1.Study of HBV-X gene mutation among patients with HBV-related chronic hepatitis,liver cirrhosis,and primary liver cancer

Objective To study the relationship between hepatocarcinogenesis and the mutation in X gene among patients with chronic hepa-titis B virus (HBV)infection,such as chronic hepatitis B (CHB),liver cirrhosis (LC)and primary liver cancer (PLC).Methods The serum samples from 89 patients with chronic HBV infection who visited the Second Affiliated Hospital of Chongqing Medical University from 201 1 to 2013 were collected.PCR was used to amplify the X gene of HBV DNA extracted from the serum samples.After sequencing,the HBV-X genome was compared with those reported in GenBank to find the variable sites and variant forms.Chi -square and one -way ANOVA were used for the statistical analysis afterwards,whereas genotypes were determined by the genotyping tool of the National Center for Biotechnology Information.Results All patients were genotype B or C.Among HBeAg-positive patients,46.2% were genotype B,and 53.8% were genotype C;among HBeAg-negative patients,81.2%were genotype B,and 18.8%were genotype C (P=0.001).PLC pa-tients had a significantly higher risk of mutation in the basic core promoter (BCP)region than the CHB and LC groups (69.2%vs 34.4%and 61.3%,P<0.05);in addition,an evident T-base deficiency was observed at nt1821 site (88.5% vs 53.1% and 71%,P=0.014).Among CHB and LC patients,those with genotype C had a significantly higher risk of BCP double mutation than those with geno-type B (61.5%vs 15.8%,P=0.007;83.3%vs 47.4%,P=0.045).The incidence of BCP double mutation was significantly higher in the low-viral load group (≤106 copies/ml)than in the high-viral load group (>106 copies/ml)(81.3% vs 47.9%,P=0.015). Conclusion The BCP double mutation and T-base deficiency at nt1821 site may play important roles in the development of PLC.

Systemic lupus erythematosus(SLE) is an autoimmune connective-tissue disorder with a wide range of clinical features.Although the clinical symptoms and immunological findings of pediatric SLE are similar to those of adult SLE patients,the pediatric SLE has some special aspects.In the recent years,although there was no abrupt development in the diagnosis and treatment of SLE,there were some new findings,maybe not mature.On the basis of recent findings outsides,this study drew attention to the advances in the novel clinical manifestation,update treatment of pediatric SLE.

5.Applications of microsatellite marker technology in the genetic structure research for closed colony of Brandt’ s voles

Objective Analysis of the genetic structure stability of Brandt ’ s vole ( lasiopodomys brandtii ) in closed colony using microsatellite marker technology.Methods Genomic DNA was extracted from tail tip using high-concentration-salt precipitation methods.Marked with fluorescent tags( Fam) , 7 microsatellite primers were filtered out by PCR, and the DNA structure of three consecutive generations of Brandt’ s vole was analyzed by microsatellite marker. Results By the analysis of the average heterozygosity and polymorphism information content, Brandt’ s vole populations maintaineda closed group of qualified genetic structure.Conclusions The present results show that the closed group of Brandt’ s vole species in our laboratory maintain a stable genetic structure.
6.Subtrochanteric Varization Osteotomy with Open Wedge Technic in a Legg-Calve-Perthes Disease

Subtrochsnteric varization osteotomy with open wedge technic was performed for the Legg-Calve-Perthes disease patients of 6 years to 9 years of age. Open wedge technic is less complicated with unwilling effect of postoperative leg shortening than closed wedge technic, but has seldom performed for the patients over 5 years of age because of possible delayed or nonunion of osteotomy site. Authors trial of this technic in a older patient(6 to 9 years of age) showed excellent post-operative result with noneventual post-operative course including delayed or nonunion.

8.Advances in the study of derivatization of ginsenosides and their anti-tumor structure-activity relationship.

Ginsenosides, belonging to a group of saponins with triterpenoid dammarane skeleton, show a variety of pharmacological effects. Among them, some ginsenoside derivatives, which can be produced by acidic and alkaline hydrolysis, biotransformation and steamed process from the major ginsenosides in ginseng plant, perform stronger activities than the major primeval ginsenosides on inhibiting growth or metastasis of tumor, inducing apoptosis and differentiation of tumor and reversing multidrug resistance of tumor. Therefore ginsenoside derivatives are promising as antitumor active compounds and drugs. In this review, the derivatization methods, ginsenoside derivatives and their anti-tumor structure-activity relationship have been summarized for providing useful information for the research and development of novel antitumor drugs.

10.The experiences of internship in nursing homes among college students of elderly service and management major: a qualitative research

Objective To explore the experiences of internship in nursing homes among college students of elderly service and management major. Methods Descriptive phenomenological methodology was adopted in this study, a semi-structured interview was conducted with 10 college students of elderly service and management major. Data were analyzed based on Colaizzi' s phenomenological research method. Results Six themes were extracted including: sense of worthlessness;heavy burden of work;respond actively and continuous learning;sense of satisfaction and achievement;the future is mixed;eager to pursue further study. Conclusions College students of elderly service and management major have heavy burden of work and feel worthlessness, they worry about the future, so educators in colleges, managers of nursing homes and national policy-makers should take targeted measures to stable talent team of old-age service.
