1.Correlation between parameters of contrast-enhanced ultrasound and microvessel density in rabbit atherosclerotic plaques

Objective To evaluate the correlation between quantitative parameters of contrastenhanced ultrasound(CEUS) and microvessel density (MVD) of rabbit atherosclerotic plaques.Methods Twenty-six male New Zealand rabbits were damaged abdominal aortic by balloon expanded,fed with high fat 12 weeks later.Thirty two plaques were detected using conventional ultrasound examination.The maximum area,luminal area stenosis rate and the maximum thickness of each plaque were recorded.CEUS was performed on 32 plaques and enhancement images were offline analyzed quantitatively by ACQ software and the parameters such as arrival time(AT),time to peak(TTP),basic intensity(BI),and peak intensity(PI) were acquired.Enhanced intensity (EI) was calculated according to the formula:EI =PI- BI.Then the experimentalrabbitswerekilledforpathologicalexaminationandCD34monoclonalantibody immunohistochemical detection.MVD was counted under the microscope.The correlation between the parameters of CEUS and MVD was analyzed.Based on pathological findings,the plaques were divided into vulnerable plaques and stable plaques.ResultsEI and MVD of vulnerable plaques group were significantly higher than that of stable plaques group [EI:(26.36 ± 1.44) dB vs (23.90 ± 2.92)dB,t =- 3.243,P =0.001 ;MVD:(5.23 ± 1.16)/mm2 vs (2.47 ± 1.12)/mm2,Z =- 4.378,P <0.001].The maximum area,luminal area stenosis rate,the maximum thickness and PI,AT,TTP had no significant difference between two groups( P >0.05).The value of EI showed significant positive correlation with MVD ( r =0.676,P <0.001).ConclusionsEI can accurately evaluate the angiogenesis of rabbit atherosclerotic plaques,which can be regarded as a useful index to distinguish between vulnerable and stable plaques.
2.Mechanism of Huanglian Jiedu Decoction in Regulating lncRNA NEAT1 to Alleviate Inflammatory Response in Abscess Wounds

OBJECTIVE
To explore the effect and mechanism of Huanglian Jiedu decoction in reducing inflammation of abscess from the perspective of lncRNA NEAT1.
METHODS
C57BL/6 mice were divided into blank group, model group, Huanglian Jiedu decoction group and control group. After the intervention, the tissue were separated and collected under anesthesia. Animal experiment: the wound tissue of each group was observed by morphology. Real-time PCR and Western blotting were used to detect lncRNA NEAT1, miR-146a-5p gene level changes and RELA, TNF-α, TGF-β1, α-SMA, Collagen Ⅲ protein expression respectively. Cell experiments: Real-time PCR was used to detect the changes of lncRNA NEAT1 and miR-146a-5p gene levels. The effect of flow cytometry on apoptosis rate was detected. Fluorescence intensity was measured by FISH. The expression of related proteins was detected by Western blotting.
RESULTS
Compared with model and control group, the wound was repaired well and the inflammatory cell infiltration in the wound of the Huanglian Jiedu decoction group was reduced. Compared with the model and control group, the level of lncRNA NEAT1 and protein expressions of RELA, TNF-α, TGF-β1 in the Huanglian Jiedu decoction group were significantly reduced, while the gene level of miR-146a-5p and the protein expressions of α-SMA and Collagen III were significantly increased(P<0.05). Compared with LPS+Lenti-control group, lncRNA NEAT1 gene level in LPS+Lenti-sh-NEAT1 group was significantly decreased, while miR-146a-5p gene level was increased(P<0.05). Flow cytometry showed that Huanglian Jiedu decoction could reduce the apoptosis rate and fluorescence intensity of lncRNA NEAT1(P<0.05). Compared with the blank group, the protein expressions of RELA, TNF-α, TGF-β1 in LPS group were significantly increased, while the protein expressions of α-SMA and Collagen III were significantly decreased (P<0.05). After sh-NEAT1 treatment, The regulatory effects of Huanglian Jiedu decoction on the expression of above molecule were reduced(P<0.05).
CONCLUSION
Huanglian Jiedu decoction can alleviate the inflammatory state of mouse abscess wound through lncRNA NEAT1, which is closely related to the regulation of miR-146a, RELA, TNF-α, TGF-β1, α-SMA and Collagen III expression.
3.Advances in the research of the relationship between wound temperature and wound healing

There are many factors that may affect the microenvironment of acute and chronic wounds.This article reviews the relationship between temperature factor in the external microenvironment of wound surface and wound healing.The temperature changes in different types and stages of wounds are closely related to the wound healing status.Therefore,wound temperature monitoring provides timely,reliable,and non-invasive method in the evaluation of wound status.As low temperature affects the physiological state of wound,relieving the low temperature state and maintaining normal temperature of the microenvironment of wound can promote wound healing.Further research is needed on the wound repair related effector cell proliferation and the mechanism of regulatory function to determine the optimal constant temperature and heat treatment duration needed for wound healing.
