1.Effects of inducible nitric oxide synthase on mechanism of osteoarthritis

Objective To observe the expression of inducible nitric oxide synthase (iNOS) in synovium of osteoarthritis and to investigate the relationship with the disease.Methods iNOS expression is investigated by means of immunohistochemical assay.Results By immunohistochemical assay , we found that there are significant expression of iNOS in synovium of osteoarthritis patients than that of control.Conclusion In OA ,there are high expression of iNOS in synovium ,so we concluded that iNOS may mediate the pathogenesis and development of OA.
2.Expression of PTEN,NF-KB p65 and Survivin in endometrial carcinoma and its clinical significance

Objective To explore the expression of PTEN,NF-KB p65 and Survivin protein and the influence in the genesis and development of endometrial carcinoma(EC).Methods The expression of PTEN.NF-KB and Survivin in 63 cases of EC and 20 cases normal endometrial tissue specimens were detected by immunohistochemistry.Resuits There were obvious differences among the positive rates of PTEN.N-KB p65 and Survivin protein in EC compared with in normal endometrial tissue specimens (P<0.001).The clinicopathological characteristics of endometrial carcinoma with PTEN,NF-KB p65 and Survivin were as follows:the expression of PTEN gene was positively correlated with the degree of histological differentiation(P<0.001),and it was negatively correlated with lymph node metastasis and TNM (P<0.005).There was inverse correlation between the expression of NF-KB p65 gene and the degree of histological differentiation(P<0.05).It was positively correlated with lymph node metastasis,deep myometrial invasion and TNM in EC (P<0.05).The expression of Survivin was positively correlated with the clinical stage and lymph node metastasis and deep myometrial invasion in EC(P<0.05).Conclusion There are difierent extent action of PTEN.NF-KB p65 and Survivin significantly in genesis and development of EC.Detection of PrEN combined with NF-KB p65 and Survivin is valuable for early diagnosing and evaluating malignancy extent of EC.
3.The analyses of hip joint lesionin ankylosing spondylities in X ray and clinic

Objective To improve the recognization of hip joint lesion in ankylosing spondylitis and it's related factors.Methods Pelvis computed radiography(CR) and HLA-B27 were examined in 100 cases of ankylosing spondylitis proved by clinical.Results 58% cases appeared differed degree of narrowing of the interticular space,osteoporosis on head of femur and acctabulum,cystis under the surface of acctabulum and joint inosculated.This probability was higner at thoe patients that had diaease at youthful or HLA-B27(+).Conclusion Hip joint lesions of ankylosing spondylitis appear characteristic signs in CR.It relates with the age that the disease come on and HLA-B27.
4.Secretory status of pancreas and pathogenesis of insulin resistance in rat with nonalcoholic fatty liver disease

Objective Rats with nonalcoholic fatty liver disease (NAFLD) were used to investigate the secretory state of pancreas and the expression of insulin receptor (IR). Methods NAFLD rat model was reproduced, and then the structure of pancreatic tissue, secretory states of ? and ? cells (serum, tissue) and the expression of IR were examined and determined by means of HE staining, ELISA, and immunohistochemistry. Results In all experimental groups, the structure of pancreatic tissue showed no obvious change; the blood sugar level tended to rise. The insulin level in serum began to elevate obviously at 4th week (P≤0.01), while the insulin content in tissue began to increase at 6th week (P≤0.01), and distributed mainly in the middle part of the pancreas with a tendency of elevation along with the time. The content of glucagon in pancreatic tissue began to increase at 8th week (P≤0.01), and reached the peak at 12th week. The expression of IR in tissue began to decrease at 6th week (P≤0.01), and then tended to be stable after 8th week. Conclusions In NAFLD, there was changes in secretory state of pancreas with the accompaniment of insulin resistance. There was a tendency of elevation of levels of insulin both in serum and pancreatic tissue, but the time of expression was different. The expression of glucagen shows an increase tendency, while the decrease of IR might be the crucial cause of insulin resistance in NAFLD.

Objective To study the expression of retinoblastoma(Rb) protein in patients with breast cancer and its significance.Methods The expressions of Rb protein were detected in 52 patients with breast cancer,20 patients with breast fibroadenoma or breast cystic hyperplasia(control group) by S-P immunohistochemical method.(Results The negative) rate of Rb expression in breast cancer(76.91%) was higher than that in control group(15.00%)(P0.05).Of all 46 follow-up patients with breast cancer,the peritoneal recurrence and metastatic rate was 10.53% in 38 cases of the negative expression of Rb protein,and while the rate was 0 in 8 cases of the positive expression of Rb protein,there was significant difference(P
6.Effect of letrozole on the proliferation of uterine leiomyoma cells in vitro

Objective To investigate the expression of aromatase cytochrome P450 (P450arom), the secretion of estradiol and the suppressive effect of aromatase inhibitor letrozole on the proliferation of uterine leiomyoma cells in vitro. Methods Uterine leiomyoma cells were cultured in vitro, after the action of letrozole with different concentration, MTT was used to examine the proliferative activity of uterine leiomyoma cells, RT-PCR and immunocytochemistry was used to determine the expression of the P450arom protein and mRNA, and the concentration of estradiol in media supernatant was measured by radioimmunity. Results Low dosage (0.06 μmol/L) of letrozole had no effect on the expression of P450arom protein and mRNA, and estradiol synthesis in uterine leiomyoma cell (P>0.05). However, high dosage(0,6-6 μmol/L) of letrozole could down-regulate the expression of P450arom, and to inhibit estradiol synthesis and the proliferation activity of uterine leiomyoma cells(P<0.05). Furthermore, the inhibition were shown to decline gradually along with the increase in the concentrations of letrozole. Conclusion The aromatase inhibitor letrozole could suppress the proliferation of uterine leiomyoma cells in the dosage-dependent manner by down-regulating the expression of the P450arom and decreasing the estradiol synthesis, suggesting that the drug may possibly be effective in the clinical treatment of uterine leiomyoma.
7.Surgical Treatment of Primary Hepatocellular Carcinoma Complicated by Intrabile Duct Obstruction

Objective To study the diagnosis and treatment of primary hepatocellular carcinoma(HCC) complicated by intrabile duct obstruction.Methods From Aug.1994 to Feb.2000,the clinical data of 13 HCC patients with intrabile duct obstruction caused by tumor were reviewed.Results There was no operative mortality and all patients were followed up for one year.10 patients were still alive,and one died.Conclusion Jaundice caused by intrabile duct tumor was not a contraindication for hepatectomy.Hepatectomy could gain a longer postoperative survival term.
8.HAI and/or PVI after radical resection of primary hepatic carcinoma for preventing tumor recurrence

Objective To study the effect of HAI and/or PVI after radical resection of hepatocellular carcinoma (HCC) for preventing recurrence.Methods From Jun. 1995 to Apr. 2000, 49 patients with HCC were undergone radical tumor resection. They were divided into 4 group: the treatment combined with HAI and PVI was given in group I(12 cases), HAV alone in group II (8 cases); PVI alone in group III (15 cases); nothing in group IV (14 cases). All patients were followed up for 6 months to 5 years. Results Recurrence rate at one year didn't show significantly lower in group I, II and III than that in grou IV. Survival rate in one year showed no significant difference in 4 groups. Recurrence rate in three years was significantly lower in group I and II than that in group IV. Survival rate in three years was significantly higher in group I and II than that in group IV. Recurrence rate and survival rate in five years showed no significante difference in four groups. Conclusions The treatment of radical resection of HCC combined with HAI and/or PVI is simple and safe. It is an useful method for preventing tumor recurrence.
9.Mutation of rspL gene of Mycobacterium tuberculosis in urine and its relationship with streptomycin-resistance

Objective To investigate the streptomycin(SM)-resistance of Mycobacterium tuberculosis (MTB) in urine and to examine their relationships with the mutation of rspL gene. Methods 47 strains of MTB were isolated from the urine of patients with renal tuberculosis.The mutation of rspL gene were analyzed by PCR-SSCP with silver-staining,and the strains were tested for the sensitivity to streptomycin. Results All the 47 strains had positive expression of rspL gene of MTB,with 28 SM-resistant and 19 SM-sensitive.Among the 28 SM-resistence strains 19(68%) had rspL gene mutation by SSCP testing.The 19 strains of SM-sensitive had no mutation. Conclusions Detecting mutation of MTB rspL gene in urine by PCR-SSCP can rapidly and exactly identify the MTB's sensitivity to streptomycin,which is significantly important in clinical treatment of anti-tuberculosis.
