1.Research progresses of regulatory T cells in asthma

Asthma is severely harmful to the health of the children with respiratory tract infection,but its etiology and pathogenesis is not yet entirely clear.Currently,it is considered that Th1/Th2 cell function imbalance is the immunological basis.Regulatory T cells (regulatory T cell,Treg) is one of the hot spots in recent years,regulating the immune system negatively.The decrease in the number of cells leads to immune suppression dysfunction,and creats the conditions for the development of the disease for asthma.Recent studies have found that Treg cells play an important role in the regulation of Th2 cell activation,which not only completes the traditional theories of Th1/Th2,but also provides a new method for preventing and controlling asthma by clinical use of Treg cells.

5.Effects of NTI-tss splint in the treatment of myofascial pain dysfunction syndrome

20 patients with myofascial pain dysfunction syndrome(MPDS)were were treated by NTI-tss splint only during night for 2 months.After treatment,VAS decreased(P <0.01)and active maximum mouth opening degree increased(P <0.01).NTI-tss splint is ef-fective in the treatment of MPDS.Its side effect needs to be further observed.

At present, more and more researches have found that cell-mediated immunity,especially T lymphocyte response plays an important role in the control of Epstein-Barr virus infection.
7.Effect of magnesium on nigral dopaminergic neurons in rats with Parkinson's disease

Objective To observe the effect and mechanism of magnesium on nigral dopaminergic neurons in rats with Parkinson's disease(PD). Methods The PD rats were prepared by unilateral injection with 6-hydroxydopamine(6-OHDA) in the nigra and then divided into magnesium sulfate group,Madopar group,mixed group(magnesium sulfate+Madopar) and control (normal saline) group,and recived corresponding therapy by gastric perfusion for 28 d.The behavior change was observed. At the damage side,the number of tyrosine hydroxylase(TH) positive neurons in nigra was detected by immunohistochemistry; the activities of superoxide dismatase(SOD),glutathione peroxidase (GSH-Px) and the level of malondialdehyde(MDA) in striatum were measured by biochemistry method,the expression of caspase-3 mRNA and the protease of nuclear factor(NF)-?B P65 in nigra were detected by RT-PCR and Western Blot. 8.Suppression of Growth and Tumorigcnicity in Human Gastric Cancer Cells by the Introduction of Exogenous p53 Gene

The p53 gene is one of the most common targets for genetic abnormalities in human tumors. Restoring wild - type p53 gene (wt-p53) into cancer cells which have p53 deletion is a strategy in cancer gene therapy. In order to explore the feasibility of this hypothesis, we selected a gastric cancer cell line BGC823 which was confirmed having deletion of chromosome 17pl3 and decreased expression level of p53 mRNA . We transfected construct pC53SN3 containing wt - p53 into BGC823 cell line with lipofectin mediated gene transferration, and G418 resistant colonies were characterized by using analysis of PCR, Southern blot hybridization, Northern blot hybridization and Western blot hybridization. These data showed that exogenous wt-p53 had successfully transferred into BGC823 cells and obtained high expression. The cell growth rates in regular medium and soft agar were inhibited from 30 to 40 percent in the BGC823 cells transfected with wt - p53. The tumorigenicity in nude mice showed that one of four mice failed to form tumor and three of them delayed to form tumor from 7 to 14 days comparing with monk and parent BGC823 cells. These results suggested that exogenous wt -p53 could suppress the growth ability and tumorigenicity of human gastric cancer cells. The method of using lipofectin mediated wt-p53 gene transfection may have a potentially therapeutic effect on human gastric cancer.
9.Effect of curcumine in regulating the balance between regulatory T cells/helper T lymphocyte 17 via SOCS3 in inflammatory bowel disease in immature rat

Objective To determine the therapeutic efficacy of curcumine on inflammatory bowel disease(IBD)and its underlying mechanisms.Methods The rat ulcerative colitis model was developed by using 30 g/L dextran sulfate sodium.Twenty-four young rats were divided into 4 groups:normal group,model group,curcumin 100 mg/kg group(curcumin 100 group),and curcumin 300 mg/kg group(curcumin 300 group).Curcumin was given through gastric gavage once a day for 7 days.General condition,disease activity index(DAI)and histopathological score(HPS)were evaluated.The proportions of CD4+ CD25+ Foxp3+ regulatory T cells(Treg)and CD4+ IL-17+ helper T lymphocyte 17(Th17)in CD4+ T cells in rats,and spleen cells were calculated by using flow cytometry.Immunohistochemical method was performed to determine the level of SOCS3 in colon tissues.Results After curcumin administered through gastric gavage for 5 days,compared with the DAI in the model group[(7.50±0.32)scores],HPS in the curcumin 100 group[(4.00±1.10)scores] and the curcumin 300 group [(5.00±0.89)scores] significantly reduced,and the difference was significant(F=12.469,P<0.05).After curcumin administered through gastric gavage for 7 days,compared with the HPS in the model group[(2.69±0.70)scores],histological score in the curcumin 100 group[(1.94±0.68)scores],the curcumin 300 group [(2.00±0.63)scores] and the normal group[(0.50±0.05)scores] significantly reduced,and the difference was significant(F=33.282,P<0.05).Compared with the model group[(0.015 1±0.000 7)D],the expression of SOCS3 in the curcumin 100 group[(0.002 0±0.000 5)D],the curcumin 300 group [(0.002 6±0.006 0)D] and the normal group[(0.002 2±0.001 0)D] was significantly inhibited(F=382.270,P<0.05),but there was no difference among the former 3 groups(F=0.828,P>0.05).Compared with the model group[(6.5±1.5)%],the propotion of CD4+ CD25+ Foxp3+ Treg cells in CD4+ T cells in the curcumin 100 group[(9.9±1.0)%],the curcumin 300 group [(9.3±0.7)%] and the normal group[(9.6±1.1)%]was obviously upregulated(F=16.635,P<0.05),but there was no difference among the former 3 group(F=1.564,P>0.05).Compared with the model group[(3.5±1.4)%],the propotion of CD4+ IL-17+ Th17 cells in CD4+ T cells in the curcumin 100 group[(2.0±0.9)%],the curcumin 300 group [(1.2±0.6)%] and the normal group[(2.0±0.9)%] was downregulated(F=5.155,P<0.05),but there was no difference among the former 3 group(F=2.073,P>0.05).There was no obvious difference between the curcumin 100 group and the curcumin 300 group in these indicators respectively(all P>0.05).Conclusions Curcumin probably attenuates IBD severity,reduces inflammation and regulates the balance of Treg/Th17 through the inhibition of SOCS3 expression.
10.Diagnostic significance of γ-glutamyl transpeptidase in infants cholestasis of different causes

Infants cholestasis is a common disorder of hepatobiliary system which is caused by abnormal cholestasis of hepatocytes or substances accumulation in the liver, blood and extrahepatic tissue due to the disorder of the intrahepatic or extrahepatic bile ducts.Serum Gamma-glutamyl transpeptidase(GGT) was widely distributed in one side of the capillary bile duct and the whole bile duct system of hepatocytes.When hypersynthesis in the liver or disorder of cholorrhagia, serum GGT increased.This review summarized the changes of GGT cholestasis of different causes in infants and its significance in differential diagnosis, for better understand the disease and provide references for clinical practice.
