1.Expression of glypican-3 in ovarian carcinoma and its clinical significance

Objective To study the expression of GPC3 mRNA and GPC3 protein in ovarian carcinoma, and to investigate the clinical significance of their expression. Methods 35 specimens of ovarian carcinomas and 23 specimens of normal ovarian tissues were obtained from the patients undergoing operation during Jan. 2004 to Oct. 2005. The expressions of GPC3 mRNA in ovarian carcinomas and normal ovarian tissues were detected by real-time fluorescence quantitative PCR (RT-PCR). The clinical and pathological characteristics of ovarian carcinomas were analyzed. The expression of GPC3 protein was detected in 4 specimens of ovarian carcinomas and 3 specimens of normal ovarian tissues by Western bloting. Results The expression level of GPC3 mRNA in ovarian carcinomatous was down-regulated compared with that in normal ovarian tissues (P0.05). The expression of GPC3 protein was consistent with the expression of GPC3 mRNA. GPC3 protein was doubly or trebly expressed in normal ovarian tissues compared with ovarian carcinomas. Conclusion The present study suggests that GPC3 may be related to the occurrence and development of the ovarian carcinoma. The simultaneous determination of GPC3 and CA125 may increase the sensitivity of diagnosis for ovarian carcinoma.
2.Determination of Fenofibrate in Fenofibrate Capsules by HPLC

Objective:To establish a method for the determination of fenofibrate in fenofibrate capsules .Methods: HPLC was performed on a Dikma Diamonsil C18column (150 mm ×4.6 mm, 5 μm).The mobile phase was acetonitrile-water (7030, adjusting pH to 2.5 with phosphoric acid) and the detection wavelength was 286 nm.The flow rate was 1.0 ml· min-1 and the column tempera-ture was 30 ℃.The injection volume was 20 μl.Results:The linear range of fenofibrate was 10.07-60.42 μg· ml-1(r=0.9996) and the average recovery was 99.26%(RSD=0.5%,n=6).Conclusion:The method is simple, accurate and special, which can be used for the quality control of fenofibrate capsules .
3.Control study of personality,interpersonal trust and coping style of juveniles in criminal gangs

ObjectiveTo study the personality traits,interpersonal trust and coping style of juveniles in criminal gangs and their relationship. Methods 15 male juveniles who committed crimes without gangs ( group1 ),52 male juveniles who committed crimes by gangs( group2 ) and 40 male lawful freshmen( group3 ) were tested with Eysenck Personality Questionnaire ( EPQ),Trust Scale and Coping Style Questionnaire.The data were statistically analyzed by covariance analysis or Pearson relation analysis.ResultsAfter the covariant effect of education levels was removed,the levels of P and N in EPQ of group1 ( P:57.49 ± 2.62,N:59.35 ± 3.55 ) and group2 ( P:57.83 ± 1.24,N:59.60 ± 1.68 ) were higher than those of group3 ( P:43.88 ± 4.72,N:39.07 ±6.40) with statistical difference (P<0.05).The level of E of group1 (63.41 ±3.86) was higher than that of group2 (53.01 ± 1.83 ) with statistical difference (P < 0.05 ).The levels of P,D and F in Trust Scale of greup1 (P:24.68 ± 1.51,D:14.36 ±2.19,F:15.49 ±2.21) and group2(P:22.95 ±0.71,D:22.48 ± 1.04,F:23.09 ±1.05 ) were lower than those of group3 ( P:33.14 ± 2.72,D:40.22 ± 3.95,F:38.44 ± 3.99) with statistical difference (P < 0.01 ).The levels of self-accusation and fantasy in Coping Style Questionnaire of group1 ( 0.80 ±0.08,0.83 ± 0.06 respectively) and group2 (group2:0.59 ± 0.04,0.68 ± 0.03,respectively) were higher than those of group3 (0.39 ± 0.14,0.44 ± 0.11,respectively) with statistical difference (P< 0.05 or 0.01 ),while the levels of problem-solving of group1 ( 0.76 ± 0.06 ) and group2 ( 0.70 ± 0.03 ) were lower than that of group3 ( 0.95 ± 0.11 ) with statistical difference (P < 0.05 ).For the male juveniles who committed crimes by gangs,the P of personality traits had a statistical linkage with the problem-resolving and self-accusation of coping style ( r =- 0.389,- 0.395,P < 0.05 ),the N of personality traits had a statiseical linkage with the F of interpersonal trust and the self-accusation of coping style ( r=-0.473,0454,P < 0.05 ),and the E of personality traits had a statistical linkage with the help-seeking of coping style ( r=0.400,P< 0.05 ).ConclusionThe personality traits,interpersonal trust and coping styles of male juveniles who committed crimes with or without gangs and the lawful men are different.These psychological factors may have certain effect on juvenile gang crimes.
4.Ultrasonic Features of Wilms′ Tumor in Children

Objective To study the ultrasonographic characteristics of Wilms′tumor (WT), aims at elevating the diagnostic level.Methods We retrospectively studied sonographic features in 30 cases of WT proved by operation and pathology.Results The tumors with heterogeneous pattern of solid and cystic tissue were shown in 22 cases.Seven cases demonstrate predominantly echogenic tumor. A huge cystic mass with numerous septas was shown in 1 case. The tumor extended into renal pelvis in 4, into renal vine in 1.The tumor metastasized to lymphnod in 1, to bilateral lungs in 1. Among 30 cases, the tumor was huge and grew beyond the renal profile in 26 cases, in 3 cases the tumors took up the renal sinus and made its structure turbulence, the renal shape was approximately normal.One case was extrarenal WT,the tumor located in retroperitonemu.Conclusions Sonographically, typical WT is a large heterogeneous mass within or without cystic areas, it also can be a pure cyst. It is easy to diagnose. When the tumor is limited to renal outline, it is easy to confusion with renal cancer. In addition, there are bilateral WT and extrarenal WT. The sonographic characteristics is similar among WT, clear cell sarcoma of kidney, malignant rhabdoid tumor and congenital mesoblastic nephroma. It is difficult to distinguish them from ultrasonography.
5.Preparation and Dissolution of Jieheling Dispersible Tablets

Objective:To prepare Jieheling dispersible tablets and establish the dissolution determination method. Methods:The dissolution of fischeriana B in the tablets was determined by HPLC. The column was Kromasil C18(250 mm ×4.6 mm,5μm)with ace-tonitrile-water (40∶60) as the mobile phase at a flow rate of 1. 0 ml·min-1 . The detection wavelength was at 290nm. The column temperature was 30℃ and the sample size was 100 μl. Results: The linear range of fischeriana B was 0. 005-0. 057 μg ( r =0. 999 6). The average recovery was between 95% and 105%. Phosphate solution with pH of 6. 8 was used as the dissolution medi-um. Conclusion:The method is simple, accurate and specific, and can be used in the quality control of Jieheling dispersible tablets.
6.Effect of 5-Aza-CdR on pancreatic carcinoma Capan-2 cell proliferation and PCDH8 gene expression

Objective To discuss the effect of 5-Aza-CdR on pancreatic carcinoma Capan-2 cell proliferation and PCDH8 gene expression.Methods Pancreatic carcinoma Capan-2 cells were treated with different doses of 5-Aza-CdR with or without gemcitabine,negative control group without drug,0.08μmol/L group with 0.08μmol/L 5-Aza-CdR,0.40 μmol/L group with 0.40 μmol/L 5-Aza-CdR,2.00 μmol/L group with 2.00 μmol/L 5-Aza-CdR,10 μmol/L group with 10 μmol/L 5-Aza-CdR,50 μmol/L group with 50 μmol/L 5-Aza-CdR. The inhibition ratio of Capan-2 cell proliferation were observed by MTT assay and PCDH8 gene and protein expression were detected by RT-PCR and Western blot. Results The inhibition ratio was increased with 5-Aza-CdR dose increasing(P<0.01),but decreased apparently with times extending(P<0.01). Inhibition ratio in 5-Aza-CdR and gemcitabine group was higher than those with only 5-Aza-CdR or gemcitabine groups(P<0.05 or P<0.01).The levels of PCDH8 gene and protein expression were increased significantly in 5-Aza-CdR treatment groups,with dose dependent (P <0.01 ).Conclusion 5-Aza-CdR can inhibit the proliferation of pancreatic carcinoma Capan-2 cell proliferation,and increase PCDH8 gene expression. The inhibition effect is strong when combined with gemcitabine.
7.Acceleration of apoptosis by transfection of Apoptin gene in retinoblastoma cells

Objective Present study aimed to observe the effects of Apoptin gene on killing retinoblastoma HXO-RB_(44) cells and illustrates its mechanisms. Methods Human retinoblastoma cells strain, HXO-RB_(44), was cultured and passaged in RPMI 1640 medium containing bovine serum. Apoptin gene was transfected into HXO-RB_(44) cells by liposome into HXO-RB_(44)/Apoptin, and pcDNA_3 was transfected in HXO-RB_(44)/peDNA_3 group. The expression of Apoptin mRNA was detected using Reverse Transcription Polymerase Chain Reaction (RT-PCR). The expression of protein of Apoptin and p53 were detected by SABC immunohistochemistry. The growth rate of HXO-RB_(44) cells was studied by constructing the growth curve and calculated as the formula: inhibitory rate = 1-cell number in experiment group/cell number in control group x 100%. Cellular apoptosis was determined by flow cytometry. Results The RT-PCR result showed the 450 kb specific band in UXO-RB_(44)/Apoptin group and absent amplification result in HXO-RB_(44) group and HXO-RB_(44)/pcDNA_3 group. The difference in SABC-positive cell number between HXO-RB_(44)/Apoptin group and control group was statistically significant (P < 0. 05). The growth of HXO-RB_(44) cells was significantly inhibited in HXO-RB_(44)/Apoptin group compared with control group (P < 0.05). Apoptosis cells increased significantly. The apoptosis rate was 38. 5% . Conclusion Apoptin gene could inhibit the growth of HXO-RB_(44) cells effectively. Up-regulation of expression of p53 gene might not be one of cell apoptosis mechanisms.
8.Statistical analysis of serum lipid classification and phenotyping of Beijing professional populations

Objective To study the characteristics of lipid level classification and phenotyping of Beijing professional populations. Method 4 033 cases (M/F=2 835/1 198, aged 40~91) were enrolled in the study. Lipid levels were classified according to the "Recommended guidelines for prevention and treatment of dyslipidemia" of China (1997) and NCEP ATPⅢ(2001) of US. Results High prevalence of lipid abnormality was found in Beijing populations, only 1/3 cases had desirable cholesterol (TC) and triglyceride (TG) level in the groups with age above 50, and more than half of the subjects had high TC levels at this age, the occurrence of high TG was much less than high TC. Hyperlipoproteinemia typeⅡa was 2~3 times more than type Ⅳ.Decreased high density lipoprotein cholesterol (HDL-C) was found in 20% male subjects, but only 5% in females. The prevalence of high level HDL-C was found in 25%~39% of female groups, and 10%~14% in male groups. High HDL-C was usually associated in cases with high TC, but low HDL-C was usually seen in high TG cases.Conclusion The results showed that the prevalence of lipid abnormality was as high as 2/3 of the studied subjects above age 50, it means that the cardiovascular disease risk is high in this population, Lipid modification may be beneficial to primary prevention of coronary artery disease.
9.Relationship between noninvasive brachial artery blood pressure and radial artery blood pressure of the right arm of patients

Objective To investigate the relationship between noninvasive brachial artery blood pressure (B) and radial artery blood pressure (R) of the right arm.Methods Two hundred and ninetyfive patients with 149 males and age of (47 ± 16) years were studied.The height of patients was 163 ± 8 cm,and weight of patients was (61.2 ± 7.8) kg.The patients with peripheral vascular disease,wounds of arm skin or subcutaneous tissue infection were all excluded.Their B (with adult cuffs) and R (with infant cuffs) of the right arm were measured and analyzed after the patients under general anesthesia and stable hemodynamics.The relationships between B and R were analyzed by linear regression,the differences between B and R of each interval were compared using one-way ANOVA and then followed by SNK procedure.Results Right brachial artery blood pressure was significantly lower than radial artery blood pressure.The differences between the two varied from 13 to 18 mmHg in systolic BP (SBP),diastolic BP (DBP) and mean blood pressure (MAP).And linear regression was most applicable to describe their correlation [r=0.841 (SBP),0.808 (DBP),0.833 (MAP),all P<0.01].Conclusions Radial artery blood pressure measured with infant cuffs can well reflect the variation of brachial artery blood pressure.
10.Passive movement and the expression of brain derived neurotrophic factor and the B cell lymphoma/leukemia-2 gene after cerebral infarction

Objective To observe the effects of passive movement on the functional outcome after occlusion of the middle artery in the brain and reperfusion, and to explore the molecular mechanisms involved. Methods Cerebral infarction models were established in rats using left middle cerebral artery occlusion ( MCAO). The survivors were randomly divided into a passive movement group and a natural recovery group. There was also a sham-operated group and a normal group. Passive movement treatment (twice a day, twenty min per time) was started at different times after reperfusion. The expression of brain-derived neurotrophic factor (BDNF) and B cell lymphoma/leukemia-2 gene (Bcl-2) were determined using real-time PCRs. Results Expression of BDNF and Bcl-2 was detected a-round the infarction area in both groups. The expression of BDNF and Bcl-2 was highest in the sub-groups where passive movement was begun 24 or 48 h after the operation. Conclusions The expression of BDNF and Bcl-2 in the brain peaks when daily, moderate intensity passive movement is administered beginning 24 to 48 h after reperfusion. Passive movement might have a protective and rehabilitative effect after cerebral infarction.
