1.Research advances of Mfn2 in tumor

Mitofusin 2 (Mfn2) plays an important role in the process of mitochondrial fusion, and is involved in regulating mitochondrial function and morphological changes. Studies have found that Mfn2 has a tumor suppressor effect in a variety of malignant tumors and their cell lines, including cervical cancer, hepatocellular cancer, pancreatic cancer, breast cancer, and bladder cancer. The difference in the expression of Mfn2 in malignant tumor tissues suggests that Mfn2 may play an important role in the proliferation, apoptosis, invasion and migration of malignant tumor cells. As a cancer-related gene, Mfn2 provides new ideas for gene therapy of malignant tumors.
2.Voxel-based analysis of diffusion tensor indices in the brain of the patients with type Ⅱ diabetes mellitus

Objective To evaluate early occult brain damage by diffusion tensor imaging in patients with type Ⅱ diabetes mellitus.Methods High resolution three-dimensional T1-weighted fast spoiled gradient recalled echo MRI images and diffusion tensor imaging were obtained from 16 patients with type Ⅱ diabetes mellitus (T2DM) and 16 normal controls (NC).Average apparent diffusion coefficient (ADC) map and fractional anisotropy (FA) map were calculated and analyzed with voxel-based analysis.Analysis of covariance was performed to compare ADC and FA between T2DM and NC.Results Increased ADC values of gray matter were demonstrated in right superior temporal gyrus,middle frontal gyrus,anterior cingulate gyrus,middle cingulate gyrus,inferior occipital gyrus,and left lingual gyrus,operculum orbitale,fusiform gyrus and parahippocampal gyrus in T2DM (voxel numbers in clusters were 176-3125,P < 0.01).And increased ADC values of white matter were demonstrated in the regions of right middle temporal lobe,linual lobe,middle frontal lobe,calcarine gyrus,superior parietal lobe,and left operculum orbitale in T2DM patients compared with NC (voxel numbers in clusters were 200-781,P < 0.01).Decreased FA values of gray natter were mainly located in left thalamus,bilateral caudate nuclei and right cuneus in T2DM(voxel numbers in clusters were 210-1402,P <0.01).Decreased FA values of white matter were demonstrated in the regions of right cuneus and the body of corpus callosum in T2DM (voxel numbers in clusters were 206,253,P < 0.01).Conclusions The occult brain damage is demonstrated as increased ADC values and decreased FA values in regional brain parenchyma in type Ⅱ diabetes mellitus.Voxel-based analysis of diffusion tensor indices may be a simple and effective tool for the evaluation of occult brain damage in type]][diabetes mellitus.
3.Clinical analysis of renal Beh(c)et's disease: 20 cases reports

Objective To investigate the clinical and pathological characteristics of renal involvement in Beh(c)et's disease (BD).Methods A retrospective analysis was carried out in BD patients complicated with renal damage who were admitted to Peking Union Medical College Hospital from June 1998 to July 2012.Results Twenty patients with renal involvement constituted 3.2% of all the 618 hospitalized BD patients.The presentation of renal disease was chronic glomerulonephritis in 6 patients (1 with nephrotic syndrome),renal tubular acidosis in 1 patient,renal artery stenosis in 7 patients,renal vein thrombosis in 1 patient,and chronic renal failure of unknown etiology in 5 patients.Kidney biopsy was performed in 5 patients,3 of them revealed glomerular minor lesion,mild mesangial proliferative glomerulonephritis and chronic tubularinterstitial nephropathy,respectively.The other 2 patients underwent a second biopsy,one with glomerular minor lesion transforming into IgA nephropathy of grade Ⅲ on Lee's glomerular grading system after 6 years,and the other with IgA nephropathy of grade Ⅱ progressing to grade Ⅳ after 2 years.After the diagnosis of renal BD,one patient with uremia underwent peritoneal dialysis,while the remaining 19 patients received immunosuppressant (or with combination of glucocorticoid,angiotensin converting enzyme inhibitors and angiotensin Ⅱ receptor blockers.etc.) therapy.Among the 8 patients with renal vascular involvement,2 underwent surgery,and several received anticoagulant therapy.During the follow-up of 13 patients,the urine protein quantifications were reduced,and renal functions remained relatively stable.Conclusion Renal damage is relatively uncommon in BD patients.There are various clinical spectrums for renal BD.Routine screening with urinalysis,serum creatinine and imaging studies should be carried out for the early diagnosis of renal BD.
4.Antagonistic effects of acanthopanasia and hoelen on mutagenesis induced by cadmium

0.05). Conclusion Both acanthopanasia and hoelen have no mutagenic toxicity founded on the genetic damage of somatic cells,and have significant antagonistic effects on the genetic damage induced by cadmium sulfate,and they are good antimutagents.

Objective To observe the effect of active components(glycyrrhizic acid monoaminate,total ginsenoside and to-tal ophiopogonoside)of Zhi Gancao Decoction(ZGD)and their combination on the triggered activity and myocardial dam-age in the isolated ischemia -reperfusion rat heart.Methods Isolated rat heart was treated with L angendroff perfusion and monophase action potential technology was applied.Results &Conclusion Total extract of ZGD,single active c omponents and their combination can markedly l ower the incidence of cardiac triggered activity,prevent the myocardiu m from injury and decrease the incidence of arrhythmia induced by ischemia -reperfusion in rats and the effects of their com bination are positively related to the doses.
6. Research Progress on Pathogenesis of Hepatic Encephalopathy

Hepatic encephalopathy (HE) is a kind of neuropsychiatric syndrome basing on metabolic disorder caused by acute or chronic liver dysfunction or abnormal portosystemic shunt. It is one of the common complications and causes of death in various end-stage liver diseases. The clinical manifestations of patients with HE can vary from personality changes, behavior abnormalities to disturbance of consciousness and coma. The pathogenesis of HE is complex. Theories about ammonia neurotoxicity and alterations in neurotransmitters have been widely accepted. Roles of inflammation, gut microbiota, premature astrocyte senescence, impaired cerebral microcirculation, hemichannel dysfunction, and bile acid also attract much attention. In this review article, the research progress on pathogenesis of HE was briefly summarized.
7.Expressions and clinical significances of Livin and vascular endothelial growth factor in human pancre-atic carcinoma

Objective To investigate the expressions of Livin and vascular endothelial growth factor (VEGF)in pancreatic carcinoma and their cilinical significances.Methods The expressions of Livin and VEGF proteins were tested by immunohistochemistry in 68 cases of pancreatic carcinomas and 44 cases of adja-cent paracancerous tissues.Results The positive rates of Livin in pancreatic carcinomas and adjacent paracan-cerous tissues were 73.5% and 4.5% respectively,and the difference was statistically significant (χ2 =48.137,P<0.001).The positive rates of VEGF in pancreatic carcinomas and adjacent paracancerous tissues were 69.1% and 13.6% respectively,and the difference was statistically significant (χ2 =29.147,P <0.001).The expressions of Livin and VEGF were related with tumor differentiation (χ2 =6.061,P=0.014;χ2 =6.592,P=0.010),TNMstage (χ2 =4.175,P=0.041;χ2 =9.992,P=0.002),lymph node metasta-sis (χ2 =11.731,P=0.001;χ2 =12.002,P=0.001)and neural invasion (χ2 =9.950,P=0.002;χ2 =7.433,P=0.006).Significantly positive correlation was found between the expressions of Livin and VEGF by using Spearman correlation analysis (r=0.320,P=0.008).Survival analysis showed that the expressions of Livin and VEGF were independent prognostic factors in pancreatic carcinoma.Conclusion Livin and VEGF involve in the development,migration and metastasis of pancreatic carcinoma.Livin may upregulate the expres-sion of VEGF,which may lead to the angiogenesis and migration in pancreatic carcinoma.
8.Application of Six-minute Walk Test in Acute Stroke Patients

Objective To apply Six-minute Walk Test (6MWT) in acute stroke patients and analyze the results. Methods From Novem-ber, 2014 to January, 2015, 29 acute stroke patients with multiple or single infarction accepted 6MWT. The distance, distance per minute, blood pressure and heart rate before and after test, adverse events or discomforts during the test were recorded. Weak and fatigue scores were calculated. Results The mean of distance in acute stroke patients was (240.1±17.2) m, that was (46.8±3.3)%of the predicted values in the healthy. Male patients walked (255.4±21.40) m, not different from (214.9±28.4) m in female (t=-1.151, P=0.26). The distance liked to be negatively correlated with age (r=-0.356, P=0.058). The distance was similar in each minute. Weak score was (46.8±3.3)%, fatigue score was mostly between-20 and 20, and no correlation was found between them. Blood pressure and heart rate increased significantly at the end of the test (t>2.476, P<0.05). Few of patients complained mild discomforts. Conclusion 6MWT is safe and suitable for acute stroke patients for physical mobility judgement, but adverse events need to be cared.
9.Effects of a new matrix metalloproteinase inhibitor, MMI-166, in nude mouse xenografts of human pancreatic cancer

Objective To investigate of the MMI-166 on the expression of MMP-2,MMP-9 and the cell apoptosis of nude mouse xenografts of SW1990 human pancreatic cancer cells.Methods Establishment of control and experimental groups,randomly,the human pancreatic cancer xenograft model of SW1990 was constructed.The control group was treated with normal saline,and experimental group was treated with MML-166 (200 mg · kg-1 · d-1).The tumor volume and tumor inhibition rate was measured by vernier caliper through length and short diameter.The expression of MMP-2 and MMP-9 protein was observed using immunohistochemistry in the tumor tissues.Apoptosis index was detected by deoxynucleotidyl transferase-mediated nick end labeling (TUNEL method).Results The tumor volume of MMI-166 group (1252.30± 464.84) mm3 was less than the control group (2241.82±208.06) mm3,significantly.The inhibition rate was 34.47% between the experimental groups (treat with MMI-166) (1.42±0.15) g and control group (2.17±0.20) g.The expression of MMP-2 (2.80 ± 1.10) % and MMP-9 (2.60 ± 1.52) % protein was significantly downregulated in MMI-166 group,compared with the control group.Apoptotic index in the experimental group (75.60±9.71) % was higher than the control group (17.40 ± 10.14) %,significantly.Conclusion The mechanism of MMI-166 inhibiting pancreatic tumor growth and inducing apoptosis may be related to the suppression of MMP-2 and MMP-9 protein expression.
10.Expression of CD4+ CD25+ FOXP3+ regulatory T cells in peripheral blood of patients with hepatocellular carcinoma

Objective To determine the expression of CD4+ CD25+ FOXP3+ regulatory T cells(Treg cells) in peripheral blood of patients with hepatocellular carcinoma (HCC) and investigate the clinical significance of Treg cells determination in clinical practice. Methods Flow cytometry was employed to measure the levels of CD4+ CD25+ T cells and CD4+ CD25+ FOXP3+ T cells in peripheral blood of 18 HCC patients, 26 hospitalized patients without HCC (clinical control) as well as 24 healthy persons (healthy control). Results The percentage of CD4+ CD25+ T cells in total CD4+T cells isolated from the HCC patients(4.25% ± 3.98 % ) was elevated significantly compared to that in the clinical control group (1.34% ± 1.14%) or healthy control group (1.29% ±0.95%) (both P<0.01). There was no difference in the percentage between the clinical control group and the healthy control group (P>0.05). Meanwhile, the ratio between CD4+ CD25+ FOXP3+ T cells and CD4+ T cells in HCC patients (2.94%0.91%) also increased significantly compared to that in the clinical control group (0.76% ± 0.34%) or healthy control group (0.81% ± 0.29%) ( both P<0. 001), which showed a more obvious increasing tendency than the ratio of CD4+ CD25+ T cells and CD4+ T cells. No difference in percentage of CD4+ CD25+ FOXP3+ T cells in CD4+ T cells was found between the clinical control group and the healthy control group (P>0.05). Conclusion As the more accurate regulatory T cells, CD4+ CD25+ FOXP3+ T cells are able to detect the increase of population in HCC patients. Therefore, it is important to determine the levels of CD4+ CD25+FOXP3+ T cells in HCC patients for prevention and treatment of malignancy.
