Objective To examine the effects of different compositions of metabolic syndrome [Overweight and (or) obesity, hyperglycemia, hypertension, dyslipidemia] on chronic kidney disease. Methods A total of 1552 health data were collected from the survey of chronic kidney diseases among Uygur adults in Moyu country in Xinjiang Uygur Autonomous Region and the relationship between metabolic syndrome and chronic kidney disease was analyzed by using SPSS 15.0 software package. Results Before and after adjusting of age and gender, the prevalence of metabolic syndrome was 14.18% and 14.45% (95% CI 14.30%?14.60%). The prevalence of albuminuria (7.27% vs 3.83%, χ2=5.42, P=0.02), reduced estimated glomerular filtration rate (9.55% vs 3.45%, χ2=16.96, P=0.00) and chronic kidney disease(13.64% vs 6.76%, χ2=12.52, P =0.00) increased in residents diagnosed as metabolic syndrome than those without metabolic syndrome. The prevalence of chronic kidney disease increased with the increasing number of metabolic syndrome elements. Conclusions The prevalence of chronic kidney disease is associated with the accumulation of metabolic syndrome compositions. Early intervention on metabolic risk factors may reduce the risk of chronic kidney disease.

AIM:To investigate the role of SDF-1? in migrating of bone marrow stromal cells to the injured areas. METHODS:Ischemic brain lesion model was created in rats by permanent middle cerebral artery occlusion (MCAO). 48 SD rats were divided randomly into 2 groups. Group 1:phosphate buffered saline (PBS 1 mL) for control (n=25); Group 2:BMSCs (2?106) were injected intravenously at 24 h after MCAO (n=24). After propagated in BMSCs,Ad5/F35 GFP (green fluorescent protein) was infected to BMSCs. The expression of SDF-1? (stromal cell-derived factor-1?) mRNA in the penrumbral tissue was assayed by real-time quantitative PCR. The expression of CXCR4 on MSCs was detected by flow cytometry. Confocal microscopy was used to detect the GFP-labeled MSCs migration. RESULTS:Ad5/F35 GFP signals was observed in almost infected BMSCs. The expressions of SDF-1? mRNA in the thalamus and hippocampus of the ischemic brains were peaked at 3rd day after stroke, followed by a decrease at 14th day post-ischemia. The expression of SDF-1? mRNA in the cortex of the ischemic brains was peaked at 7th day post-ischemia,still at high level at 14th day post-ischemia. The median percentage of surface CXCR4 expression in BMSCs was 14%. GFP labeled BMSCs were detected in the origination of the middle cerebral artery (olfactory area) at 6 h,after 3 days in the prenumbra tissue such as thalamus, and in the cortex more labeled cells were found after 14 d post-ischemia.CONCLUSION:BMSCs can pass through the blood brain barrier of ischemic rats. Its mechanism might be associated with the expression of SDF-1? in the ischemic brain.

Objective To prove the remarkable inhibitive effect of multiple siRNAs targeting a proliferation-inducing ligand (APRIL) on the human colorectal cancer cell. Methods We constructed a multiple short hairpin RNA(shRNA) expression vector containing four shRNAs (pG4) as well as four single one (pGsh644,pGsh1451,pGsh1938,pGsh2231) against APRIL gene in SW480 cell, and then transfected them into the human colorectal cancer cell line by cationic liposome.Ultimately,SW480 were screened by EGFP to obtain expression cell lines.APRIL expression levels including mRNA and APRIL protein were detected after transfected with all different kinds of vectors. Results A multiple shRNA expression vector containing four shRNAs (pG4) and four single ones were successfully constructed.Four single vectors (pGsh644,pGsh1451,pGsh1938,pGsh2231) and the multiple siRNAs expression vector (pG4) all decreased the APRIL mRNA by 56.2%,49.5% ;50.9%,49.2% and 79.3%.And APRIL protein expression was also remarkably reduced, especially by multiple siRNAs expression vector(87.5%).Conclusion Multiple siRNAs expression vector produced a more significant knockdown effect of APRIL than the vectors containing only one APRIL shRNA.What we found suggested us using the vector containing multiple shRNA to silence the expression of APRIL might be exploited as a novel therapeutic strategy for tumors.
