1.Determination of 7 Residual Organic Solvents in Shengxuening Tablets by Headspace Gas Chromatography

Objective:To establish a method for the determination of 7 residual organic solvents including acetone,2,3 -dimethyl-pentane,3-methylhexane,n-heptane,2,2-dimethylhexane,o-xylene and 2,4,6-trimethylpyridine in Shengxuening tablets. Methods: A headspace gas chromatography method was adopted. The determination was performed on a DB-5MS capillary column ( 30 m × 0. 25 mm,0. 25 μm) with programming temperature. Nitrogen was used as the carrier gas at the flow rate of 2. 5 ml·min-1 . The tem-perature of injector was 200℃,and a flame ionization detector was used as the temperature of 250℃. The containers of headspace in-jector were in equilibrium at 95℃ for 30min. DMF was used as the solvent and an external standard method was used for the determi-nation of the 7 residual solvents. The injection volume was 1 ml. Results:Under the chromatographic conditions, the 7 residual organic solvents could be well separated from each other . The concentration of each solvent showed a good linearity with the peak area within the investigated concentration range (r≥0. 994 4). The average recoveries were 98. 72%-99. 71% (RSD=0. 14%-0. 71%)(n=9). Conclusion:The established method is simple, rapid and sensitive, which can be used for the determination of multiple residual or-ganic solvents in Shengxuening tablets.

3.Investigation on high-arsenic water:a report of 12 counties in Shaanxi Province in 2006

Objective To explore the distribution of high-arsenic water in Shaanxi Province in order to provide theoretical basis for the control of arsenism.Methods The contents of arsenic of 4042 water sampled from 1898 nature villages in 12 counties were assessed using semi-quantitative reagent-knit,and the water samples of Shanyang County,the targeted area,was quantified using quantitative atomic fluorescent speemetry.Results The contents of arsenic of 6 water samples in 2 villages exceeded 0.05 mg/L,accounting for 0.15%(6/4042),were 0.46,0.53,0.63,0.69,0.74,1.53 mg/L,respestively,and located in Shanyang County.A crowd of 1146 exposed to hish asenic water.among whom 555 were children.Conclusions Arsenic in drink water in Shanyang County in Shaanxi Province is higher than the standard,to which a large number of people are exposed,water-improving and defluoridotion must be carried out as soon as possible,reduces harm of high arsenic.

Most of doctors paid special attention to surgical technique, rather than to some related issues of operation. Timing is crucial for ptosis surgery. Both cosmetic and function need to be considered for a success surgery, the evaluation of extraocular muscle disorder is impor- tant for surgical result and postoperative complications.
7.Changes of Plasma Somatostatin in Children with Septic Shock

Objective To explore the changes of plasma somatostatin(SST) in children with septic shock.Methods The level of plasma SST in children with septic shock (test group,n=21) on an empty stomach at shock stage,blood pressure and heart rate recovery stage,recovery stage(at that time clinical symptoms and signs disappeared,infection indicators such as blood routine and CRP returned to normal,about 6-12 days after admission) were detected by competive radioimmunassay,the level of SST in healthy children(healthy control group,n=25) on an empty stomach on morning was detected,too.The levels of plasma SST between septic shock concbined with paralytic ileus group and without paralytic ileus group were compared.Results 1.Level of plasma SST of test group at shock stage[(44.60?16.83) ng/L]was significantly lower than that of control group[(123.15?6.57) ng/L](t=-12.16 P0.05).The level of plasma SST of children with paralytic ileus [(28.10?7.0) ng/L] was significantly lower than that of children without paralytic ileus [(56.98?9.44) ng/L](t=-7.70 P
8.Biological Characteristics of Human Colon Cancer Cell Line CW-2 Stem Cells

Objective To isolate cancer stem cells from human colon cancer cell line CW-2,and observe the biological characteristics.Methods The percentage of CD44+EpCAM+ cancer stem cells which were isolated by multi-combination method was evaluated by flow-cytometry.The biological characteristics of the stem cells were analyzed by cell cycle analysis,in vitro invasion assay and in vivo tumorigenicity assay.Results The percentage of CD44+EpCAM+ cancer stem cells was 89.57%,and their capabilities of generation,invasion,tumorigenicity were higher than non-cancer stem cells(P 0.05).Conclusion CD44+EpCAM+ cancer stem cells showed low generation,high invasiveness,high tumorigenesis,and they could be isolated by multi-combination.
9.Effect of granulocyte colony-stimulating factor on murine thymic emigration and subsets reconstitution after a sublethal dose of irradiation

Objective To investigate the effects of recombinant human granulocyte colonystimulating factor(G-CSF) on murine thymic emigration and subsets reconstitution after a sublethal dose of irradiaton.Methods Female BALB/c mice were irradiated with a 6.0 Gy of γ-ray total-body irradiation and then randomly divided into GCSF group and control group.For mice in the GCSF group,recombinant human G-CSF 100 μg · kg-1 · d-1 was injected subcutaneously once daily for 14 continuous days and mice in the control group were given the same volume of phosphate buffered solution (PBS).At 7,14,21 and 28 days later,mice were killed and thymus mononuclear cell suspension were analyzed by flow cytometry for the percentage of the four stages of thymic CD4 -CD8 - double negative cells (DN1-4) and the CD4 + CD8 + double positive ( CD4 + CD8 + DP),CD4 + CD8 - single positive ( CD4 + SP),CD4 -CD8 + single positive cells (CD8 + SP).Real-time PCR was used for detection and quantitation of murine T cell receptor rearrangement excision circles(sjTRECs) of the thymic cells of 30 and 60 d after irradiation.Results The percentage of thymic DN1 cells in GCSF group was significantly higher than that of the control group 7 d after irradiation (t =9.59,P < 0.05 ).21 d later,the proportion of thymic DN3 and DN4 cells were higher than those of the control group ( t =16.37,7.6,P < 0.05 ).The percentage of thymic CD4 + CD8 + DP cells decreased 7 d after irradiation,increased at 14 d,decreased again at 21 days,and then got a permanent recover.The percentage of thymic CD4 + CD8 + DP cells in the GCSF group recovered to normal and was significantly higher than that of the control group 28 days after irradiation (t =12.22,P< 0.05).The percentage of thymic CD8 + SP cells of the GCSF group was significantly higher than that of the control group 21 d after irradiation ( t =3.77,P < 0.05 ),while G-CSF had no obvious influence on the percentage of the thymic CD4 + SP cells.The sjTRECs copies in the GCSF group was significantly higher than that of the control group 30 d after irradiation ( t =5.95,P < 0.01 ),which disappeared 60 d later.Conclusions G-CSF could promote the proliferation and differentiation of thymic DN and DP cells,enhance the recent thymic emigrants and accelerate central immunologic reconstitution after acute irradiation.
10.Mesenchymal stem cells protecting multiple organ failure

Objective The patients with lethal irradiation after sucessful hematopoietic stem cells transplan-tation had blood recovery, but did not avoid to died of multiple organ failure(MOF). To overcome the block, the article investigated mechanisms of mesenchymal stem cells (MSCs) protecting lethal radiated mice from multiple organ failure after haploid bone marrow cells transplantation. Method BALB/c mice irradiated with 8Gy60COγ-rays were randomly divided into two groups: MSCs group, infused MSCs labeled with cm-DiI and bone marrow monocytes of CB6F1 mice; Control group, only infused bone marrow monocytes; normal group, mice were infused cm-DiI marked MSCs without irradiation. The distribution of MSCs and the serous densities of Il-2, Il-10 and TNF-α in the recipients were observed after transplantation. Results MSCs collected in the bone marrow and the intes-tine in normal group at 15 d,in MSCs group MSCs enriched the different organs at 3,15 and 30 d. MSCs regulated down the secretion of IL-2 and TNF-α,and up the IL-10 density. Conclusions MSCs protected mice from multiple organ failure through above effects and may be open a new treatment strategy on acute radiation syndrome by stem cells.
