1.Percutaneous transhepatic gallbladder drainage under color Doppler ultrasound in critically ill patients

Objective:To study percutaneous transhepatic gallbladder drainage(PTGD) under color Doppler ultrasound in critically ill patients with acute calculus cholecystitis.Methods:A paracentetic needl was stuck into the cholecyst after going straight through 1.5-2.5 cm hepar under color Doppler ultrasound.The operator took out the bullet core,put in a guide wire,extracted the needl,inserted a central venous catheter along the guide wire,and then drew out the bile and clusised the cholecyst with metronidazole until the extracted fluid was clear and bright.Results:PTGD immediately relieved the pain in the of upper right quadrant in all the patients.Body temperature and WBC descended obviously 24 hours and returned to normal 5 days after the operation. Conclusion:Color Doppler ultrasound guided PTGD,easy to perform and with few complications and high suness rate,is an returned effective and reliable procedure for critically ill patients who are intolerant of cholecystectomy or for whom the operation is difficult to perform.
2.Pharmaceutical Care of Parenteral Nutrition Therapy in A Patient with Hyperemesis Gravidarum

Objective To explore the role of clinical pharmacists in nutrition support therapy in the patients with hyperemesis gravidarum. Methods The clinical pharmacist played a positive role in nutrition support care of a patient with hyperemesis gravidarum by analysising disease characteristics and adverse drug reactions, providing suggestion on the selection of fat emulsion and offering an individualized pharmaceutical care. Results The clinical pharmacist recognized the potential risk in nutrition support plan, took modifications timely, and prevented the occurrence of unfavorable clinical outcomes. Conclusion The participation of clinical pharmacists in nutrition support therapy of the patients with hyperemesis gravidarum is beneficial to improve the efficacy and safety of nutrition support and promote the rational use of drugs.
3.An epidemiological survey of functional dyspepsia and its relationship with psychological factors in college students of Zhejiang Province

Objective To investigate the prevalence of functional dyspepsia (FD) and its relationship with psychological factors in college students of Zhejiang Province. Methods The students of two colleges in Zhejiang Province were selected.The multi-stage stratified random cluster sampling method was conducted.The Rome Ⅲ adult FD diagnosis questionnaire (ROME Ⅲ-DQ) and psychological symptoms self rating scale questionnaire (SCL-90) was applied in the investigation.The data were analyzed by x2 test and t test.Results A total of 1870 college students of Zhejiang Province were surveyed.The prevalence of FD was 5.78%.The prevalence in female was higher than that in male (7.53 % vs 4.14 %,x2 =9.884,P<0.05).The prevalence in senior students was highest in all grade (x2 =13.83,P<0.05).In FD subtypes,postprandial distress syndrome was more than epigastric pain syndrome.The functional constipation and functional defecation disorders were most common in the overlap with other functional gastrointestinal disorders. In SCL-90 questionnaire survey,the scores of each factor of FD group were higher than those of the healthy control group.Conclusions The prevalence of FD was high in the college students of Zhejiang Province. The incidence was correlated with psychological factors.
4.An epidemiological study of functional bowel disorders in Zhejiang college students and its relationship with psychological factors

Objective To study the epidemiologic features of functional bowel disorders (FBD)among medical students and science and engineering students in Zhejiang province,China,and to explore the relationship between psychological factors and FBD.Methods This study was a survey of random sampling based on the Rome Ⅲ Diagnostic Questionnaire ( Rome Ⅲ -DQ) conducted from November 2010 to March 2011.The survey was carried out among medical students and science and engineering students respectively in two universities in Zhejiang province.Symptom Check List 90 ( SCL-90 ) was used for psychological analysis.Results Among 1870 participants,1033 were diagnosed as FBD based on Rome Ⅲ criteria,with an overall incidence of 55.24%.The majority cases were unspecified functional bowel disorder,with an incidence of 26.58% (497/1870),others were functional constipation 16.95%(317/1870),irritable bowel syndrome 6.90% ( 129/1870),functional bloating 4.12% (77/1870) and functional diarrhea 0.70% (13/1870).The prevalence varied in different genders,majors and grades.In the multivariate analysis,women had a higher risk of any FBD than men [ 68.33% (617/903) vs 43.02%(416/967) ] except for functional diarrhea; and medical students had a higher risk of any FBD than science and engineering students [ 68.53% ( 734/1071 ) vs 37.42% ( 299/799 ) ].Generally higher grade undergraduates had relatively higher risks than lower grade students,especially in medical students.Among the participants with FBD,44.05% (455/1033) had other functional gastrointestinal disorders.In those overlapped parts,9.00% ( 93/1033 ) had belching disorders,and 8.23% ( 85/1033 ) had functional dyspepsia.Students with FBD got higher scores in SCL-90 than healthy students.Conclusions This study revealed a high rate of FBD among college students in Zhejiang province.The prevalence varied in different majors,and female higher than male.Psychological factors also had close relationships with FBD.

Objective To investigate the effects of antioxidant response element (ARE) activator- 5,6-dihydrocyclopenta[ C ]-1,2-dithiole-3-thione (CPDT) on organotypic spinal cord cultures and to study whether this activation can protect motor neurons from oxidative stress.Methods Organotypic spinal cord cultures were prepared using lumbar spinal cord slices from 8-day-old rat.Threo-hydroxyaspartate (THA) was continuously added into the culture medium for 3 weeks,which caused selective motor neuron death. Thus,the in vitro model of amyotrophic Lateral sclerosis (ALS) was established.15,30 ?mol/L of CPDT were added into the culture medium respectively.Ventral motor neurons survival was evaluated by immunohistochemical staining with monoclonal antibody SMI-32,a nonphosphorylated neurofilament marker. Ultrastructure was observed with electronic microscope.Results The pretreatment of organotypic spinal cord cultures with different concentrations of CPDT significantly increase the total number of ventral motor neurons (15?mol/L:(15.81?6.97) perexplant;30?mol/L:(16.25?6.74) perexplant respectively) compared with THA group ((5.31?5.76) perexplant) and the former had plentiful neurite extensions (n= 15,P
6.Effect of Polygonum Cuspidatum Extract on the Expression of ICAM-1 and NF-?Bp65 in Synovial Membrane of Acute Gouty Arthritis Rats

Objective To study the therapeutic mechanism of Polygonum cuspidatum extract on the gouty arthritis induced by monosodium urate (MSU). Method Acute gouty arthritis rats model was induced by MSU. Twelve hours after modeling,the positive expression area and integral optical density of intercellular adhesion molecule-1 (ICAM-1) and nuclear factor-?B p65 protein (NF-?Bp65) in synovial membrane were observed by immunohistochemical assay. The effect of Polygonum cuspidatum extract on ICAM-1 and NF-?Bp65 was observed. Results There existed less expression of ICAM-1 and NF-?B p65 protein on synovial membrane in the normal group,and their expression was much increased in the model group. Polygonum cuspidatum extract in the dose of 360 mg/kg significantly reduced the positive expression area and integral optical density of ICAM-1 and NF-?Bp65 protein. Conclusion Polygonum cuspidatum extract can inhibit the abnormal expression and activation of ICAM-1 and NF-?Bp65 in synovial tissue,which may be an important therapeutic and preventive mechanism for gouty arthritis.
7.miR-126 promotes endothelial progenitor cell migration and targets KANK2

Objective To investigate the role of miR-126 (micro RNA-126) in rat endothelial progenitor cells (EPCs) proliferation and migration and the starget gene of miR-126 by bioinformatics and experimental survey.Method EPCs were transfected with control oligoes and miR-126 mimics or inhibitor by electroporation.MTT was performed to evaluate the growth of EPCs subjecting to miR-126 overexpression.Cell migration analysis was done by wound healing and transwell assay.The target genes of miR-126 were predicted by TargetScan and validated by Western blot.Result (1) miR-126 mimics promoted EPCs growth at 24 h post cell transfection (P < 0.01).In contrast,the EPCs growth was immue from miR-126 application at 48 and 72 h.(2) Both the wound healing and transwell assay show that miR-126 promotes EPCs migration (P < 0.01) and miR-126 inhibitor inhibits EPCs migration (P < 0.01).(3)It is predicted that KANK2 is the potential target gene of miR-126 by TargetScan online software.(4) The results of Western blot indicated that miR-126 mimics repress the expression of KANK2 compared with NC but miR-126 inhibitor enhances KANK2 expression.Conclusions miR-126 has a transient effect on the promotion of EPCc growth.miR-126 promotes EPCs migration and targets KANK2 protein.
8.Effects of intraoperative expansion on expression of stenosis-related genes in saphenous vein graft

Objective To investigate the effect of high pressure distention on the expression of stenosis-related genes of saphenous vein graft(SVG) during the coronary artery bypass grafting(CABG).Methods The biopsy specimens of saphenous vein collected from 10 patients who have undergone CABG,were divided into expansion group and no-expansion group.Real-time PCR and immunohistochemical staining were performed for examination of mRNA and protein expression of VE-cad,Egr-1,VCAN respectively.Student's t and Chi-square test were used to do statistic analysis.Results The results of RT-PCR showed that the mRNA transcription of Egr-1,VCAN in the expansion group were statistically significantly higher than those in no-expansion group(P<0.05).The mRNA transcription VE-cad in expansion group was statistically significantly lower than that in the no-expansion group(P<0.05).The immunohistochemical staining results showed that the expression of Egr-1 and VCAN in expansion group were significantly stronger than those in no-expansion group,while the expression of VE-cad was significantly lower than no-expansion group.Conclusion The intraoperative expansion of SVG can increase the expression of stenosis-related genes Egr-1 and Versican,and decrease the expression of stenosis-related gene VE-cad,which may be related with the SVG stenosis after CABG.
9.The activin receptor-like kinase Ⅰ promotes proliferation and migration in HUVECs

AIM: To study the function of activin receptor-like kinase Ⅰ(ALK1) gene in vascular endothelium.METHODS: The human umbilical vein endothelial cells(HUVECs) were cultured,and the change of expression of ALK1,ALK5 in activation of HUVECs was analyzed.The full-leng coding sequence of ALK1 was cloned into pcDNA3.1+using standard protocols.The constructed pcDNA3.1+ALK1 plasmid were transfected into HUVECs.The proliferation and migration of HUVECs were detected by boyden champer and flow cytometry.RESULTS: The expression of ALK1 was up-regulated in resolution.ALK1 promoted the proliferation and migration of HUVECs.CONCLUSION: ALK1 has an important function in remodeling by promoting the proliferation and migration of endothelial cells.
10.Construction of the eukaryotic expression vector of mouse Sim2 and its effect on cell cycle in PC12 cells

AIM: To observe the effect of mouse Sim2 (mSim2) eukaryotic expression vector transfection on the cell cycle in PC12 cells in vitro and to explore the role of Sim2 in the pathogenesis of Down syndrome. METHODS: The full open reading frame of mSim2 was amplified by reverse transcription-polymerase chain reaction (RT-PCR) and cloned into the vector pcDNA3. Then the constructed pcDNA3-Sim2 vectors were transiently transfected into PC12 with Lipofectamine~ TM . The expression of mSim2 was detected by RT-PCR. The effect of mSim2 on the cell cycle was observed by flow cytometry. RESULTS: The eukaryotic expression vector mSim2 was successfully constructed. There was notable expression of mSim2 in the cells transfected with pcDNA3-Sim2. There were more cells in G_0/G_1 phase in the pcDNA3-Sim2 transfected cells than that in the control (P
