1.Influence of rough extraction of QiZhu decoction on VEGF& KDR/flk-1 protein expression in MGC-803 cell

Objective To evaluate the influence Qizhu decoction on VEGF and its receptor KDR/flk-1 protein expression in MGC803 cell. Methods Four groups of MGC-803 Cells were established, and intervened with blank control, DMSO control, high dose Qizhu decoction, and low dose Qizhu decoction respectively. VEGF and KDR/flk-1 protein were detected by flow cytometry and western bloting. Results The value of VEGF expression was 120.0±10.8, 116.8±14.7, 95.0±12.5, and 108.4±13.5 respectively in each group. The difference between high dosage Qizhu decoction group and the bland control group was significant. Value of KDR/flk-1 were 10.4±3.5, 9.0±3.4, 6.8±2.3, and 6.8±3.5 in each group respectively. There was no significant difference among these 4 groups. The grayseale values of VEGF expression was 14.45±4.61, 12.32±3.27, 2.58±0.84, and 3.45±1.12 in each group respectively. There was significant difference between the QiZhu groups and the blank control group. Meanwhile grayseale values of KDR/flk-1 expression were 3.87±1.05, 3.55±1.32, 3.62±1.01, and 3.73±0.88 in each group respectively, showing no significant difference among 4 groups. Conclusion Rough extraction of QiZhu decoction down-regulated the protein expression of VEGF, but had no effects on the expression of KDR/flk-1.
2.Study on Therapeutic Effect and Mechanism ofQi-Zhu Er-Zhu Er-CaoTang in Treatment of Chronic Atrophic Gastritis Precancerous Lesion Based on Pathway of NF-κB (p65) and CyclinE

This study was aimed to observe the therapeutic effect and mechanism ofQi-Zhu Er-Zhu Er-Cao Tang(QZEZECT) in the treatment of chronic atrophic gastritis (CAG) precancerous lesion. A total of 56 clean grade healthy Wistar rats were randomly divided into 6 groups. In the negative control group (NC), model group,Wei-Fu-Chun(WFC) group,Ren-Zhu Jian-Wei Ke-Li(RZJWKL, RZ) group, there were 10 rats in each group. In the high-dose QZEZECT (QZ-H) group and low-dose QZEZECT (QZ-L) group, there were 8 rats in each group. CAG/PLGC model was established by MNNG in the model group, WFC group, RZ group, QZ-H group and QZ-L group. Intragastric administration of corresponding decoctions at the dose of 0.39 g·kg-1, 3.2 g·kg-1, 54.4 g·kg-1, 13.6 g·kg-1 were given to rats in the WFC, RZ, QZ-H and QZ-L groups once a day for 28 consecutive days, respectively. The same volume of normal saline was given to the NC group and the model group. Histomorphological changes of gastric mucous membrane in rats of each group were observed. Expressions of NF-κB/p65 and CyclinE protein were detected. The results showed that compared with the NC group, the degrees of infiltration and dysplasia and expressions of NF-κB/p65 and CyclinE significantly increased in the model group with statistical significance (P < 0.01). Compared with the model group, the expression of NF-κB/p65 and CyclinE in the WFC group reduced with statistical significance (P < 0.05). The expression of NF-κB/p65 and CyclinE in the RZ and QZ-L group obviously reduced with statistical significance (P < 0.01). The overall effective rates of WFC group, RZ group, QZ-H group and QZ-L group were 50%, 63.3%, 43.3% and 73.3%, respectively. It was concluded that QZEZECT can treat CAG precancerous lesion, which may take effect by improving and inhibiting pathologic changes of the transcription factor of inflammation.
3.Relationship between serum cystatin-C levels and vibrating perception threshold in patients with Type 2 diabetes mellitus.

OBJECTIVE:
To explore the relationship between serum cystatin-C (Cys-c) levels and vibrating perception threshold (VPT) in patients with Type 2 diabetes mellitus (T2DM).
METHODS:
According to the symptoms, signs and results of lab examination, a total of 352 patients with T2DM were divided into a diabetic peripheral neuropathy group (DPN group, n=107) and a non-diabetic peripheral neuropathy group (NDPN group, n=245). Serum Cys-c levels were measured by radioimmunoassay method. The relationship between serum Cys-c levels and VPT, estimated glomerular filtration rate (eGFR), urinary albumin-to-creatinine ratio (ACR), glucose and blood pressure and other parameters were also analyzed by correlation and multiple regression analysis. All T2DM patients were divided into a high Cys-c levels group (n=89) and a low Cys-c levels group (n=263) according to the upper quartile of Cys-c, and the incidence of DPN and VPT levels in each group were compared. Risk factors of DPN in T2DM patients were analyzed by binary logistic regression analysis and receiver operating characteristic (ROC) curve was used to identify the optimal cutoff of serum Cys-c levels for predicting DPN in patients with T2DM.
RESULTS:
Serum Cys-c levels were significantly higher in the DPN group than that in the NDPN group [(1.04±0.43) vs (0.80±0.25) mg/L, P<0.01]. Correlation analysis showed that serum Cys-c levels were positively correlated with age, body mass index (BMI), serum creatinine (SCr), blood urea nitrogen (BUN), ACR, VPT, pulse pressure (PP), white blood cell( WBC) , red cell distribution width (RDW), hemoglobin A1c (HbA1c) and fasting blood glucose (FBG) (r=0.410, 0.115, 0.613, 0.433, 0.291, 0.300, 0.156, 0.129, 0.282, 0.314, 0.236, respectively, P<0.05 or P<0.01); and negatively correlated with diastolic blood pressure (DBP), eGFR and indirect bilirubin (IBIL) (r=-0.135, -0.647, -0.114, respectively, P<0.05 or P<0.01). Serum Cys-c levels in T2DM patients were positively correlated with VPT after adjusting for gender, age, BMI, ACR and eGFR (r=0.235, P<0.01). Multiple regression analysis revealed that VPT, age, SCr, eGFR, PP, ACR and HbA1C were independent related factors affecting serum Cys-c levels in T2DM patients. Compared with those in the low Cys-c levels group, the prevalence rate and VPT value was increased in the high Cys-c levels group (all P<0.01). Binary logistic regression analysis found that age, Cys-c and HbA1C were independent risk factors for predicting DPN in T2DM patients (all P<0.01). ROC curve analysis revealed that the optimal cutoff of Cys-c to predict DPN in T2DM patients was 0.996 mg/L, the sensitivity was 43.9%, the specificity was 83.7%, and the area under curve was 0.663.
CONCLUSION
Serum Cys-c levels are well correlated with VPT in patients with T2DM. When the serum Cys-c levels>0.996 mg/L, the predicts have high risk of DPN in T2DM patients, which might be related to diabetic nephropathy, oxidative stress and inflammatory reaction induced by advanced age, hyperglycemia and hypertension.

4.Comparison Between Two Marking Methods in CT/MRI Image Registration

Objective To compare the results of CT/MRI registration by using two different marker materials. Methods CT and MR transversal images (using electric wire and thin tube filled with lohexol injection as matching points respectively) were obtained from nine cases with brain tumor. Another CT and MR transversal images were also obtained by using the cod-liver oil capsule filled with gadopentetate dimeglumine injection as matching points with the same scanning position. The images were transferred to the TPS for image registration and the effect was evaluated. Results The CT/MRI CRE range for using different kinds of marker material was 0.16-0.28cm,while was 0.06-0.16cm for using the same marker material. Conclusion Comparing with the method using different kinds of material,the method using the same spherical material can get better results in CT/MRI registration.

Objective: To explore the position change of the setup reference markers on the sternum due to the change of hands grip position. Methods: Six hands grip positions were simulated for 8 patients and the set-up reference points marked on the sternum skin were compared while the hands grasped different positions of the hands. Results: The setup markers on the sternum were deviated by all five hands grip positions. The maximum deviated distance was 14mm. Conclusion: The change of the hands grip position can affect the position of setup reference markers on the sternum of the patients and thus the isocenter of plan may be aligned to the wrong area.
6.Effect of loneliness on online deviant behavior of students in a police college: a moderated mediation model

ObjectiveTo explore the relationship among loneliness, Internet addiction and online deviant behavior of students in police colleges, and to discuss the related mechanism. MethodsIn March 2020, 766 students from a police college in Sichuan were selected by cluster sampling method, and assessed using University of California at Los Angeles-Loneliness Scale (UCLA-LS), Diagnostic Scale for Internet Addiction Disorder (DSFIAD) and College Students' Online Deviant Behavior Questionnaire, then SPSS 20.0 software was used for statistical, and Mplus 8.0 software was used to test the mediating and moderating effects. Results①The Online Deviant Behavior Questionnaire score of male students in police college was higher than that of female students (t=7.560, P<0.01). ②UCLA-LS score was positively correlated with DSFIAD score (r=0.406, P<0.01), and Online Deviant Behavior Questionnaire score was positively correlated with UCLA-LS and DSFIAD scores (r=0.279, 0.592, P<0.01). ③Loneliness could positively predict online deviant behavior (β=0.279, t=9.041, P<0.01). ④Internet addiction played a full mediating role between loneliness and online deviant behavior, with a mediating effect size of 0.233 (95% CI: 0.190~0.279, P<0.01), accounting for 83.51% of the total effect. ⑤Gender played a regulatory role in the second half (Internet addiction → online deviant behavior) of the intermediary path of "loneliness → Internet addiction → online deviant behavior" (β=-0.192, t=-4.461, P<0.01). ConclusionThe loneliness of police college students cause a direct impact on the online deviant behavior, and also pose an indirect impact on online deviant behavior through Internet addiction. Furthermore, loneliness has a moderated mediation effect on online deviant behavior, and is only significant in the male students.
7.Expressions of phospho-Erk1/2 and phospho-Akt1 in the corpus cavernosum of aged rats.

OBJECTIVENitric oxide (NO) is a key factor for penile erection. Its generation is mainly regulated by nitric oxide synthase (NOS), while both phospho-Erkl/2 (P-Erkl/2) and phospho-Aktl (P-Aktl) can affect the expression and activity of NOS and consequently penile erection. This experiment aimed to study the expressions of P-Erkl/2 and P-Aktl in the corpus cavernosum and their possible roles in erectile dysfunction in aged rats.
METHODSWe included 10 two-month-old male SD rats in Group A and another 10 eighteen-month-old ones in Group B, measured the levels of serum testosterone, and detected the expressions of P-Erkl/2 and P-Aktl in the corpus cavernosum by immunohistochemistry and RT-PCR.
RESULTSCompared with Group A, Group B showed a significantly decreased level of serum testosterone (9.57 +/- 1.57 nmol/L vs 4.73 +/- 0.94 nmol/L, P < 0.05), and remarkably increased mRNA expressions of P-Erk1 and P-Erk2 and protein expression of P-Erkl/2 (0.47 +/- 0.09, 0.61 +/- 0.11 and 7.50 +/- 1.81 vs 0.95 +/- 0.06, 0.92 +/- 0.05 and 32.09 +/- 8.45, P < 0.05). But there were no significant differences in the mRNA and protein expressions of P-Akt1 between the two groups (0.97 +/- 0.04 and 11.67 +/- 5.61 vs 0.94 +/- 0.05 and 10.93 +/- 3.06, P > 0.05).
CONCLUSIONThe overexpression of P-Erk1/2 in the corpus cavernous may be one of the important mechanisms of aging-related erectile dysfunction.

8.Raf/MEK/ERK1/2 signaling pathway and penile erection.

Penile erection is regulated by the relaxation of corpus cavernosum smooth muscle cells (CCSMCs). It has been recognized that the Ras/MEK/ERK1/2 signaling pathway is closely related to the functions of CCSMCs and endothelial cells, and it is involved in the regulation of penile erection, mainly via phosphorylation of NO synthase. ERK1/2 phosphorylates, inhibits eNOS activity, and thus reduces the relaxation of CCSMCs and penile erection. But the site of phosphorylation is not yet clear. In CCSMCs and endothelial cells, the ERK1/2 pathway interacts with other cascades and regulates the erectile function of the penis. This article presents an overview of the researches on the ERK1/2 signaling cascade, its regulatory role and its interaction with other signaling pathways in penile erection.

9.The progressive effects of chronic intermittent hypoxia on cognitive function and the cholinergic neuron in rats.

OBJECTIVETo investigate the relation between the progressive effects of chronic intermittent hypoxia (CIH) on cognitive function and the change of cholinergic neuron.
METHODSForty adult male Sprague-Dawley rats were randomly averagely divided into four groups: control group, CIH 1 week group, CIH 3 week group and CIH 5 week group. The cognitive function was assessed by the Morris Water Maze. The necrosis neurons in prefrontal cortex and hippocampus were observed and counted. The cholin acetyltransferase (ChAT) immunostained cells in prefrontal cortex and hippocampus were identified and quantitated.
RESULTSThe spatial learning and memory impairments progressed from 1 to 5 5 weeks in rats. Compared with the control group, the cognitive impairments in CIH5w group were significant (P < 0.05). The degeneration or necrosis neurons in prefrontal cortex and hippocampus were significantly increased in CIH rats, and worsen gradually along with the hypoxia. The ChAT immunostained cells in prefrontal cortex and hippocampus were gradually reduced. The ChAT immunostained cells of prefrontal cortex and hippocampus in CIH3w group and CIH5w group were less than that in control group (P < 0.05).
CONCLUSIONChronic intermittent hypoxia induced slowly progressive spatial learning and memory impairments in rats, which maybe associated with the damage of neurons and the reduction of ChAT in prefrontal cortex and hippocampus.

10.Efficacy of incision and drainage versus percutaneous catheter drainage in treatment of severe acute pancreatitis complicated by pancreatic abscess

ObjectiveTo investigate the clinical features and treatment of severe acute pancreatitis (SAP) complicated by pancreatic abscess (PA). MethodsThe clinical data of 17 SAP patients with PA who were admitted to Affiliated Hospital of Luzhou Medical College from January 1, 2005 to August 25, 2015 were analyzed retrospectively. The clinical manifestations, therapeutic methods, and outcome were summarized. ResultsOf all the 17 patients, 12 patients underwent surgical operation, among whom 9 were cured, 1 experienced postoperative intestinal fistula, and 2 experienced recurrence of abscess and underwent the surgery again (1 died of multiple organ failure), and the mean hospital stay was (108.29±52.37) d; 5 patients underwent percutaneous catheter drainage, among whom 4 were cured, and 1 underwent surgical treatment due to inadequate drainage, and the mean hospital stay was (53.03±6.71) d. ConclusionAdequate drainage should be performed once a confirmed diagnosis of PA is made, and appropriate drainage methods should be selected based on the patient′s actual condition. Minimally invasive treatment has a good effect, a short length of hospital stay, and few complications, and holds promise for clinical application.
