1.Treatment of malignant gliomas with stem cells mediated oncolytic adenovirus

Oncolytic adenoviruses are genetically altered to replicate within tumor cella while exhibiting a high degree of infectivity,good safety and specificity.The use of oncolytic adenoviruses to treat malignant brain tumors has been proved to be a promising mode of cancer therapy.Local injection of adenoviral vectors fails to reach scattered infiltrative tumor cells within the brain parenchyma and the therapeutic effect of intracereberal injections of oncolytic adenoviruses is only seen in the vicinity of the injection site.To remedy this and prevent possible tumor recurrence,stem cells have been explored as vehicles for gene therapy in brain tumors given that they possess an intrinsic tropism for brain glioma.
2.Influence of amitriptyline on GLAST in spinal cord of rat in neuropathic pain

Aim To investigate influence of amitriptyline on glutamate-aspartate transporter(GLAST)in spinal cord of rats after spared nerve injury.Methods 60 male SD rats were randomly divided into 4 groups with 15 rats in each:control(A),SNI(B),amitriptyline(C),SNI+amitriptyline(D).Respectively 4 groups were treated with intraperitoneal injections of 0.2 ml saline(A and B),10 mg?kg-1 amitriptyline(C and D),bid.The L3~L6 segments of the spinal cord were isolated in 1,3 and 5 days after surgery.Expression of GLAST was determined by western blot and semiquantitative RT-PCR.Also,changes of mechanical withdrawal threshold(MWT)were measured.Results Compared to control,a markedly decreased MWT was showed in group B and the expression of GLAST at both protein and mRNA level increased firstly and decreased later.There were no changes of MWT in group C,but the expression of GLAST increased gradually.Rat MWT in group D did not change any more at the third days after surgery.In addition,the expression of GLAST in group D was higher than control group and kept stable.Conclusions Amitriptyline could increase the expression of GLAST,which may be one of its mechanisms in the treatment of neuropathic pain.

6.The superposition effect of moxibustion and ear acupoint pressing method in Sodium Hyaluronate Eye Drops treatment of dry eye

Objective To observe the effect of moxibustion and ear acupoint pressing method in treating dry eye syndrome.Methods Will meet the inclusion criteria of 63 patients were randomly divided into a treatment group (n=31) , treated with moxibustion and ear acupoint pressing method, and a control group (n=32) who were given with sodium hyaluronate eye drops. After 4 weeks' treatment, clinic symptom,break-up time of tear film(BUT), fluorescence staining(FL) and tear meniscus height(TMH) were measured and compared respectively.Results After treatment, the BUT (8.28 ± 1.73 svs.5.00 ± 1.51 s,t=11.948) in the treatment group was longer than the control group (P<0.01), FL score (1.00 ± 0.90vs.1.85 ± 1.45,t=4.057) in the treatment group were lower than the control group; TMH(0.28 ± 0.06 mm vs.0.20 ± 0.06 mm,t=6.768) in the treatment group were higher than the control group (P<0.01). After treatment, dry sense (1.61 ± 0.67 vs.2.60 ± 0.89,t=6.053), foreign body(1.00 ± 0.45 vs.1.85 ± 1.45,t=10.460), burning(0. 39 ± 0.67 vs.1.59 ± 0.87, t=6.142), visual fatigue (0.61 ± 0.67 vs.1.50 ± 1.16,t=3.725), eye expansion (0.29 ± 0.46 vs.1.28 ± 1.02, t=4.980), Photophobia (0.77 ± 0.76 vs.1.67 ± 0.90,t=4.092), cry (0.29 ± 0.46 vs.0.59 ± 0.91,t=2.038) integral in the treatment group were lower than those in the control group (P<0.05,P<0.01).Conclusion The effect of moxibustion and ear acupoint pressing method in treating dry eye syndrome is more effective than sodium hyaluronate eye drops.
7.Advances in oncolytic adenovirus-loaded mesenchymal stem cells for treating tumors

Oncolytic adenoviruses have been used in cancer gene therapy because of their ability to selectively transfect and repli-cate in tumor cells. However, systemically administering oncolytic adenoviruses is often inefficient because the host's immunity also at-tacks the viruses. Delivering oncolytic adenoviruses by loading them into mesenchymal stem cells effectively solves this problem. Mes-enchymal stem cells have been described to possess a tumor-homing ability, as well as the capacity to deliver oncolytic adenoviruses. This review discusses the current challenges faced by oncolytic adenovirus delivery via mesenchymal stem cells.
8.The correlation between plasma von Willebrand factor and the extent of coronary artery lesion in acute coronary syndrome

Objective To investigate the correlation between plasma von Willebrand factor(vWF) and the extent of coronary artery lesion in acute coronary syndrome (ACS).Methods One hundred and forty eight ACS patients (ACS group) who performed coronary angiography (CAG) were divided into single stenosis group (54 patients),double stenosis group (51 patients) and multitude stenosis group (43 patients) by the number of stenosis through CAG.They were divided into mild group (Gensini score ≤ 20 points,31 patients),midrange group (Gensini score 21-40 points,47 patients) and severe group (Gensini score ≥ 41 points,70 patients).Fifty patients with stable angina pectoris (SA) were randomly selected and classified as SA group,while 50 patients with healthy coronary were randomly selected as control group.Blood sample of 248 patients were collected to detect the level of vWF,and blood routine examination,blood clotting function,liver and renal function,high sensitivity C-reactive protein (hs-CRP),blood glucose,blood fat were detected.The correlation between vWF and the extent of coronary artery lesion were analyzed.Results The level of white blood cell count (WBC) and hs-CRP in ACS group were significantly higher than those in SA group and control group (P < 0.05).But the level of WBC and hs-CRP between SA group and control group had no significant difference (P > 0.05).The level of blood platelet count,prothrombin time,partial prothrombin time,fasting blood glucose,thrombin time,2 h postprandial glucose,triglyceride,total cholesterol had no significant difference among three groups (P > 0.05).The level of vWF in ACS group was significantly higher than that in SA group and control group [(143.25 ± 20.42)% vs.(102.77 ± 11.84)%,(97.63 ± 10.31)%] (P <0.05).The level of vWF in single stenosis group,double stenosis group and multitude stenosis group was (127.34 ± 9.35)%,(144.81 ± 12.02)%,(159.55 ± 18.62)%,and significantly higher than that in control group (P < 0.01).The level of vWF in mild group,midrange group and severe group was (124.77 ± 14.31)%,(132.53 ± 16.38)%,(155.06 ± 18.53)%,and significantly higher than that in control group (P < 0.01).The level of vWF had significantly positive correlation with the number of stenosis and Gensini score (r =0.698,0.683,P < 0.01).Conclusion The level of vWF in peripheral blood is closely related with the severity of ACS and has important clinical significance in evaluating the condition and guiding therapy in patients with ACS.
9.Pathogenic bacteria and drug resistance analysis of 72 cases of premature infants with nosocomial fungal infection

Objective To understand the distribution of pathogen and antibiotic resistance of nosocomial fungal infection,and provide evidence for the prevention of fungal infection in premature infants in hospital.Methods The clinical data of 72 cases of premature infants with nosocomial fungal infections were retrospectively analyzed.Results The top three of the pathogenic of fungal infection in premature infants in our hospital were 38 strains of Candida pelliculosa (52.8%,38/72),10 strains of Candida albicans (13.9%,10/72),9 strains of Candida parapsilosis (12.5%,9/72).The weights of 72 cases with nosocomial fungal infection in preterm infants were below 2000 g,who were dominated by very low birth weight infants.Gestational age of 27 ~ 30 weeks were in the majority,which accounted for 36.1% of 27 ~ 28 weeks (26/72) and 34.7% of 29 ~30 weeks (25/72),each group were dominated by bacteria pathogen candida.Seventy-two strains of fungi were isolated to amphotericin B drug resistance,the 1.4% (1/72) resistance rate to fluconazole,4.2% (3/72) pairs of voriconazole resistance rate,2.8% (2/72) for itraconazole resistant rate.There were no significant differences among the last three resistant rate (x2 =1.02,P > 0.05).The 5-fluorine cytosine resistance accounted for 59.7% (43/72),and the 4 kinds of drug resistance rate had significant difference (x2 =57.73,P < 0.05).There was no significant difference in resistance rate between amphotericin B and fluconazole (x2 =1.01,P > 0.05).Conclusion Fluconazole could be used to those premature infants with high risk factors of fungal infection.Once the fungal infection is diagnosed,intravenous fluconazole could be the first choice,when necessary,amphotericin B can be used together.
10.Contribution of clinical laboratory in handling public health emergencies

Objective Accidental public health emergencies occurred frequently all over the world in the last decade. The problem of how to set up an effective system for prevention and control of infectious diseases has aroused the attention of all the nations worldwide. The present paper discussed the contributions of the clinical laboratory in hospital during public health emergencies by collecting samples, establishing and applying a rapid diagnosis platform for infectious pathogens, ensuring laboratory biosafety, and developing a strategy in case of emergency. These countermeasures may be helpful for the army to establish an effective system and mechanism in handling public health emergencies.
