1.Effects of Astragalus polysaccharide on gene expression profiles in islets of NOD mice with microarray technique

Objective To further investigate the molecular mechanism underlying the preventive effect of Astragalus polysaccharide (APS) on type 1 diabetes mellitus (DM) in NOD mice. Methods In order to explore the gene expression profiles of NOD mice′s islet cells treated with APS, cDNA microarray technique was applied. Results In APS group 5.47% gene expressions were obviously changed, 28 genes up-regulated, 35 genes down-regulated, and of which, 17 genes were functionally related to immunity. Conclusion The preventive effect of APS on type 1 diabetes in NOD mice is probably related to the action of APS in correcting the imbalance of Th 1/Th 2 cytokines.
2.Effects of Nimodipine on Peripheral Facial Paralysis and Hearing in Mimetic Convulsion Patients after Mi-crovascular Decompression

OBJECTIVE:To observe the effects of nimodipine on peripheral facial paralysis and hearing in mimetic convulsion patients after microvascular decompression. METHODS:162 mimetic convulsion patients underwent microvascular decompression were enrolled in this study,and then randomly divided into control group and observation group,81 cases in each group. Control group was given routine microvascular decompression,and observation group was additionally given intravenous pump of Nimodip-ine injection 40 mg,qd,2 mg/h for consecutive 3 days after operation,and then given Nimodipine tablet 40 mg,qd,for 10 days. The incidence of peripheral facial paralysis and hearing disorder,onset time,duration time and severity of facial paralysis were ob-served in 2 groups. RESULTS:The incidence of peripheral facial paralysis and hearing disorder were 8.6% and 6.2% in observa-tion group,which were significantly lower than 14.8%and 11.1%of control group,with statistical significance(P<0.05). The on-set and duration time of peripheral facial paralysis was(15.32±3.15)d and(36.52±5.84)d in observation group,which were sig-nificantly later and lower than(12.21±2.16)d and(44.73±6.57)d of control group,with statistical significance(P<0.05). Ac-cording to House-Brackman classification method,the incidence of Ⅲ、Ⅳ facial paralysis was lower in control group than that in observation group,with statistical significance (P<0.05). CONCLUSIONS:Nimodipine can significantly decrease the incidence of peripheral facial paralysis and hearing disorder in mimetic convulsion patients after microvascular decompression,and contrib-utes to the recovery of injured nerve function.
3.Efficacy of tea polyphenols in inhibiting inflammatory damage to islet beta cell of rats with metabolic syndrome

A rat model of metabolic syndrome was induced by high glucose plus high fat diet and treated by tea polyphenols for 10 weeks.The results showed that the levels of fasting blood glucose,triglyceride,cholesterol,low-density lipoprotein cholesterol,and free fatty acid in tea polyphenols treatment group were significantly lower than those in control group(all P<0.05),along with decreased TNF-α,IFN-γ,iNOS mRNA and protein expressions,and IL-1β protein expression in pancreatictissue (all P<0.05).Under electron microscope,more secretory granules in islet beta cells and impoveddisorganization of cellular organ were shown in tea polyphenols treatment group compared with metabolic syndrome group.The results suggest that tea polyphenols are able to protect islet β cells against oxidative damage via inhibiting the production of inflammatory cytokines.
4.Research about shorten preoperative fasting time for patients with digital cerebral angiography

Objective To know the feasibiliy and the effects of shorten preoperative fasting time for patients with digital cerebral angiography. Methods Divided 110 patients with digital cerebral angiogra-phy into the observation group and the control group according to their sequence enter the hospital. Routine fasting method was used in the control group, shorten fasting time was used in the observation group, com-pared the incidence rate of fasting-related complications between the two groups. Results The incidence rote of belly discomfort, thirstily, strvation and hypoglyeemic during the digital cerebral angiogrsphy in the con-trol group was significant higher than those of in the observation group, while there were no significant difference about the incidence rate of postoperative complication between the two groups. Conclusions For patients with digital cerebral angiography can shorten the preoperative fasting time properly, which can avoid certain complieaitons.
5.The preliminary study on hemodynamics change of brain before and after vertebrobasilar angioplasty and stenting in the patients with intracal stenosis

Objective To evaluate the hemodynamics change of brain before and after vertebrobasilar angioplasty and stenting in the patients with intracal stenosis.Methods All the indexes were compared among thirty-eight patients with intracal arterial stenosis,including mean peak flow velocity,systolic peak flow velocity,and resistance index of hemodynamics ; Cerebral blood volume ( CBV),cerebral blood flow (CBF)and middle time to peak(MTT) before and after the operation.Results After vertebro-basilar angioplasty and stenting in the 38 patients,the Vs of PCA were significantly lower in the affected side [ (226 ±27)cm/s vs (135 ± 18)cm/s; t =21.74,P <0.01 ].Significant difference was found between pro-and post stenting of rMTT and rCBF in the blood territories of stenotic cerebrovascular ( t =3.987,P =0.001 )and( t =-2.027,P =0.029),There was no significant difference between pro-and post stenting of rCBV in the blood territories of stenotic cerebrovascular ( t =-0.901,P =0.329).Conclusions Transcranial Doppler and CT perfusion can be used to evaluate hemodynamic changes before and after pereutaneous transluminal angioplasty and stenting.
6.Effects of monoclonal antibody of insulin-like growth factor-Ⅰ receptor on proliferation and apoptosis of papillary thyroid carcinoma cells

Objective To investigate the role of monoclonal antibody of insulin-like growth factor Ⅰ receptor (IGF-ⅠR) in the proliferation and apoptosis of papillary thyroid carcinoma cell line CGTH W-3, and the effect of IGF-ⅠR monoclonal antibody on papillary thyroid carcinoma cells and normal follicular cells in primary culture. Methods MTT and TUNEL method were used to determine the proliferation and apoptosis of cells respectively after IGF-ⅠR monoclonal antibody intervention. Results The inhibitory effect on cell proliferation was enhanced with the increasing IGF-ⅠR monoclonal antibody concentration and duration of action. IGF-ⅠR monoclonal antibody induced apoptosis of CGTH W-3 cells remarkably even in the concentration of 50 nmol/L. Objective To investigate the effect of astragalus polysaccharide(APS)on expression of interleukin-4(IL-4)and interferon-γ(IFN-γ)in non-obese diabetic(NOD)mice.Methods 20 NOD mice were randomly divided into the APS group and control group with 10 animals in each group.The mice of the APS group took APS 2 g/kg/d orally for two months.The animals of the control group took same quantity saline.The incidence rate of type 1 diabetes mellitus of two groups was calculated,and pancreatic histopathology was observed with microscope,and expression of IL-4 and IFN-γ were examined with RT-PCR for both APS group and control group.Results The incidence rate of type 1 diabetes mellitus of the APS group was lower than the control group,and histopathology of islets were more protected,expression of IFN-γ was lower and expression of IL-4 was higher in the APS group.Conclusion APS may correct the imbalance of cytokines.

9.Effects of astragalus polysaccharides on lipid metabolism and PPAR-α gene expression in myocardium of diabetic hamsters

Objective This article was designed to observe the effects of astragalus polysaccharides (APS) on glucose and lipid metabolism, and on expressions of proxisome proliferator activated receptors-α (PPAR-α) and its downstream genes in diabetic hamsters cardiomyopathy. Methods Forty-five hamsters were divided into 3 groups randomly: normal control group (15 normal hamsters), diabetic control group [15 streptozotocin (STZ)-induced diabetic hamsters], and astragalus polysaccharides (APS)-therapy group (15 STZ-induced diabetic hamsters administered with APS 2 g/kg per day orally for 10 weeks). The levels of insulin, C-peptide, myocardial enzymes, glycosylated serum protein (GSP) and lipoprotein of all hamsters were measured. The ultrastructure of myocardium was studied, and the gene and protein expressions of PPAR-α, FATP and ACS were also detected by fluorescent quantitative RT-PCR and Western blot. Results It was shown that Compared with DM group, the levels of GSP, myocardial enzymes and lipoprotein of hamsters in APS-therapy group were lower, the myocardial ultrastructure of hamsters in APS-therapy group was well-protected, and the gene and protein expression of PPAR-α, FATP and ACS of hamsters in APS-therapy group were higher. Conclusions APS is partly effective in treating diabetic cardiomyopathy.

Objective To investigate the relationship between microalbuminuria (MAU) and insulin resistance syndrome. Methods A total of 240 patients(130 of them with hypertension) were stratified by oral glucose tolerance status 〔normal glucose tolerance(NGT), impaired glucose tolerance(IGT), and type 2 diabetes mellitus(DM)〕.
