1.Estabilishment of automated particle-enhanced turbidimetric immunoassay for the measurement of human and evaluation on methodology

Objective To evaluate methodologically particle‐enhanced turbidimetric immunoassay (PETIA ) for determining quantitatively Cystatin C .Methods Cystatin C was determined quantitatively with PETIA by Cobas 8000 automatic biochemical analyzer ,and imprecision ,veracity ,sensitivity ,linearity range of the method were investigated .The correlation of fresh serum sam‐ple results between PENIA and PETIA was analyzed .The reference interval was verified .Results The within‐run CV imprecision of samples with low and high concentration of adiponectin was 3 .67% and 1 .15% respectively ,the between‐run CV imprecision of samples with low and high concectration of adiponectin was 4 .08% and 1 .53% respectively .The accuracy of the measurement devi‐ation was only -1 .25% .The sensitivity was 0 .07 mg/L .When the adiponectin concentration was between 0 .2 -8 .0 mg/L ,the method showed good linearity .A strong correlation between PETIA and PENIA was confirmed (Y = 0 .945 8X+ 0 .048 6 ,r2 =0 .991 3 ,r=0 .995 6) .Among 40 cases of healthy physical examination sample ,2 cases were with cystatin C level out of the refer‐ence range supplied by reagent kit .The values that were more than 90% in validation interval .Conclusion The PETIA assay is a convenient ,rapid ,cheap ,accurate and reliable method for quantitating the human serum concentration of adiponectin .This assay is suitable for clinical routine analysis .
2.The development and assessment of Knee Osteoarthritis Self-Care Questionnaire for Elderly

Objective To develop Knee Osteoarthritis Self-Care Questionnaire for Elderly (KOSCQE), and test reliability and validity of the scale. Methods Based on self care theory, with reference to International Knee Documentation Committee Subjective Knee Form, combined with literature analysis and through semi-structured interviews, the item pool of the scale was established. A total of 148 elderly were investigated by questionnaire survey. Results KOSCQE extracted 5 common factors with 26 items, and the total variance contribution rate was 68.972%;the item load factor was 0.576 to 1.872, item level content validity index was 0.901-0.928, factor in overall content validity index of 0.916;total Cronbach αcoefficient was 0.898, test-retest reliability was 0.864, and the confirmatory factor analysis showed that:χ2/df=1.09, goodness of fit index =0.942, root mean square error of approximation=0.020, adjusted goodness of fit index=0.964, normed fit index=0.960, comparative fit index=0.948, relative fit index=0.931, incremental fit index=0.918, parsimony goodness of fit index=0.891. Conclusions The reliability and validity of the KOSCQE of the elderly accord with the requirements of the measurement of the scale, it can improve the elderly's awareness of self care and quality of life of the knee osteoarthritis.
3.Study on the anti-inflammatory effect of proanthocyanidins and its mechanism

Objective To investigate the anti-inflammatory effect of proanthocyanidins and its mechanisms.Methods Inflammation models such as dimethylbenzene-indueed ear swelling and carrageenan-induced hind paw edema in mice and rats were prepared.The contents of PGE2 in exudate from edema paws of rats were measured by ultraviolet spectrophotography and the protein expression of COX-2 in edema paws of rats by Western-blot and immunohistoehemistry(IHC)assay.Results Pro-anthocyanidins remarkably inhibited the ear edema induced by dimethylbenzene in mice at the dose of10 mg/kg and 20 mg/kg;paw edema of rats induced with carrageenan was significantly inhibited byproanthocyanidins 5,20 mg/kg ip from 2 to 5 h;proanthoeyanidins 5,20 mg/kg ip reduced the contents of PGE2 in exudate from edema paws of rats induced by carrageenan;proanthocyanidins 5,20 mg/kg ip inhibited the protein expression of COX-2.Conclusion Proanthocyanidins has an anti-inflammatory effect in vivo which may be related to inhibition of protein expression of COX-2 and downregutation of PGE2 biosynthesis.

This paper introduces the job descriptions and competence of case managers, the development of cancer case management domestic and overseas; so as to offer reference for the domestic implementation of cancer case management. Consequently realize the comprehensive rehabilitation of physical, psychological, social and spiritual function for cancer patients.
5.Hemostasis effect comparison between artificial pressing and Perclose vascular closure device on femoral artery

Objective:To observe hemostoisis efficacy and safety of Perclose vascular closure device on femoral artery in transfemoral pathway radiofrequency ablation (RFA) of supraventricular tachycardia (SVT) .Methods :A total of 199 patients undergoing transfemoral RFA of SVT were divided into Perclose vascular closure group (Perclose group ,n=98) and artificial pressing group (n=101) .Immediate success rate ,hemostasis time ,braking time and in-cidence rates of postoperative complications were observed between two groups .Results:There was no significant difference in immediate success rate of hemostasis (98% vs .100% ) between Perclose group and artificial pressing group , P> 0.05. Compared with artificial pressing group , there were significant reductions in hemostasis time [ (18.9 ± 9.1) min vs .(2.7 ± 0.7) min] ,braking time [ (21.6 ± 3.4) min vs .(6.3 ± 2.4) min] and total inci-dence rate of complications (16.8% vs .4.1% ) in Perclose group , P<0.05 or <0.01. Complications:incidence rate of vagal reflex in Perclose group was significantly lower than that of artificial pressing group (0 vs .7.9% ,P<0.05) ,other incidence rates of complications between two groups were no significant difference ( P> 0.05 all) . Conclusion:Perclose vascular closure device could shorten the duration of hemostasis and braking ,and reduce the complications .It could be the preferred choice for femoral artery hemostasis .
6.Research on measuring the velocity and displacement of the coxa and knee based on video image processing.

Based on repeated experiments as well as continuous researching and improving, an efficient scheme to measure velocity and displacement of the coxa and knee movements based on video image processing technique is presented in this paper. The scheme performed precise and real-time quantitative measurements of 2D velocity or displacement of the coxa and knee using a video camera mounted on one side of the healing and training beds. The beds were based on simplified pinhole projection model. In addition, we used a special-designed auxiliary calibration target, composed by 24 circle points uniformly located on two concentric circles and two straight rods which can rotate freely along the concentric center within the vertical plane, to do the measurements. Experiments carried out in our laboratory showed that the proposed scheme could basically satisfy the requirements about precision and processing speed of such kind of system, and would be very suitable to be applied to smart evaluation/training and healing system for muscles/balance function disability as an advanced and intuitional helping method.

7.The effect of radiofrequency ablation with sublethal temperature on the stem cells of hepatocellular carcinoma

Objective To study the effect of radiofrequency ablation (RFA) with sublethal temperature on the production of liver cancer stem cells (LCSCs) and on the expression of LCSCs-related transcriptional factors.Methods Mouse hepl-6 hepatoma cell line and clinical samples of patients with hepatocellular carcinoma (HCC) were used to test the expressions of LCSCs-related markers and transcriptional factors.Results Different temperatures were used to stimulate Hep1-6 cells,and it was proved that the temperature of 45℃ was a sublethal temperature that could not induce cell death.Flow cytometry testing showed that treatment with 45℃ could obviously increase CD13+,CD44+,CD90 and CD133+ Hep1-6 cells,suggesting that treatment with 45℃ could increase the production of above mentioned types of LCSCs in hep1-6 cells.Real-time quantitative polymerase chain reaction (RT-qPCR) assay indicated that the temperature of 45℃could cause significant increase in CD13,CD90 and CD133 mRNA.In all 5 HCC patients,CD13 mRNA in the recurrent HCC lesions was remarkably increased,CD133 mRNA was increased in 4 patients with recurrent HCC,and CD90 mRNA was increased in only one patient with recurrent HCC.Flow cytometry testing revealed that CD13+ LCSCs were strikingly increased in 4 recurrent HCC patients,while CD133+LCSC was increased in only one patient,suggesting that more close correlation existed between the increase of CD13+ LCSCs and the temperature of 45℃.RT-qPCR assay showed that in 4 recurrent HCC patients with increased CD13+ LCSC,the Sox2 and Stat2 among 13 LCSCs-related transcriptional factors were obviously increased.Flow cytometry testing showed that 45℃ treatment also increased the expression of Sox2 and Stat1 mRNA in Hep1-6 cells.Finally,Sox2 and Stat1 could be knockdown by siRNAs,indicating that both Sox2 and Stat1 transcriptional factors were involved in 45℃-induced production of CD13+ LCSCs in Hep1-6 cells.Conclusion In RFA therapy,the use of sublethal temperature of 45℃ can increase CD13+LCSCs,which is related to the promotion of Sox2 and Stat1 expression.The results of this study can be used for reference in the research of liver cancer recurrence.
8.Application of KDIGO classifcation of chronic kidney disease for analyzing the prevalence of kidney disease and other vascular diseases in 1645 type 2 diabetic patients

Objective To analyze the prevalence,risk factors of kidney disease in type 2 diabetic patients with KDIGO classification of chronic kidney disease,also to study cardiovascular and cerebrovascular diseases and death in these patients,so as to investigate the significance of the KDIGO classification system.Methods One thousand six hundred and forty-five type 2 diabetic patients who were in hospitalization from June 2008 to December 2012 were grouped according to the KDIGO classification of chronic kidney disease and the incidence of vascular disease was analyzed based on the classification.Clinical features were compared between patients with or without kidney disease.The risk factors of kidney disease and the death of diabetic patients were also investigated.Results There were 915 male and 730 female,aged a median (57.86±12.54) years with (6.35±6.30) years duration of diabetes mellitus among the 1645 cases,and 37.2% of patients had concomitant kidney disease.According to the classi fi cation of CKD,patients in CKD group 3a,group 3b and CKD group 4-5 accounted for 5.7%,3.5% and 7.6%,while 33.4% of patients had proteinuria,among which 19.5% with microalbuminuria,13.5% with macroalbuminuria.On complications,patients with hypertension accounted for 49.5%,hyperlipidemia 67.7%,diabetic retinopathy 27.4%,cardiovascular and cerebrovascular diseases 18.5% (coronary artery disease 16.5%,cerebrovascular diseases 8.8%).Statistical difference was detected in the incidence of diabetic retinopathy,coronary artery disease and cerebrovascular diseases between CKD group 3a and 3b (P < 0.05).The duration of diabetes,concomitant hypertention especially with elevated systolic blood pressure,diabetic retinopathy and hyperuricemia were the independent risk factors for type 2 diabetic patients with kidney disease.Age,Scr,complicating cardiovascular and cerebrovascular diseases and advanced CKD stage were the independent risk factors for the death of type 2 diabetic patients with kidney disease.Conclusion KDIGO classification of chronic kidney disease enables better staging of kidney diseases in diabetic patients for management and prognosis.Diabetic patients have a higher prevalence of renal diseases and cardiovascular and cerebrovascular events than the general population.Early control of factors such as blood pressure and serum uric acid can delay the progression of kidney disease,and the predictive role of diabetic retinopathy should be emphasized.
9.Research on video measuring method of knee joint angle based on cross ratio invariability.

With an un-calibrated camera a new method is proposed in this paper to measure dynamically the bending angle of knee-joint based on cross ratio invariability. Firstly, by video image processing technique, we detected 25 circular objectives distributed in the aided measurement tool and determined their geometric centers in the video frame respectively. Then we further computed the extending direction of thigh and shank by detecting position of two rectangular targets tied on them. Finally, using of the principle of cross ratio invariability in Projection Geometry, we could realize real-time dynamic measurement of the bending angle of knee-joint based on the method. Experiments have proved that the measurement precision of the introduced approach could satisfy basic requirements of clinical applications.

10.Efficacy and safety of bone cement injection via unipedicular and bipedicular approaches in the treatment of osteoporotic vertebral compression fractures: a Meta-analysis

BACKGROUND: In the treatment of osteoporotic vertebral compression fractures with percutaneous vertebroplasty, the efficacy and safety of bone cement injection by unipedicular and bipedicular approaches are still controversial. Some studies suggest that bone cement injection via unipedicular approach can shorten operation time and reduce postoperative complications, while the other studies suggest that bone cement injection via bipedicular approach can make bone cement distribute more evenly in the vertebral body and relieve pain better. OBJECTIVE: To systematically assess the efficacy and safety of percutaneous vertebroplasty via unipedicular versus bipedicular approach in the treatment of osteoporotic vertebral compression fractures. METHODS: Randomized controlled trials about unipedicular versus bipedicular percutaneous vertebroplasty in the treatment of osteoporotic vertebral compression fracture published before September 18 t h, 2018 were retrieved in the PubMed, Cochrane library, Embase, CNKI, VIP, WanFang data and CBM. Two researchers independently screened all the literatures, carried out data extraction and used improved Jadad to evaluate the methodological quality of the included studies. Meta-analysis using Revam 5.3 was conducted. Egger's test was utilized to evaluate the publication bias. RESULTS AND CONCLUSION: Totally 14 randomized controlled trials including 900 cases were eventually included, 452 cases in unipedicular approach group and 448 cases in bipedicular approach group. The Meta-analysis results showed that compared with the bipedicular approach, the unipedicular approach required shorter operation time [weighted mean difference (WMD) =-16.59, 95% confidence interval (CI) (-19.25, -13.94), P < 0.001], smaller amount of bone cement injected [WMD=-1.27, 95% CI (-1.64, -0.89), P < 0.001], and had lower incidence of cement leakage [relative risk=0.70, 95% CI (0.53, 0.92), P =0.01]. There were no significant differences in short-and long-term Visual Analogue Scale scores, short-and long-term Oswestry Disability Index scores, and the postoperative incidence of adjacent vertebral fractures between the two groups (P> 0.05). Overall, bone cement injection both via bipedicular and unipedicular approaches can lead to a significant improvement in pain relief and living quality of osteoporotic vertebral compression fracture patients, but bone cement injection via unipedicular approach can shorten operation time, reduce cement volume and lower the incidence of cement leakage compared with the bipedicular approach.
