1.The expression of TGF-?1 and type I receptor following moderate lateral fluid percussion brain injury in rats

Objective Study on the pattern of changes of TGF-?1 and type I receptor occurred in the experimental fluid percussion brain injury model for the purpose of providing the scientific basis for molecular pathological diagnosis, forensic identification, clinical treatment as well as further ascertaining the molecular mechanism of brain injury. Methods Male Sprague-Dawley rats were divided into normal control, sham operation control and injury groups. The rats of injury groups were subjected to moderate lateral fluid percussion brain injury (0.2 MPa). The injury groups were then subdivided into 30min, 1h, 3h, 6h, 12h,1d, 3d and 7d sub-groups according to the time elapsed after injury. Immunohistochemistry SP method was used for studing the immunoreactivity of both TGF-?1 and T?R I factors. Results (1) In the brain of normal control and sham operation control groups, the low expression levels of TGF-?1 and T?R I were observed; (2) The gradual increase of TGF-?1 and T?R I immunoreactivity could be observed 1 to 3d after injury both in cortex and brain stem, and sustained at the high level up to 7d; (3) In hippocampus, the gradual increase occur during 12h to Id after brain injury, and sustained the high level at 3d, then declined at 7d. Conclusion The results suggested that brain injury induced the gene eypressions of the TGF-?1/ T?R I . The TGF-?1/ T?R I may contribute to maintance of nerve cell survival and the repair of damaged neural tissues after CNS injury and the patterns of their level change were quite regular and can be used for timing of injury in forensic medicine aspect.
2.Level of bFGF and its receptor FGFR1 mRNA in rat brain following injury caused by moderate lateral fluid percussion

Objective Study on the pattern of changes of bFGF and FGFRl mRNA occurred in the experimental brain injury model in order to provide scientific basis for the diagnosis, forensic identification and clinical treatment, and also for further ascertaining the molecular mechanism of brain injury. Methods Male Sprague-Dawley rats were divided into 3 groups: normal control, sham operation, and injury groups. The rats of injury groups were subjected to moderate lateral fluid percussion brain injury (0.2MPa). The injury groups was then subdivided into 30min, 1h, 3h, 6h, 12h, 1d, 3d and 7d groups according to the time elapsed after injury. In situ hybridization (ISH) and RT-PCR were used for studying the mRNA expression of both bFGF and FGFRl factors. Results (1) In the brain of normal control and sham operation control groups, mRNA levels of bFGF and FGFRl were low; (2) There is gradual increase of bFGF and FGFRl mRNA levels could be observed 6h to 3d after injury both in cortex and brain stem, then partly declined at 7d; (3) In hippocampus, the gradual increase occurred during 3h- 1d after injury, then partly declined at 3d, and returned to basal level at 7d. Conclusions The results suggested that brain injury induced the gene expressions of bFGF and FGFR1. The bFGF may contribute to maintenance of nerve cell survival and the repair of damaged neural tissues after CNS injury and the patterns of their level change were quite regular. It is potentially useful for timing of injury in forensic medical practice.
3.Typing of fragment length polymorphism marker with primer SNP-binding at the last second 3'end

Objective To successfully get the full PCR alleles of the Insertion/Deletion marker rs10644346 in which a SNP-binding exists at the 3'end region of the primer.Methods Based on the AS-PCR,a common forward primer and two reverse primers with allele-specific oligonucleotides at the last second 3'end instead of the terminus were tentatively designed for typing 150 unrelated individuals and 10 father-mother--child trios from Htnan province in South-central China.Simultaneously,9 samples were typed with all the above three primers (the two primer sets which consist of the common forward primer and one of the reverse primers).Results PCR amplicons were well detected in the 150 unrelated individuals after being typed with the three primers,and the amplified fragments of parental and filial generations among 10 father-mother-child trios conformed to Mendel's principles.Allele missing was found in the two-primer group.Conclusion The primers designed by locating the specific nucleotide at the last second 3'end rather than terminal position were demonstrated also effective in getting specific alleles if perfect mismatch and PCR conditions are guaranteed,and the design strategy can provide an optional reference to rescue markers of SNP-binding primers for forensic practice.
4.Clinical Pathological Analysis of 86 Cases of Malignant Melanoma.

Objective To study the clinical pathological characters of malignant melanoma.Methods To analyze the clinical pathological materials of 86 cases of malignant melanoma.Results The original sites of 49 of the whole 86 cases locate at limbs,among those 26 cases at sole or heel.49 cases of them have melanotic nevus history,36 cases have trauma history.30 cases were diagnosed as carcinomatous change of ulcer before the surgical operation while 51 cases were diagnosed as other pathological diseases.Only 5 cases were diagnosed as malignant melanoma.The major histological type of the 86 cases is the epitheliod cell type,which made 50%(43 cases) of all the cases.61 cases(70.9%) of them has lymphnodes metastasis when the diagnosis was made.Conclusion Malignant melanoma most commonly located at limbs,especially at sole or heel.It's inclined to misdiagnose the disease and the disease is likely to metastasize before surgical operation.Final diagnosis should be confirmed by pathology examination.
5.Investigation of magnetic motor evoked potentials in patients with Graves′ disease

Magnetic motor evoked potentials (MEP) were measured in 46 patients with Graves′ disease (GD) and 50 normal controls, and its latency、amplitude and central motor conduction time (CMCT) were recorded. Prolonged latency, lowed amplitude and shortened CMCT in MEP suggest the damage of the neurons.

8.Injectable small intestinal submucosa is co-cultured with adipose-derived mesenchymal stem cells in vitro

BACKGROUND:The decel ularized porcine smal intestinal submucosa is a kind of bioactive extracel ular matrix, which is mainly composed of col agen, glycoprotein, proteoglycan and rich in col agen, glycosaminoglycan and various growth factors, and these components play an important role in promoting the differentiation and proliferation of tissue cel s. OBJECTIVE:To prepare the injectable smal intestinal submucosa and to investigate its co-culture with rat adipose-derived mesenchymal stem cel s in vitro. METHODS:The injectable smal intestinal submucosa and rat adipose-derived stem cel s were prepared. Cel counting kit-8 test for cel proliferation:Passage 3 adipose-derived stem cel s were seeded onto the injectable smal intestinal submucosa (experimental group) and cel s cultured under normal condition as control group. The cel proliferation was observed at 1, 3, 5 and 7 days of incubation. Live/dead staining test for the survival of cel s:Passage 3 adipose-derived stem cel s were respectively cultured in the injectable smal intestinal submucosa extracts (experimental group) and complete culture medium (control group). Cel survival was determined at 1, 3, 5 and 7 days of culture. RESULTS AND CONCLUSION:Scanning electron microscope oval and strip adipose-derived stem cel s adhered onto the material. The absorbance values in the experimental group were higher than those in the control group at 1 and 5 days of incubation (P<0.05). Cel survival:The number of cel s appeared to be in a rising trend with time in both two groups;after 1-day co-culture, al cel s in the two groups survived. Then dead cel s appeared in both two groups, showing no significant difference. These results show that the injectable smal intestinal submucosa exhibits a good cytocompatibility.
9.Effect of psychological factors on quality of life in patients with irritable bowel syndrome with diarrhea

Objective To investigate the effects of anxiety and depression on the quality of life (QOL) in patients with irritable bowel syndrome with diarrhea (IBS‐D) and the difference in gender .Methods IBS‐D patients met the Rome Ⅲ diagnostic and subtyping criteria were consecutively enrolled .The intestinal symptoms , psychological status , and QOL of patients were evaluated using IBS‐specific symptom questionnaires , the Hamilton anxiety scale (HAMA) ,the Hamilton depression scale (HAMD) ,and the Chinese Version of IBS‐QOL instrument .The data were analyzed by chi‐square test variance analysis ,t test or Spearman rank correlation analysis .Results A total of 155 IBS‐D patients were enrolled .Among them ,115 were complicated with anxiety and/or depression .The number of male and female with comorbid psychological disorders was 69 cases(71 .13% ) and 46 cases(79 .31% ) ,respectively ,and the difference was not statistically significant (χ2 = 1 .267 ,P= 0 .26) . Compared with 40 patients without anxiety or depression ,there was no statistically significant difference (F=1 .143 ,P> 0 .05) in the scores of intestinal symptoms in 59 patients with comorbid anxiety alone and 56 patients with anxiety and/or depression (20 .85 ± 5 .84 vs .21 .71 ± 7 .47 vs .22 .87 ± 6 .09) .The total IBS‐QOL score of the 155 patients was 71 .61 ± 19 .22 .There was negative correlation between IBS‐QOL score and HAMA , HAMD scores(r= -0 .262 and -0 .268 ,both P= 0 .001) .The total IBS‐QOL score of patients with comorbid anxiety and depression or depression were lower than that of patients without anxiety or depression (66 .05 ± 22 .88 vs 77 .22 ± 15 .35 , F = 4 .412 , P = 0 .005) .Except health worrying and sexual ,the scores in six dimensions(including dysphoria ,interference with activity ,body image ,food avoidance and social reaction) were all significantly decreased (F= 3 .309 ,3 .279 ,4 .177 ,3 .765 ,6 .041 and 3 .830 , P= 0 .013 ,0 .012 ,0 .013 , 0 .007 ,0 .001 and 0 .010) .In male patients ,the total score of IBS‐QOL and the scores of dysphoria ,body image and social reaction of patients with anxiety and depression or depression were lower than those of patients without anxiety and depression (t = 2 .143 ,2 .110 ,2 .279 ,3 .061 ; P = 0 .036 ,0 .039 ,0 .027 ,0 .003) .In female patients ,the score of food avoidance domain significantly decreased (t= 2 .812 ,P= 0 .008) .The scores of IBS‐QOL and six dimensions (including dysphoria ,interference with activity ,body image ,food avoidance and social reaction) of patients with history of severe mental disorders and being abused were even lower than those patients without such histories (t= 3 .241 ,3 .433 ,2 .499 ,2 .296 ,2 .514 ,3 .413 and 2 .601 ;P= 0 .001 ,0 .001 ,0 .014 , 0 .023 ,0 .013 ,0 .001 and 0 .010) .Conclusions The QOL of patients with IBS‐D was significantly affected by psychological factors with gender differences .The approaches to improve the psychological status of IBS patients were helpful to enhance the efficacy of comprehensive therapy .
10.Analysis of D1S1656 polymorphisms in Hu'nan Chinese Han population

Objective To get the genetic information and forensic parameters for the STR locus of D1S1656 in Hu'nan Chinese Han population . Methods All 451 unrelated individuals of Hu'nan Chinese Han population were selected to analyze genetic polymorphisms by using the Powerplex 21 system kit. The deviation of allele frequencies from Hardy-weinberg equilibrium was tested and the forensic parameters were counted by software. Results The marker of D1S1656 showed no significant deviation from Hardy-weinberg expectations, and 16 alleles were found with Power of Discrimination was 0.954 and Power of Exclusion 0.667. Conclusion The STR marker of D1S1656 present rich polymorphisms in Hu'nan Chinese Han population and could provide effective application in forensic field.
