2.Purification of Dayuan Dropping Pills extract with macroporous resin

Objective To study the adsorption function of macroporous resin for purifying the extract of Dayuan Dropping Pills and to investigate the purification process of Dayuan Dropping Pills by macro-porous adsorption resin. Methods Taking the content and the extraction rate of baicalin as the main standard index, and the adsorption characters as well as the purification capacities of eight types of macroporous adsorption resins,such as NKA-12, HPD-600, HPD-500, HPD-300, HPD-100, D101, AB-8,and NKA-9, used to treat baicalin of Dayuan Dropping Pills were investigated preliminarily. Results The appropriate adsorption conditions were: the concentration of baicalin was 2.65 mg/mL, flow rate was 3 BV/h, the effect of desorption was satisfactory when 50% alcohol (5 BV) and water (4 BV) were used as eluant. Conclusion HPD-300 resin can be applied to refine the Dayuan Droppingp Pills.
3.Clinical Observations on Coiling-dragon Warm Needling at Huatuo Jiaji Points as Main Treatment for Ankylosing Spondylitis

ObjectiveTo investigate the clinical efficacy of coiling-dragon warm needling atHuatuo jiaji points plus Chinese herbal fumigation in treating ankylosing spondylitis (AS).MethodEighty AS patients were randomly allocated to treatment and control groups, 40 cases each. The treatment group received coiling-dragon warm needling at Huatuo jiaji points plus Chinese herbal fumigation and the control group, oral administration of salicylazosulfapyridine (SASP). An investigation was made of the Bath Ankylosing Spondylitis Disease Activity Index (BASDAI), the patients’ overall assessment, the Bath Ankylosing Spondylitis Functional Index (BASFI), the total number of swollen joints, the Bath Ankylosing Spondylitis Measurement Index (BASMI) and laboratory inflammatory indices: erythrocyte sedimentation rate (ESR), platelet (PLT) and C reactive protein (CRP).ResultThere was a statistically significant difference in the marked efficacy rate between the two groups (P<0.05). Clinical indices: disease activity index and functional index and laboratory indices: ESR, PLT and CRP improved somewhat inboth groups of patients after treatment (P<0.05). After two courses of treatment, the therapeutic effect was better in the treatment group and there was a statistically significant difference compared with the control group (P<0.05).ConclusionCoiling-dragon warm needling at Huatuo jiaji points plus Chinese herbal fumigation can effectively relieve the clinical symptoms and reduce inflammatory reactions in the active stage in AS patients. It is an effective way to treat AS.
4.The effects of angiotensin Ⅱ on the synthesis of albumin and type Ⅰ collagen in human hepatic cells

Objective To investigate the effects of angiotensin Ⅱ (AngⅡ) on the expression of albumin and the synthesis of type Ⅰ collagen in human normal hepatic cells. Methods HL-7702 cells (human normal hepatocyte) were cultured and divided into control group, Ang Ⅱ treated group, an AngⅡ+irbesartan (co-stimulated) group. The expressions of albumin and type Ⅰ collagen were detected by immunofluorescence and Western blotting, respectively. The mRNA level of type Ⅰ collagen was measured by real time-PCR(qRT-PCR). Results After stimulated with 10-7 mol/L Ang Ⅱ for 72 hours, the expression of albumin significantly decreased in Ang Ⅱ treated group compared with control group (0.85±0.11 vs 1. 41±0.23,P=0.000), while the mRNA expression increased in AngⅡ treated group compared with control group (1.00±0.08 vs 3.72±0.19,P=0.000). In costimulated group, however, the expression of albumin significantly increased (0.85 ± 0.11 vs 1.38 ±0.32,P=0.000),and mRNA expression (3. 72±0.19 vs 2.86±0.13,P=0.000) and synthesis of type Ⅰ collagen were reduced when compared with Ang Ⅱ treated group. Conclusions The reduction of albumin and elevated systhesis of type Ⅰ collagen in HL-7702 cells are induced via Ang Ⅱ AT1 receptor.
5.Separation and purification of total alkaloid from Rhizoma Coptidis by macroreticular adsorbent resin

Objective To study the technology for purification of total alkaloids from Rhizoma Coptidis by maeroreticular adsorbent resin.Methods LD605,D101,DA201,NKA-9,and AB-8 types of macroreticular adsorbent resins werre used to separate and purify the total alkaloid from Rhizoma Coptidis.The yields and purities of the products were compared as indexes.Results AB-8 Type macroreticular adsorbent resin had optimum adsorption and elution parameters with its dynamic saturated adsorption ratio up to 1.23 mg/g.After eluted with 2 BV of distilled water and 2 BV 40% ethanol,the yields of total alkaloid was 85%,and content was 80%.Conclusion The AB-8 type macroreticular resin showsa better comprehensive adsorption property and can be used to separate and purify the total alkaloids from Rhizoma Coptidis.
6.Sequence and Structural Analyses of the Complete Genome of Bovine Papillomavirus 2 Genotype Aks-01 Strain from Skin Samples of Cows in Southern Xinjiang, China.

To study the complete genomic sequence, genomic characteristics, and genetic variation of the bovine papillomavirus 2 genotype (BPV-2) Aks-01 strain at the molecular level, genotyping of this strain from the skin samples of cows in southern Xinjiang (China) was first detected by the polymerase chain reaction with FAP59/FAP64 primers. Based on the complete genome of the BPV-2 reference strain, specific primers and sequencing primers were designed, and the complete genome of the Aks-01 strain amplified and sequenced. Sequence analyses showed that genotyping of the Aks-01 strain belonged to BPV-2. The Aks-01 strain had the structural characteristics of BPV-2. The 7944-bp full-length genomic sequence of the Aks-01 strain was compiled using DNAStar™. The sequence of the Aks-01 strain had 98% similarity to the reference strain from GenBank. The Aks-01 strain was most closely related to BPV-1 and BPV-13. BPV-2, BPV-1 and BPV-13 were grouped within the genus Deltapapillomavirus. The Aks-01 strain is the first BPV-2 strain reported in southern Xinjiang.

7.Clinical study of ultrasound evaluation of fiberoptic bronchoscope guided tracheotomy

Objective:To explore the clinical effect of ultrasound evaluation of fiberoptic bronchoscope (FB) guided tracheotomy, which can provide help for difficult tracheotomy and new operators.Methods:The operating protocol was standardized for ultrasound evaluation of FB guided tracheotomy. Ten patients with difficult tracheotomy admitted to the department of critical care medicine of Donge Hospital from October 2019 to January 2020 were enrolled. According to this protocol, FB guided tracheotomy was performed under the ultrasound evaluation, and the amount of blood loss, operation time and related complications during procedures were collected.Results:The preparation of supplies, personnel, patients and the operation, the process of FB guided tracheotomy under the ultrasound evaluation were standardized. When tracheotomy was preformed for patients with difficult tracheotomy, it was necessary to use ultrasound first to evaluate the neck condition and vascular disorientation of patients, and the tracheotomy plan (tracheotomy site, incision size, and incision depth) was designed, and then the tracheotomy process was monitored under the guidance of FB. Among the 10 patients with difficult tracheotomy, 6 were male and 4 were female; body mass index was (32.2±1.4) kg/m 2. Tracheotomy was successfully completed within 10 minutes in all the 10 patients, with less than 5 mL blood loss, and no complications occurred. Conclusion:Ultrasound evaluation of FB guided tracheotomy can improve the clinical operations and ensure patient safety.
