function recovered well .Conclusion PFNA treatment on intertrochanteric femur frac-tures is with little trauma ,reliable fixation ,fewer complications ,with which the hip function recovered well ,and it is the preferred method of operation in treatment of senile intertrochanteric femur fracture .
3.Comparative study of magnetic resonance elastography and T2WI in assessing stiffness of tissue

Objective To preliminarily compare accuracy of magnetic resonance elastography (MRE) and T2WI in assessing stiffness of tissue.Methods Agar gel phantoms of 23 different concentrations ranging from 0.8%-3.0% were produced.All the phantoms were examined with MRE and T2 map scanning respectively.After the raw images were processed by using local wavelength estimation (LEF),shear modulus of scan slice was measured,and T2 value of the same slice was measured with GE ADW4.3 workstation.Pearson correlation analysis was used to test the correlation between shear modulus and agar concentration,as well as T2 value and concentration.Rank sum test was used to compare the correlation coefficients of them.Results All the shear modulus and T2 values of the phantoms were successfully obtained,which were (49.1 ± 23.5) kPa and (57.8 ± 21.8) ms respectively.Shear modulus was positively correlated with concentration of agar gel(r1 =0.985,r1 2 =0.970,P < 0.01),while T2 value was negatively correlated with concentration of agar gel (r2 =-0.901,r22 =0.812,P < 0.01).The differences of these two correlation coefficients were statistically significant (Z =5.459,P < 0.01).Conclusion MRE is more accurate to reflect stiffness of tissues relative to T2WI,and shows great clinical prospect.
4.Design and clinical application of three-dimensional puncture line guider in percutaneous vertebroplasty

According to the anatomy characteristics of vertebral pedicle and on the basis of puncture site on the skin to the level distance of vertebral process, a three-dimension guider was designed. Eighty-three cases were received percutaneous vertebroplasty from December 2006 to December 2008, including 43 cases with three-dimension guider. Thirty-six cases of above 43 cases puncture site were guided by fluoroscopy, and then performed percutaneous vertebroplasty by three-dimension guider without fluoroscopy monitoring under guidance of preoperative CT measures. The rest 7 cases of 43 were performed percutaneous vertebroplasty by the monitored by fluoroscopy. All cases were successful, and the needle tip could reach the ideal position. Forty cases were received primary two-dimesion guider to perform the percutaneous vertebroplasty, which were monitored by fluoroscopy. The mean puncture time was 1.9 minutes in two-dimension guider while 1.5 minutes in three-dimension guider. Results demonstrated that three-dimension guider has the virtue of logical design, suit for the body anatomy and easy to perform, which can reduce the expose time of x-rays and more safety and efficiency to operators.
5.Nursing of cancer patients with anal fistula treated with blood stem cells transplantation

Objective To summarize the experience of nursing cancer patients with anal fistula treated with blood stem cells transplantation (BSCT). Method 8 critical cancer patients with anal fistula treated with BSCT from August 2013 to November 2014 were intervened by nursing measures such as prevention of constipation and diarrhea , protecting the anus from infection and health education . Results Two patients of them contracted with mild crissum infection , 1 moderate , 0 severe . With the pertinent nursing intervention , no serious complications occurred . The hospital stay was 20~30 d , with an average of ( 24 . 75 ± 2 . 30 ) . Conclusion The nursing measures including preventing constipation and diarrhea, keeping the anus from infection and health education can be effective in avoiding anal fistula infection and its related complications and reducing patients′pain , which are worth clinical promotion .
6.Effects of Gestation Exposure to Dibutyltin Dilaurate (DBTD) on Enzymes Activity in Testis and Spermatogenesis Function of Offspring Rats

Objective To explore the effects of gestation exposure to dibutyltin dilaurate (DBTD) on the activity of enzymes in the testis and the spermatogenesis function in the mature male offspring of Wistar rats. Methods The pregnant rats were treated with DBTD by gavage at the doses of 0, 10, 20 and 30 mg/kg for days, from 12th-20th days of gestation. On 70th postnatal day, 10 male offspring rats were selected randomly from each group for measurements of the viscera coefficient of testis and the sperm count in epididymis, the activity of enzymes of testis was determined by spectrophotometry. Results There were no significant differences seen in the body weight gain of pregnant rats from days 12-20 of gestation, litter size, sex ratio of fetuses and body weight of male offspring rats between the experimental groups and the control group. 7.Molecular targeting therapy of gastric cancer

It has been demonstrated that surgery is currently still the most effective approach to completely cure gastric cancer. However, while only 30%-40% of gastric cancer can obtain curative outcome through pure surgery, most of patients with gastric cancer would died of recurrence or distant metastasis of the tumor. It appears to be more critical to search for other therapeutic strategies for gastric cancer other than surgery. Molecular targeting therapy has become the focus and hotspot of comprehensive treatment of gastric cancer.
8.Clinical value and nursing intervention of prone position ventilation in treating patients

Objective To investigate the effect of position ventilation(PPV) on oxygenation,the airway sputum drainage and hemodynamic of acute respiratory distress comprehensive syndrome(ARDS).Methods From June 2015 to April 2016,13 cases with moderate or severe ARDS were researched.The changes of PaO2,PaCO2,SpO2,PaO2/FiO2 and airway sputum drainage were observed as well as HR,MAP,CVP,CO,CI,SVV,etc.Results Totally 56 PPV were taken.Compared with pre-treatment of PPV,the number of PaO2 and PaO2/FiO2 increased from (74.7±13.18) mm Hg and 168.65±22.12 to (92.13±17.82) mm Hg and 208.80±24.04(P<0.05) respectively;SpO2 and airway sputum drainage increased from (95.32±2.76)% and (7.84±1.68)mL to (97.04±1.84)% and (13.25±3.55)mL(P=0.000).However,there were no significant differences in HR,MAP,CVP,CO,CI and SVV(P>0.05).Conclusion PPV can effectively improve oxygenation in patients with ARDS and promote airway sputum drainage,and hemodynamic effects are not obvious.However,nursing intervention still should be strengthened.
9.Application of CT measuring puncture route in Percutaneous Verebroplasty

Objective To explore optimal puncture route and method for advance pierce accuracy when perform percutaneous vertebroplasty. Methods 248 cases (365 vertebrae )with osteoporotic vertebral body fractures in thoracic vertebral and lumbar vertebral were treated by percutaneous vertebroplasty. Total injured vertebraes were examined and measured puncture routes under CT before operation. Then according to measure route pierce and perform percutaneous vertebroplasty, when the pricker arrived preconceive place, take some radiographs obverse and side, after operation,scan injured vertebraes with CT again. Pierce accuracy was evaluated. Results 365 vertebrae were pierced successfully,the pierce successful rate was 100%. 324 vertebra piercing route were coincident with CT measure routes,coincident rate 88.8%. 41 vertebra piercing route weren' t coincident with CT measure routes, deflective rate was 11.2%. Conclusion CT measure puncture route and directing pierce was effective means when percutaneous vertebroplasty.
10.Molecular docking of variant of quinolone resistance-determining region in the DNA gyrase subunit A to substrates in Klebsiella pneumonia

Objective To investigate the binding capacities of two variants of quinolone resistance-determining region in the DNA gyrase subunit A to substrates in Klebsiella pneumonia (K.pneumonia).Methods Tertiary structures of two variants (type Ⅰ and type FH) of quinolone resistance-determining region in the DNA gyrase subunit A in K.pneumonia were predicted by homology modeling referring to that of wild type.Then,DOCK module in ArgusLab 4.1 software was used to perform molecular docking of two variants and wild type to seven kinds of quinolones substrates,and calculate binding free energies (△G).Moreover,numbers and distances of interaction between amino acid residues of DNA gyrase subunit A and ciprofloxacin were calculated.Results Molecular docking showed that binding free energies of type Ⅰ and type FH to pipemidic acid,ciprofloxacin,gatifloxacin were-26.607 50,-29.530 39,-29.493 09 kJ/mol and-26.696 44,-28.972 83,-29.590 50 kJ/mol,respectively,which declined greater than those of wild type (-27.188 82,-30.872 00 and-30.244 04 kJ/mol,respectively) and showed drug resistance.While binding free energies of type Ⅰ and type FH to levofloxacin were-29.013 81 and-29.497 57kJ/mol,respectively,and that of wild type was-28.016 20 kJ/mol.The binding free energies of type Ⅰ and type FH to nalidixic acid,norfloxacin,ofloxacin increased or declined.Moreover,if distance was less than 5 angstroms,atom pairs formed between wild type of DNA gyrase subunit A and ciprofloxacin had 16 pairs,while type Ⅰ and type FH had 2 pairs and 4 pairs,respectively.If distance was less than 4 angstroms,atom pairs formed between wild type and ciprofloxacin had 8 pairs,while type Ⅰ and type FH had no atom pairs.Conclusion Decline of binding capacities of two variants of DNA gyrase subunit A in K.pneumonia to ciprofloxacin played a role in drug resistance.
