1.Incidence of Helicobacter pylori infection in 108 youngsters and drug resistance of the isolated strains

Objective To investigate the incidence of Helicobacter pylori in teenagers and analyze the drug resistance of isolated H.pylori strains.Methods Gastric mucosa samples were harvested from 108 young patients by gastroscopic biopsy.The Kirby-Baner test was performed to test the drug susceptibility of isolated H.pylori strains.Results Thirty-six strains of H.pylori were isolated from the gastric mucosa of 108 patients(33.3%).Seven of the 36 H.pylori strains were originated from the patients aged 6-10 years,the isolation rate in this age group was 16.7%;4 strains were isolated from the patients aged 10-14 years,the isolation rate in this age group was 31.3%;26 strains were from the age group of 14-18 years with the isolation rate of 48.1%.The isolation rates of H.pylori were significantly different among the there age groups(P0.05).The drug susceptibility test indicated that the drug-resistant rate of the 36 H.pylori strains to clarithromycin,amoxicillin,gentamicin,fruranzolidone,metronidazole and levofloxacin was 8.3%(3/36),33.3%(12/36),0%(0/36),16.7%(6/36),94.4%(34/36) and 0%(0/36),respectively.Conclusions The frequency of H.pylori infection may incidence with the increase in age.For the isolated H.pylori strains in present study,the drug-resistance to metronidazole and amoxicillin is higher,to clarithromycin and fruranzolidone is lower,and no drug-resistance is found to gentamicin and levofloxacin.
2.Study on application of hospital-school combined three-dimensional teaching mode in the operation nursing practice teaching

Objective To explore the effect of application of hospital-school combined three-dimensional teaching mode in the operation nursing practice teaching.Methods 129 students were collected by cluster sampling,then based on class unit,they were randomized into the experimental group (69 students) and the control group (60 students).The experimental group used hospital-school combined threedimensional teaching mode and the control group used traditional teaching mode.Then,both groups had finished an operation nursing comprehensive theoretical examination and a self-report questionnaire for mastery of basic knowledge of operation nursing.Besides,the experimental group had a questionnaire survey of cognitive evaluation for the new teaching mode.Results The average score of operation nursing comprehensive theoretical examination of the experimental group was (77.81 ±9.87),higher than the control group (70.35±12.37); The self-report score for mastery of basic knowledge of operation nursing of the experimental group was (3.94±0.45),higher than the control group (3.68±0.46).The students of the experimental group also considered that the new teaching mode improved their comprehensive knowledge and innovative abilities,critical thinking ability,humanistic care spirit,communication ability,team cooperation ability and innovation ability,etc.Conclusions The effect of application of hospital-school combined three-dimensional teaching mode in the operation nursing practice teaching was good,but it still had some limitations which needed us to pay attention to and to perfect.
3.Chondrogenic differentiation of bone marrow mesenchymal stem cells

BACKGROUND: Bone marrow mesenchymal stem cells are important seeded cells for construction of tissue-engineered trachea, but there is no special surface marker. Therefore, identification of bone marrow mesenchymal stem cells is mostly based on morphology, phenotype antigen and the function of differentiation. OBJECTIVE: To explore the feasibility of the tracheal chondrogenic differentiation of bone marrow mesenchymal stem cells under a special condition through isolation, cultivation and identification of bone marrow mesenchymal stem cells. METHODS: Rabbit bone marrow was acquired in the sterile environment to isolate and culture bone marrow mesenchymal stem cells to passage 2 by bone marrow adherence and screening method. Flow cytometry identified the phenotype CD44, CD45 of bone marrow mesenchymal stem cells at passages 1 and 2. Rabbit tracheal samples were acquired in the sterile environment, the tracheal chondrocytes were isolated and cultured by enzyme digestion, and toluidine blue staining was used to detect aggrecan. Bone marrow mesenchymal stem cells were co-cultured with tracheal chondrocytes by Transwel and transforming growth factor β1. Cel morphology was detected under an inverted microscope. Real-time quantitative PCR and toluidine blue staining detected the extracel ular matrix components, such as type Ⅱ col agen and aggrecan.RESULTS AND CONCLUSION: After isolation and culture, cells were spindle and irregular in morphology, and passaged cells thrived that were gathered into a fish-like colony growth. For passage 1 bone marrow mesenchymal stem cells, the positive rates of phenotype antigen CD44 and CD45 were respectively 96.97% and 13.72%; for passage 2 cells, the positive rates of phenotype antigen CD44 and CD45 were 99.11% and 8.54%, respectively. Tracheal chondrocytes were positive for toluidine blue staining. The morphology of induced bone marrow mesenchymal stem cells changed from long fusiform to triangular or irregular shape, indicating the chondrocytes expressed type Ⅱ col agen and aggrecan, and toluidine blue staining was positive. These results showed bone marrow adherence and screening method could acquire bone marrow mesenchymal stem cells, and the purity of passage 2 cells is higher. Under a special condition, bone marrow mesenchymal stem cells have the potential of chondrogenic differentiation, and can be selected as seed cells for construction of tissue-engineered trachea.
