Objective:To study the effect of ?-3 PUFA on the proliferation and apoptosis of tumor cells transfected with transmembrane TNF? (tmTNF?) expression vector containing peroxisome proliferator responsive element (PPRE). Methods:The tmTNF? expression vector was transfected into HL-60 and MCF-7 tumor cells. The effects of ?-3PUFA and/or exogenous tmTNF? on cell proliferation, cell cycle distribution and apoptosis were measured by growth curve, flow cytometry and DNA ladder assay. Results: The production of exogenous tmTNF?gene was located in the cell membrane. After treatment with?-3 PUFA, the expression of exogenous tmTNF? in transfectant was significantly increased. The proliferation was decreased in HL-60 and MCF-7 cells treated with 6.0?10-5 mol/L ?-3 PUFA,but cell cycle was arrested at G0/G1 and the DNA ladder was detectable only in HL-60 cells. These changes were more obvious in transfected cells than non-transfected cells. Conclusion:?-3 PUFA and the product of exogenous tmTNF? gene induced by ?-3 PUFA could inhibit tumor cell growth more effectually, and induction of apoptosis may play a key role in this process.

Objective To study the apoptosis and its molecular mechanism of MCF-7 cells induced by ?-3 polyunsaturated fatty acid(?-3 PUFA)combined with the product of exogenous transmembrane TNF-?(tmTNF-?) gene. Methods The tmTNF-? eukaryotic expression vector containing the PPRE-tk promoter was transfected into the human breast cancer MCF-7 cells. The effects of ?-3 PUFA and/or exogenous tmTNF-? on the cell proliferation and apoptosis were measured by MTT and DNA ladder assay. The activity of caspase-8 was examined by using special fluorescence substrate, and the expression of caspases(1,8 and 9) were analyzed by RT-PCR and Western blotting. The inhibitor of caspases was used to confirm the function of caspase. Results In 6.0?10~(-5)mol/L ?-3 PUFA-treated MCF-7 cells, only the growth suppression was found. In transfected MCF-7 cells after treated with ?-3 PUFA,not only the proliferation capacity was decreased but the DNA ladder was detectable. The expression changes of caspases(1,8 and 9) and caspase-8 activity were obvious in MCF-7 transfected cells treated with ?-3 PUFA.Growth inhibition and apoptosis induced by ?-3 PUFA and tmTNF-? were partly prevented by the special caspase inhibitor. Conclusion These results suggested that up-regulated expression and activity of caspase might promote MCF7 cells apoptosis induced by ?-3 PUFA and exogenous tmTNF-?,indicating that ?-3 PUFA and exogenous tmTNF-? could cooperate in inhibition of the MCF-7 cell growth and induction of apoptosis.caspase network pathway may play a key role in these processes.

Objective To discover the operating problems through analysis of the financial situations of the rehabilitation institutions. Methods According to the different regions and different levels of rehabilitation institutions, 416 representative institutions from 26 provinc-es, municipalities and autonomous regions were surveyed. Results 329 effective questionnaires were recalled. 57.75%institutions relied on the superior departments in charge of funding, 51.09%institutions were in breakeven state. They were lack of sustainable development pow-er, and the capital expenditure was mainly used for staff salaries and equipment purchase. Conclusion To solve the shortage of funds in the development of rehabilitation institutions, we should establish a long-term mechanism of internal and external compensation.

The Koch's triangle of 110 human hearts (adults 70, children 40) has been dissected. The morphology, relationship and landmark of the atrioventricular node were observed and measured. Muscle bundles from the interatrial septum and coronary sinus orifice above and below are connected with the node. In the children the tendon of Todaro is usually tendinous, but in the adults, its posterior part is usually muscular. Deep in the triangle, there is a pyramidal space bounded by the left and right atrial walls and the top of the interventricular septum, in which there are vessels and nerves leading to the atrioventricular nodal area. According to their construction, the Koch's triangle is divided into five parts. The functional and surgical significance of these structures has been discussed.

Objective To investigate the serum concentration of interleukin (IL)-17 in patients with active rheumatoid arthritis and the clinical implication. Methods Serum IL-17 level of 51 active RA patients were assessed by enzyme-linked immunosorbentassay (ELISA). The relationship between serum levels of IL-17 in active RA patients and clinic features were analyzed. Two independent samples t test, ANOVA for repeated, one-way ANOVA and Spearman's Chi-square test were used for statistical analysis. Results ① Serum IL-17 levels were elevated in active RA patients (365±63) pg/ml than in normal controls (86±13) pg/ml (P<0.01); ② Serum IL-17 levels in active RA was correlated with morning stiffiness duration, swelling index, Ritchie index, CRP, ESR, RF, X-ray score, DAS score (r =0.423, 0.213, 0.252, 0.276, 0.346, 0.251, 0.412, 0.398; P<0.05). ③ The levels of IL-17 between phase I (296±79) pg/ml and Phase II (360±89) pg/ml, phase II and phase HI (464±74) pg/ml, Phase I and Phase Ⅳ (369±83) pg/ml in RA group were statistically significantly different (P<0.05). Conclusion The levels of serum IL-17 in active RA patients are higher than the normal controls. The elevated serum IL-17 levels may be the useful markers for active RA.In addition,interleukin-17 is involved in disease progression and bony erosion of RA, and it maybe a new treatmenttarget of RA.

OBJECTIVETo establish social stress induced depression-like model in mice of C57BL/6 strain, and to assess its reliability using differenf behavioral methods.
METHODSTotally 20 male mice of C57BL/6 strain were divided into the normal group and the stress model group by random digit table,10 in each group. Another 10 CD1 mice were subjected to social stress. Mice in the normal control group received no stress, while those in the model group received social stress for 10 successive days. Behavioral assessment was performed using social interaction test (SIT), the elevated plus-maze (EPM) test, tail suspension test (TST), respectively. Serum cortisol level was detected by ELISA to assess the reliability of the model.
RESULTSIn the social interaction test when the social target (CDI mice) was inexistent, mice in the normal control group spent longer time in the social interaction zone and less time in the corner zone (P < 0.05); mice in the model group spent less time in the social interaction zone and more time in the corner zone (P < 0.05). Compared with the normal group when CDI mice existed, mice in the model group spent less time in the social interaction zone and more time in the corner zone (P < 0.05). Compared with the normal control group, the total times for entry into open arms, close arms, and the maze were obviously reduced (P < 0.05), and the proportion of entering open arms was significantly reduced (P < 0.05) in the model group. In TST, the motionless time within the last 4 mm was prolonged in the model group (P < 0.05). The serum cortisol level in the model group was obviously elevated (P < 0.01).
CONCLUSIONSocial stress induced depression-like animal model in mice of C57BL/6 straineasquite reliable and possibly suitable to be used in integrative medicine research of combination of disease and syndrome model.
