1.The Mechanisms of Arsenic Trioxide-induced Apoptosis in Human Gastric Adenocarcinoma SGC7901 Cells

OBJECTIVE: To investigate the possible role of the alteration of mitochondrial transmembrane potentials(Δ Ψ m) in arsenic trioxide(As2O3)-induced apoptosis of human gastric adenocarcinoma cells(SGC7901).METHODS: Cell apoptosis was assessed through morphology, cell viability, the percentage of sub-G1 cells and phosphatidylserine(PS) externalization.the Δ Ψ m was detected on flow cytometry through double staining of Rhodamine 123 (Rh123) and propidium iodide(PI).RESULTS: The As2O3-induced apoptosis was associated closely with the disruption of the Δ Ψ m.CONCLUSION: The disruption of the Δ Ψ m is one of the mechanisms of As2O3-induced apoptosis.
3.Investigation of the pathway of apoptosis induced by arsenic trioxide in cancer cells

AIM To investigate the possible role of the mitochondrial transmembrane potential (??m) and caspase 3 in arsenic trioxide(As 2O 3)-induced apoptosis of cancer cells. METHOD Namalwa, SGC7901 and Bcap37 cell lines were used as in vitro models. Apoptosis was confirmed by sub-G 1 cells content as well as phosphatidylserine(PS) externalization. The ??m was detected on flow cytometry through double staining of Rhodamine 123(Rh123) and popidium iodide(PI). In addition, the effect of DEVD-CHO, a selective inhibitor of Caspase 3, on As 2O 3-induced apoptosis was studied. RESULT The As 2O 3-induced apoptosis closely associated with the externalization of the ??m and the activation of Caspase 3. As 2O 3 induced cells necrosis when Caspase 3 was inhibited. CONCLUSION As 2O 3 may selectively activates Caspase 3 after it induces externalization of ??m, which causes cancer cells apoptosis.
4.Detection of Telomerase Activity in the Lesion of Psoriasis

Objective To investigate telomerase activity in patients with psoriasis. Methods The telomerase activity in the skin lesions of patients was detected by TRAP (Telomeric repeat amplification protocol)——ELISA which is based upon PCR amplification of the initial telomerase product and detected by ELISA. Results Telomerase activity was detected in the lesions of psoriatic patients, but the level of it was lower in comparison with that in tissues of malignant tumor and K562 cell line. Conclusion These findings support that the increase of telomerase activity may be linked to the pathogenesis of psoriasis and that the telomerase activity can be detected not only in malignant tumors but also in nonmalignant skin diseases.
5.Effects of ERC61-Extract from Rhizoma Curcumae on Hematopoietic Functions in Mice Treated by Chemotherapy

OBJECTIVE:To explore the effects of ERC61,an extract from Rhizoma Curcumae on hematopoietic functions of mice pretreated by cyclophosphamide(CTX) and the proper regime of the administration of ERC61.METHODS:The experimental mice were treated with CTX(100mg? kg-1? d-1) by ip for 3 consecutive days to establish leucopenia model.ERC61 was ip administered to the prevention group,the treatment group,and the combined group(prevention+ treatment).The numbers of blood leucocytes and bone marrow cell were counted by conventional methods;spleen index was measured as well.RESULTS:ERC61 enhanced the count of leucocytes and nucleated cells in bone marrow and improved the level of spleen index.CONCLUSION:ERC61 is able to improve the disorders of hematopoietic functions induced by CTX,and the combination therapy of prevention and treatment is the optimum one in efficacy.
6.Determination of Curdione in Nano-micelles Loaded with Curdione by UV-Vis Spectrophotometry

OBJECTIVE:To establish a UV-Vis spectrophotometric method for content determination of Curdione in Nano-micelles loaded with Curdione.METHODS:Curdione was colored by sulfuric acid-vanillin reagent.The absorbance was measured by visible-spectrophotometry at a wavelength of 500nm.RESULTS:The content of Curdione in the range of 0.050~ 0.400mg? mL-1 was in accordance with the Lambert-Beer law(r=0.999 3).The average recovery was 99.14%,and the RSD was 1.23%(n=9).CONCLUSION:This method is simple,accurate and suitable for the content determination of curdione in Nano-micelles loaded with Curdione.
7.Analysis of the risk factors of female breast cancer

Objective To explore the risk factors of female breast cancer and provide the basis for the etiology of female breast cancer,the identification of the high-risk population and the development of effective intervention measures.Methods The breast cancer patients who were confirmed by pathology were collected,and screened for healthy women whose age and living environment were similar to breast cancer patients.The questionnaire was used to collect risk-factors of breast cancer.The x2 test were used to analyze statistical difference of case group and control group.1 ∶ 2 logistic regression was used to analyze the relationship between risk factors and female breast cancer.Results There were 184 breast cancer patients and 368 healthy women in research.Benign breast disease,breastfeeding,abortion,depression,smoking,passive smoking,eating fried or smoked foods,drinking tea had statistical differences between case group and control group.And benign breast lesions(OR =1.553,95% CI 2.045 ~ 10.924),abortion(OR =0.775,95 % CI 1.423 ~ 3.309),smoking(OR =0.674,95% CI 1.009 ~ 3.817) passive smoking (OR =2.98,95% CI 1.680 ~ 3.720) were the risk factors of breast cancer,and drink tea(OR =-0.425,95% CI 0.446 ~ 0.958),eating soy products (OR =-0.687,95 % CI 0.284 ~ 0.889) breast-feeding (OR =-0.827,95%CI 0.193 ~ 0.991) were the protective factors of breast cancer.Conclusion The risk factors of female breast cancer in Binzhou are benign breast disease,abortion,passive smoking,depression,and the protective factors are drinking tea and breast-feeding.
9.Study of mutant prevention concentrations of macrolide-resistant Streptococcus pneumoniae

Objective To detect the mutant prevention concentration (MPC) of macrolide resistant Streptococcus pneumoniae, to amplify the resistance gene of Streptococcus pneumoniae mutants selected in the mutant selection window (MSW), and to investigate the mechanism in the resistance of Streptococcus pneumoniae to macrolides. Methods The Streptococcus pneumoniae strain ATCC49619 was enriched in broth, and the bacterial concentrations were adjusted to 1010 colony forming units per milliliter. The minimal inhibitory concentration (MIC), MIC for 99 % of input cells (MIC99), provisional MPC (MPCpr), and MPC of roxithromycin and azithromycin for Streptococcus pneumoniae were determined by agar plate dilution method. The ermB and mefA of Streptococcus pneumoniae mutants selected in the MSW were obtained by PCR method and sequencing. Results The MPCs of roxithromycin and azithromycin for ATCC49619 were 0.80 ?g/ml and 0.51 ?g/ml, and the MPC/MIC99 were 5.0 and 3.9. The ermB gene was detected in Streptococcus pneumoniae mutants selected in the MSW. Conclusion Adjustment of the drug dose may limit the enrichment of macrolide-resistant Streptococcus pneumoniae mutant. The mechanism in the resistance of Streptococcus pneumonia to macrolides may be associated with ermB genes carried.
10.Efficacy and side effects of methotrexate in the treatment of ectopic pregnancy

Objective To study the efficacy and side effects of methotrexate with different protocols in the treatment of ectopic pregnancy (EP). Methods Total of 648 patients who had EP and were treated only with MTX were analysed.Patients were divided into six groups according to protocol: Group 1,100 mg intravenously( IV); Group 2,100 mg, IV, followed by citrovorum factor; Group 3, 20 mg, IV every day for five days; Group 4, 20 mg intramusculary (IM) every day for five days; Group 5, 75 mg,IV; Group 6, 75 mg, IM.Results The rates of repeated MTX injection in group1~6 because of inadequate decrease of hCG were 23.3%,25.0%,21.4%,20.6%,24.4% and 22.4% respectively .Success rates were 87.4%,85.4%,90.5%,92.6%,86.3% and 91.4% respectively.Rates of liver dysfunction were 10.3%,8.3%,64.3%,69.1%,8.7% and 31.0%.Conclusion Single-dose of 75mg MTX IV injection may be the best regimen in the treatment of EP because of the same efficacy but the least side effects.
