Chinese Journal of Gastroenterology 2015;(7):439-441
Gastroesophageal reflux disease(GERD)is a commonly seen digestive disease. So far, there is no golden standard for the diagnosis of GERD. The main diagnosis methods used in clinical practice include gastroscopy,24 h esophageal pH monitoring and proton pump inhibitor test, however, these methods are either invasive or complex in procedure and with low diagnostic accuracy rate. Gastroesophageal reflux disease questionnaire(GerdQ)possesses multiple advantages such as simple,non-invasive and easily acceptable. This article reviewed the use of GerdQ in diagnosis of GERD.

NHL proteins, which play important roles in regulation of cell proliferation and differentiation, have been extensively studied on mammals. Here, we cloned a member of NHL protein family namely BmBrat in silkworm. The full-length cDNA sequence of BmB rat was obtained by means of the rapid amplification of cDNA ends (RACE), including 3 614 bp. The ORF is 2 580 bp long, encoding a protein with 859 amino acid residues. The molecular weight is 94.3 kDa and the isoeledtric point (pI) is 6.65. The BmBrat expression profile was detected by RT-PCR at L5D3 larval stage, and it was expressed in all tissues, including silk gland, midgut, fat body and malpighian tubule. However, it was highly expressed in ovary and head. The expression profile was also detected at different stage of embryo development, and reached a peak at the 4th and 5th days of the embryonic period. Anti-BmBrat polyclonal antibody was generated f6llowing prokaryotic expression, protein purification and mice immunization, which is highly specific and effective for recognizing BmBrat protein through Western blotting and immunofluorescence staining. Subcellular localization of BmBrat in hemocytes revealed that it was specifically expressed in cytoplasm. This study provides a foundation for further research of the biological function of BmBrat gene.

Objective To explore the preventive effect of Bailing capsules(BL) on myocardial fibrosis in the viral myocarditis(VMC) mice models and to clarify the possible mechanism. Methods The models of VMC were established by injecting Coxsackie virus B3 (CVB3) solution into 50 BALA/C mice. The 31 mice were divided into model group(n =11),large dose BL group(n =10)and small dose BL group (n =10).At the same time the control group (n =10) was established. The mice in model group and control group were fed with water 1 mL/d,the mice in large dose BL group were fed with BL at a dose of 7.5 g/(kg · d),and the mice in small dose BL group were fed with BL at a dose of 2.5 g/(kg · d).After 60 d,the levels of angiotensin Ⅱ (Ang Ⅱ) in blood serum of mice in various groups were detected by using ELISA.Myocardium tissue of mice was stained by Masson and collagen volume fraction(CVF)was accounted. At the same time, the content of periostin mRNA and transforming growth factor β1 (TGF-[β1) mRNA in myocardium tissue of mice were detected by adopting reverse transcription(RT)-PCR.Results Compared with the control group, the levels of serum Ang Ⅱ,CVF,TGF-β1 mRNA and periostin mRNA were increased (all P < 0.01) in the model group.Compared with the model group, the levels of serum Ang Ⅱ,CVF and TGF-β1 mRNA were obviously decreased in large dose BL group and small dose BL group (all P < 0.05).Compared with the model group, the level of periostin mRNA was obviously decreased in large dose BL group (P < 0.05),but the level of periostin mRNA in small dose BL group was not decreased obviously (P > 0.05).Conclusions BL can offer some protection to myocardial fibrosis in VMC mice. The possible mechanism may be performed through inhibiting the expression of serum Ang Ⅱ and TGF-β1 in myocardium tissue to reduce the expression of periostin.

Objective A new type adsorbent for removal of bacterial ET was prepared by immobilizing L-serine covalently onto polyvinylidene difluoride membrane (PVDF-Ser) to study the effect of hemoperfusion on sepsis induced lung injuries in a pig model. Method Endotoxemia were induced in thirteen pigs by injecting endotoxin (1 μg/(kg·h) ) intravenously for 2 h. One hour later, the pigs were treated with absorber of PVDF-Ser volumn (group hemoperfusion) or empty adsorber (group control) for 2 h. Extravascular lung water indexed to body weight and alveolar capillary permeability, levels of serum endotoxin, tumor necrosis factor-α, interleukin-6, malondialdehyde, glutathione, myeloperoxidase activity, and lung histological changes and myeloperoxidase immunohistochemical analysis were measured. Results Hemoperfusion treatment with PVDF-Ser volumn significantly improved extravascular lung water, pulmonary alveolar capillary permeability and lung injuries ( P < 0. 05). In addition, hemoperfusion attenuated both serum and pulmonary cytokines, and decreased lung malondialdehyde and myeloperoxidase levels ( P < 0. 05). Conclusions Hemoperfusion with the adsorber of PVDF-Ser volumn effectively reduced inflammatory responses and improved lung injuries and oxidative stress in endotoxemia. It has a high potential of clinical application for treatment of patients with severe sepsis.
