1.CT findings of central nervous system in congenital syphilis infant

Objective To investigate the CT features of the central nervous system in congenital syphilis infant. Methods CT findings of central nervous system in 11 infants with clinically proved congenital syphilis were analyzed retrospectively. Results CT findings in 10 syphilis neonates were diffuse hypodense lesions in the white matter, with subarachnoid and intra-encephalic hemorrhage in 3 and 1 cases, respectively. One 2-month-old syphilis infant case and 5 cases of follow-up after 45 days to 6 months of treatment demonstrated bilateral widened sulci and cistern with enlarged ventricles in 3 of them. Conclusion CT findings of the central nervous system in congenital syphilis infant are similar to those of hypoxic-ischemic encephalopathy in neonates, and extra-encephalic hydrocephalus or brain hypogenesis ensues later on.
3.cDNA microarray in the gene expression pattern in lymphatic metastasis of colon carcinomas

Objective To analyze differential gene expression profiles by cDNA microarray in colon carcinomas with or without lymphatic metastasis. Methods cDNA microarray was prepared by spotting polymerase chain reaction (PCR) products of 16 000 human genes onto specially treated glass slides. The cDNA probes were prepared by labeling cancer tissue mRNA and lymphatic metastasis tissue mRNA with Cy3-dUTP and Cy5- dUTP through reverse transcription. The mixed probes were ,then hybridized to the cDNA microarray. The chips were scanned by Agilent fluorescence scanner and analyzed by gene Pix QuantArray. Results Among the 16 000 target genes, 999 genes were screened out for differences in gene expression level in the cases with colon carcinoma and lymphatic metastasis, among which 537 were up-regulated and 462 down-regulated. There were many genes evolved in the metastasis of colon carcinoma, including oncogenes, tumor-suppressor genes, adhesion molecular, matrix metalloproteinases, signal transduction factors, metabolism, immune associated genes, etc. Conclusion The genes, being closely associated with carcinoma metastasis, could be considered as potential markers to predict metastasis and targets for antimetastasis intervention.
4.Analysis of clinical index for the aged suffering from type 2 diabetes mellitus in southern part of Guizhou among different nationalities

Objective To investigate the difference and significance of the clinical index for the aged suffered from type 2 diabetes mellitus in southern part of Guizhou. Methods A total of 240 old patients of type 2 diabetes mellitus were divided into 5 groups according to the complications (non-complications, 43; cerebrovascular disease, 20; macroangiopathy, 43; peripheral neuropathy, 67; diabetic retinopathy, 67) and 5 groups according to the different nationalities (Han 90, Miao 60, Buyi 70, Shui 12, Other 8). 7.Role of protein tyrosine phosphatase SHP-2 in group I metabotropic glutamate receptors-induced allodynia

Aim To investigate whether the pain modi-fication by group I metabotropic glutamate receptors (mGluRs)required the involvement of Src homology-2 domain-containing phosphatase-2 (SHP-2 ).Methods Co-immunoprecipitation was performed to examine the possible interaction between SHP-2 and group I mGluRs in spinal cord dorsal horn of mice.By measur-ing the paw withdrawal thresholds,the effects of SHP-2 inhibitor NSC-87877 or its catalytically inactive SHP-2 (C459S ) mutant on allodynia induced by group I mGluRs agonist DHPG (50 nmol)were observed.Re-sults Anti-mGluR5 antibody was able to co-immuno-precipitate SHP-2 from spinal dorsal horn of mice, while no SHP-2 was precipitated by anti-mGluR1 anti-body.Inactivation of SHP-2 by NSC-87877 (6 nmol) or SHP-2 (C459S ) effectively attenuated allodynia caused by DHPG.Conclusion SHP-2 can physically interact with mGluR5.The activation of SHP-2 may be necessary for group I mGluRs to process the nocicep-tive information.
8.Determination of valproic acid in human plasma by LC-MS/MS method

Objective To develop a rapid and sensitive liquid chromatography-tandem mass spectrometric(LC-MS/MS) method for the determination of valproic acid in human plasma.Methods After treating human plasma sample by acetonitrile protein precipitation method,the analytes were separated on a Shimpack VP-ODS analytical column(150 mm×2.0 mm I.D,5 μm) with the mobile phase of methanol and 5 mmol/L ammonium acetate (55∶45,v/v)at a flow rate of 0.4 mL/min.Detection was carried out by adopting the multiple reaction monitoring(MRM) scanning mode in the API3200 triple quadrupole tandem mass spectrometer,electrospray ionization source,negative ion mode,selected monitoring ionic reactions were m/z 142.9→m/z 142.9(valproic acid) and m/z 179.0→m/z 179.0(1-sulfonic acid).Results Valproic acid and internal standard 1-sulfonic acid retention time were 3.03 min and 2.38 min respectively.The plasma valproic acid linear range was 0.800-80.0 μg/mL(r>0.99) with the lower limit of quantitation(LLOQ) 0.800 μg/mL.The intra-and inter-batch relative standard deviations(RSD) were both less than 15%,and the relative errors(RE) were within ±15%.The mean extraction recovery rate was(84.1±2.4)%,and the mean matrix effect factor was(104.3±2.0)%.In the stability study,valproic acid was found to be stable in plasma under various storage conditions.Conclusion This method is suitable for the determination of valproic acid in human plasma and human pharmacokinetic study of valproic acid semisodium sustained release tablet.
9.The effect of different illumination condition on drosophila circadian rhythm and 24 h sleep time

Objective To investigate the influence of illumination changes on the drosophila circadian rhythm and 24 h sleep time.Methods We collect the unmated fruit flies (a total of 128,single-sex) under CO2 anesthesia within 8 hours of eclosion.And put them into the group of normal light group,day and night reversed group,24 h light group and 24 h dark group with the simple random sampling method,male and female with 4 single-sex groups(n=32).Change the illumination conditions of the living environment of the fruit flies and use Drosophila Activity Monitoring System (DAMS) and Data Acquisition System (DAS),to observe and compare the changes of drosophila circadian rhythm and 24 h sleep time between the different illumination conditions.Results Compared with the normal light group,drosophila circadian rhythm of the day and night reversed group was significantly reversed,the number of fruit flies' activities of 24 h light group was increased,and circadian rhythm was decline,drosophila circadian rhythm of the 24 h dark group is still there,but frequent activity appears during the day,and the 19 : 00 activity peak disappears.Conclusion The drosophila circadian rhythm and 24 h sleep time changes with illumination conditions change.
10.Progress in microRNAs associated with major avian viruses.

Recently, avian viral diseases have become one of the main models to study mechanisms of viral infections and pathogenesis. The study of regulatory relationships and mechanisms between viruses and microRNAs has also become the focus. In this review, we briefly summarize the general situations of microRNAs encoded by avian herpesviruses. Also, we analyze the regulatory relationships between tumorigenicity of avian herpesviruses and microRNAs. Additionally, the possible applications for prevention and treatment of viral diseases (such as infectious bursal disease, avian influenza and avian leucosis) using the regulatory mechanisms of microRNAs are also discussed.
