Objective In vitro detection of anti-proliferative effects of P161 combined with cisplatin ( DDP) on multiple cancer cells. Methods Growth inhibition rates of HepG2, HT29, IE8, Panc-1 and MA-782 treated by different concentrations of DDP,P161 and P161 combined with DDP were determined by MTT assay. Results DDP and P161 dose-dependently inhibited proliferation of multiple tumor cells. A synergistic effect was found in DDP combined with P161 and there was a significant difference in the effect between DDP combined with P161 and DDP alone (P<0. 05). DDP dose could be decreased to reach the same inhibitory effect. In the same concentration gradient of DDP combined with P161,the inhibition rate of Panc-1 was low and that of MA-782 was high. Conclusion P161 can increase the sensitivity of tumor cells to DDP. The combination of P161 and DDP can reduce the effective therapeutic concentration of DDP.

The individual variability should be considered in precision medicine-prevention and treatment strategies.Medical research using genomics, proteomics, metabolomics, systems analyses, and other modern tools has made big progress. In 2002, the members of the Complex-Trait Consortium proposed to develop a new mouse genetics resource called the Collaborative Cross (CC).The CC is a genetic reference panel of recombinant inbred lines of mice, designed for the dissection of complex traits and gene networks. It will provide a powerful measure for functional studies of biological networks, which will be essential to understand the intricacies of disease processes.

Objective:To prepare compound Xiaoshangtong spray films and establish an HPLC method for quality control. Meth-ods:Chitosan hydrochloride and PVP as the main film-forming materials, and HPMC as the film-forming assitant agent, the com-pound Xiaoshangtong spray films were prepared. Lidocaine and mupirocin were simultaneously determined by HPLC. A Hypersil ODS2 column(250 mm × 4. 6 mm, 5 μm)was used. The mobile phase was composed of 0. 5% ammonium dihydrogen phosphate-methanol (40∶60, adjusting pH to 6. 0 ± 0. 5 with sodium hydroxide). The flow rate of mobile phase was 1. 0 ml·min-1 and the temperature of the column was 30 ℃. The detection wavelength was 222nm and the injection volume was 20 μl. Results: The linear range of lido-caine was 25. 0-400. 0 μg·ml-1(r=0. 999 7) and the average recovery was 100. 14% (RSD=1. 21%,n=9). The linear range of mupirocin was 25.0-400.0 μg·ml-1(r=0.999 9)and the average recovery was 101.13%(RSD=0.57%,n=9). Conclusion:The preparation process is reasonable. The established determination method is accurate and reliable, and suitable for the quality con-trol of the compound Xiaoshangtong spray films.

AIM:To investigate the effect of signal transducer and activator of transcription 1 ( STAT 1 ) on proliferation and interferon-β(IFN-β) sensitivity of human non-small-cell lung cancer H1299 cells.METHODS:STAT1 or EGFP gene was transfected into H1299 cells by the lentiviral vectors system. The cell number was counted under a mi-croscope and cell proliferation was tested by MTT assay. In addition, the cells transfected with STAT1 and EGFP were trea-ted with IFN-βand cell viability was measured by MTT assay. The protein levels of p-STAT1, ICAM-1 and PCNA were de-tected by Western blot.RESULTS: Over-expression of STAT1 inhibited H1299 cell proliferation (P<0.05).H1299 cells transfected with STAT1 gene had a higher sensitivity to IFN-βthan the control cells transfected with EGFP ( P <0.05).Overexpression of STAT1 increased the protein level of p-STAT1, and reduced IACM-1 expression in H1299 cells. Moreover, STAT1 enhanced STAT1 phosphorylation and downregulated the expression of PCNA in H1299 cells treated with IFN-β.CONCLUSION:STAT1 inhibits the proliferation and enhances the IFN-βsensitivity of non-small-cell lung cancer H1299 cells.

Journal of China Pharmaceutical University 2017;48(5):583-589
Extraction experiment of volatile compounds in disposable syringes from five manufacturers was undertaken by GC-MS with high-space injection and semi-volatile compounds were tested by GC-MS.MS detection ranged from 30 to 650 with electron ionization (EI) source.Vanillin was used as internal standard to monitor the sample pretreatment process.Liquid-liquid extraction (LLE) and solid-phase extraction (SPE) were compared to acquire a simple processing method with more extractables in syringe materials.HCl,NaOH,Tween 80 and ethanol-aqueous solution were used as extraction solvents. The method was successfully developed to determine the unknown volatile extractables and the results were used to investigate the safety of disposable syringes comprehensively.

Objective:To investigate the association between pulse pressure(PP) and new-onset diabetes in overweight and obese people. Methods:A prospective cohort study was conducted in overweight or obese participants selected from Kailuan Study who underwent 2006-2007 annual checkup and met the inclusion and exclusion criteria. PP was calculated by blood pressure and participants were divided into 4 groups according to PP quartile. The cumulative incidence of new-onset diabetes of different PP groups was calculated by Kaplan- Meier method and compare by Log- Rank test. The multivariate Cox proportional hazards model was used to analyze the association between different PP groups and new-onset diabetes. Results:During an average follow-up of 8.45 years, 8 922 diabetes was identified. The cumulative incidence rate of the Q1, Q2, Q3, and Q4 groups were 22.12%, 24.48%, 27.97%, and 33.44% respectively, which were statistically different( χ2=368.16, P<0.01). Cox proportional hazards regression analysis showed that after adjusting for multiple confounding factors, compared with Q1 group, the hazard ratio for diabetes in Q2, Q3, and Q4 groups were 1.07(1.00-1.14), 1.13(1.05-1.21), and 1.17(1.09-1.27) respectively. And the HR of diabetes event in pulse pressure(per 1 SD increase) was 1.04(1.02-1.07). Similar results were found in participants who were over-weight, obese, with normal blood pressure or hypertensive without drugs use. Conclusion:PP is positively correlated with the new-onset diabetes. High PP is one of the risk factors for developing diabetes in overweight and obese people.

In order to promote the connotation construction of the resident standardized training bases and improve the training quality, a tertiary hospital made key breakthroughs against the existing problems in the base under the guidance of national key training base standards in September 2020. Optimization and innovation were explored in terms of residential teaching supervision, performance appraisal system, regular teaching meeting, educational fund management, pilot project of full-time residential teaching secretaries, clinical skill center construction, cultivation of faculty development, and so on. These experiments have paved the way to high quality and sustainable development of residents standardized training. By the end of December 2021, the hospital had achieved significant progress in such aspects as residential teaching atmosphere, resident learning initiative, and training base branding. These achievements improved the quality of resident standardized training of the hospital in general, and provided a reference for promoting the high-quality development of residential training in China.
