1.Risk factors for no-reflow after emergency coronary intervention in aged patients with acute ST eleva-tion myocardial infarction

Objective:To explore risk factors for no‐reflow after emergency coronary intervention in aged patients with a‐cute ST elevation myocardial infarction (STEMI) . Methods:According to presence of no -reflow (≤TIMI grade Ⅲwas considered as no-reflow) after operation or not ,a total of 700 aged STEMI patients hospitalized in our hospital during 2010-2013 were divided into no-reflow group (n=190 ,27. 14% ) and reflow group (n=510 ,72. 86% ) . Clinical data , PCI and coronary angiography data were collected ,compared and analyzed between two groups . Results:Compared with reflow group ,there were significant rise in percentages of patients with TIMI grade 0-1 (61.17% vs. 82.11% ) ,coro‐nary collateral blood flow grade 0 (64.12% vs. 74.21% ) ,5 thrombus scores before PCI (58.83% vs. 80.00% );signifi‐cant reduction in systolic blood pressure (SBP) at hospitalization [ (111.2 ± 24.6) mmHg vs. (101.7 ± 25.9) mmHg] in no-reflow group , P<0. 01 all. Multi-factor Logistic regression analysis indicated that SBP<101 mmHg at hospitaliza‐tion ,collateral blood flow grade 0 before PCI and 5 thrombus scores before PCI were risk factors for no‐reflow after emer‐gency PCI (OR=1.006~4.398 , P<0.05 or <0.01) .Conclusion:In aged acute STEMI patients ,those with risk factors for no-reflow after emergency PCI should take corresponding preventive and therapeutic measures in order to improve their prognosis .

To build the Dendrobium nobile -T2DM network, and elucidate the molecular mechanism of D. nobile to type 2 diabetes (T2DM). Collect the chemical composition of D. nobile and the targets on T2DM by retrieving database and documents, build the network of D. nobile to T2DM using the entity grammar systems inference rules. The molecular mechanism of D. nobile to T2DM includes: (1) regulating lipid metabolism by lowering triglyceride; (2) reducing insulin resistance; (3) protecting islet cells; (4) promoting the glucose-dependent insulin tropic peptide (GIP) secretion; (5) inhibiting calcium channel. Under the guidance of network pharmacology, through entity grammar systems inference rules we elucidate the molecular mechanism of D. nobile to T2DM, and provide the basis for the further development of health care products based on D. nobile.

3.Analysis of risk factors in elderly type 2 diabetes mellitus with cerebral infarction

Objective To explore the risk factors in elderly type 2 diabetes mellitus with cerebral infarction. Methods Retrospective investigarion was performed on 148 elderly hospitalized patients with type 2 diabetes.The patients were classified based on the presence or absence of cerebral infarction and compared with 60 controls.Logis- tic regression analysis was used to reveal the risk factors for cerebral infarction.Results The levels of systolic blood pressure(SBP),body mass index (BMI),fasting blood glucose (FBG),total cholesterol (TC),triglyceride (TG) and plasma fibrinogen(Fg) were higher in the patients with cerebral infarction[141.15?17.46)mmHg,(23.81?3.53)kg/m~2,(8.82?2.81)mmol/L,(5.69?1.15)mmol/L,(2.08?0.75)mmol/L and (4.08?0.65)g/L] than those without cerebral infarction[(129.78?14.65) mmHg,(22.18?3.22)kg/m~2,(7.06?1.72 )mmol/L,(5.09?1.12)mmol/L,(1.62?0.43)mmol/L and (3.48?0.58)g/L].The logistic analysis showed COUR,SBP,FBG, TC,TG and Fg were the independent risk factors for cerebral infarction.Conclusion Early intervention of the inde- pendent risk factors including SBP,FBG,TC,TG and Fg in elderly patients with type 2 diabetes was important for reduction and postponement of cerebral infarction.

5.Studies on antitumor effects of podophyllotoxin nanoliposome.

OBJECTIVETo compare the antitumor effects of the podophyllotoxin nanoliposome and the suspensions of podophyllotoxin in bearing-tumor mice.
METHODThe experimental mice were inoculated by suspension liquid of tumor cell in axillary region near the forelimb, 0.2 mL each. The mice had been randomly divided into 8 groups 24 h latter. The podophyllotoxin nanoliposome, suspension liquid of the podophyllotoxin, CX or NS was given to each group respectively. Except CX was given by celiac injection once, the other groups were injected every 4 days for three times. The mice were killed by hauling necks on the twelfth day after treatment, the tumor was weighed and the inhibitory rate was calculated.
RESULTWhen the dosage of podophyllotoxin nanoliposome and the suspensions of podophyllotoxin reached 5.0 mg x kg(-1), the rate of inhibiting tumor were 52.37% and 38.25% to the H22 liver cancer of mice respectively.
CONCLUSIONPodophyllotoxin nanoliposome and the suspensions of podophyllotoxin have the effect in anti-liver cancer. And podophyllotoxin nanoliposome have stronger inhibitory rate compared with suspension liquid of the podophyllotoxin in same dose.

ObjectiveTo establish a new method for preparing synaptosomes.MethodsDensity gradient centrifugation method was used to isolate synaptosomes of mouse, checking by transmission electron microscopy.ResultsSynaptosomes prepared by this method had intact morphological characteristics, surrounding with a continuous oval-shaped membrane structure, moreover, mitochondrion and lots of synaptic vesicle in them.ConclusionThis method is applicable to establish a rapid, convenient and useful method for preparing synaptosomes.

A quantitative HPLC-DAD method was developed for simultaneous determination of N-trans-p-coumaroyloctopamine and N-trans-p-coumaroyltyramine in Solani Melongenae Radix from different cultivation regions in China The separation was performed on an Agilent Eclipse XDB C18 column (4.6 mm x 250 mm, 5 microm) at 30 degrees C with a gradient elution of methanol and 0.1% formic acid in water as mobile phase. The flow rate was set at 1.0 mL x min(-1) and the detection wavelength was 300 nm. The calibration curves of N-trans-p-coumaroyloctopamine and N-trans-p-coumaroyltyramine were linear over the ranges of 2.84-68.16, 3.10-74.40 mg x L(-1), and the average recoveries (n = 9) were 99.30% and 102.8%, respectively. The developed method was successfully applied for the analysis of sixteen samples from different cultivation regions in China, which indicated that the method is simple, rapid, accurate, and reliable for quality evaluation of Solani Melongenae Radix.

Objective To study the bioequivalence of domestic and im-ported cefiibuten tablets in healthy volunteers.Methods A single oral doses of 400 mg ceflibuten was given to 22 healthy volunteers in an open randomized crossover design.The concentrations of ceftibuten in plasma were determined by a HPLC method.The pharmacokinetic parameters of domestic and imported were calculated by 3 P97 software.Results The main phannacokinetic parameters of domestic and imported were as follow:t_(max) were(2.18±0.97)and(2.43±0.81)h;C_(max) were(18.58±2.70)and(18.95±4.38)μg·mL~(-1);t_(1/2) were(2.41±0.86)and (2.36±0.70)h;AUC_(0-t) were(95.76±14.54)and(96.14±18.87)μg·h·mL~(-1),respectively.The relative bioavailability of ceftibuten was(101.77±17.72)%.Conclusion The results demonstrated that the two preparations of cefiibuten were bioequivalent.
10.In vitro metabolism of forscolin isolated from Coleus forskohlii.

This paper is to report the study of the metabolism of forscolin in plasma and liver microsomes for guiding clinical therapy. Forscolin was quantified by HPLC-MS/MS. The metabolic stability of forscolin in rat, Beagle dog, monkey and human plasma and liver microsomes, mediated enzymes of forscolin and its inhibition on cytochrome P450 isoforms in human liver microsomes were studied. Results showed that forscolin was not metabolized in plasma of the four species but metabolized in liver microsomes of the four species. The t1/2 of forscolin in rat, Beagle dog, monkey and human liver microsomes were (52.0 +/- 15.0), (51.2 +/- 5.9), (6.0 +/- 0.2) and (11.9 +/- 1.8) min; CL(int) were (75.6 +/- 18.7), (60.9 +/- 6.8), (513.8 +/- 14.3) and (176.2 +/- 25.6) mL x min(-1) x kg(-1); CL were (34.8 +/- 4.5), (23.3 +/- 1.0), (40.3 +/- 0.5) and (17.9 +/- 0.3) mL x min(-1) x kg(-1), respectively. Forscolin was metabolized by CYP3A4 in human liver microsomes. There was definite inhibition on CYP3A4 at the concentrations of forscolin between 0.1 ng x mL(-1) and 5 microg x mL(-1). Therefore, forscolin is rapidly excreted from liver microsomes. Attention should be paid to the drug interaction when forscolin was used along with other drugs metabolized by CYP3A4 in clinics.
