1.Effects of Salvianolate Injection on the Levels of Serum L-1βand IL-18 in Acute Exacerbated Chronic Obstructive Pulmonary Disease Patients
Journal of Zhejiang Chinese Medical University 2013;(11):1312-1314
[Objective] To investigate the effect of salvianolate injection on the levels of Serum interleukin-1β(IL-1β)and interleukin-18(IL-18) in acute exacerbated chronic obstructive pulmonary disease(AECOPD) patients. [Methods] Fifty-eight patients with acute exacerbated COPD from Respiratory Dep. of thrid Hospital Affiliated to Zhejiang Chinese Medical University, were randomly assigned to the treatment group(28 cases) and the control group(30 cases). Routine therapies for COPD were given to patients in the control group, while 20mg salvianolate injection was given to those in the treatment group by intravenous dripping, once daily for total 10 days. The levels of IL-18, IL-1β and blood gas analysis were evaluated before and 10 days after therapy. The levels of the cytokins were compared with 30 normal controls. [Result] There were no significant differences in the value of IL-1β,IL-18,arterial partial pressure of carbon dioxide(PaCO2),arterial partial pressure of oxygen(PaO2) before treatment between two groups. Compared with the blank control group, the levels of serum IL-1β,IL-18 in patient with AECOPD were increased significantly( P<0.01). When compared with before treatment and with the control group, the levels of IL-1β,IL-18,PaCO2 decreased more significantly in the treatment group after treatment. The PaO2 in treatment group was higher than in control group. There were significant differences between two groups.[Conclusions] Salvianolate injection can reduce the secretion IL-1β, IL-18 in patient with AECOPD, which maybe through certain anti-inflammatory effects to improve clinical efficacy of COPD pa-tients.
2.Effect of glutamine on p38MAPK and IL-8 expression in PBMC of patients with chronic obstructive pulmonary disease
Chongqing Medicine 2016;45(19):2593-2595,2605
Objective To investigate the effects of glutamine (Gln) on the p38MAPK activity and IL‐8 expression in periph‐eral blood mononuclear cells(PBMC) of the patients with chronic obstructive pulmonary disease(COPD) .Methods Thirty‐two pa‐tients with acute exacerbation of COPD (AECOPD ) were chosen as the research subjects(AECOPD group) .Then the patients con‐verting to the stable stage of COPD after treatment were set as the SCOPD group .PBMC of each group were extracted and conduc‐ted the Gln intervention .At the same time 16 healthy people were selected as the healthy control group .The real‐time PCR(RT‐PCR) was used to detect the levels of p38MAPK and IL‐8 expression in PBMC among the five groups .Results (1)The expressions of p38MAPK and IL‐8 in the AECOPD blank control group and the SCOPD blank control group were both higher than those in the healthy control group (P<0 .01 or P<0 .05) ,while the expression levels of p38MAPK and IL‐8 in patients with AECOPD was higher than those of patients with SCOPD ,the difference was statistically significant (P< 0 .05) .(2)The expression levels of p38MAPK and IL‐8 in the AECOPD Gln group were lower than those in the AECOPD blank control group (P<0 .01);the expres‐sion levels of p38MAPK and IL‐8 in the SCOPD Gln group were lower than those in the SCOPD blank control group (P<0 .05) . Conclusion Gln could inhibit the p38MAPK pathway activation in inflammatory cells of COPD patients and reduce the expression level of IL‐8 .

BACKGROUND:Low-temperature rapid prototyping technology is a new kind of rapid prototyping technology, and it is rapidly used in the preparation of bone tissue engineering scaffolds because it can make scaffold forming control able and can keep the biological activity of the materials, also can easily realize the scaffold with porous of three-dimensional structure and other advantages.
OBJECTIVE:To investigate the preparation process of polyethylene glycol-modified polylactic acid-glycolic acid/nano-hydroxyapatite (PLGA-PEG/n-HA) using the low-temperature rapid prototyping, and to test its performance.
METHODS:PLGA-PEG/n-HA and PLGA/n-HA were prepared by low-temperature rapid prototyping equipment. Under an electron microscopy, we observed ultra-structure of the scaffolds. Immersion (ethanol) method was used to test the porosity, and electronic testing machine was used to determine the material mechanical properties. Then these two kinds of scaffolds with rat osteoblasts were cultured in vitro, the cel adhesion rate was detected by precipitation method after 12 hours, and cel counting kit-8 method was used to determine the cel proliferation at culture days 1, 3, 5, 7, 9, 12.
RESULTS AND CONCLUSION:Both of the two scaffolds had ideal aperture range and high porosity. But the aperture range of PLGA-PEG/n-HA scaffolds had large fluctuations, and the average aperture was smal er than that of PLGA/n-HA. Some pores were closed up. The cel adhesion rate and the cel growth curve of PLGA-PEG/n-HA was better than that of PLGA/n-HA (P<0.05), but the mechanical properties were less than PLGA/n-HA (P<0.05). The results showed the PLGA-PEG/n-HA scaffolds had good cel compatibility.
