1.Correlation between exposure to polycyclic aromatic hydrocarbons during pregnancy and placental mitochondrial DNA copy number

Objective:
To examine the correlation between exposure to polycyclic aromatic hydrocarbons ( PAHs ) and placental mitochondrial DNA ( mtDNA ) copy number among pregnant women, so as to provide the evidence for evaluation of potential effects of PAHs exposure during pregnancy on offspring health.
Methods:
A total of 200 pregnant women delivered at a tertiary hospital in Urumqi City during the period from January to October 2019, and their newborns were recruited, and grouped according to the time of delivery, including the heating group [delivery during the heating period ( from January to April ) ] and the non-heating group [delivery during the non-heating period (from July to October) ]. Subjects'age, ethnicity, educational level and type of home heating were collected, and the total concentration of 16 PAHs was determined in the blood samples of pregnant women and their babies using gas chromatography-mass spectrometry. Placental DNA was extracted, and placental mtDNA copy number was measured using real-time fluorescence quantitative PCR assay. In addition, the correlation between PAHs concentration and placental mtDNA copy number was examined using the Spearman rank correlation analysis.
Results:
There were 100 subjects in the heating group, which had a median age of 29 ( interquartile range, 3 ) years and had a mean gestational age of ( 275.06±0.72 ) days, and there were 100 subjects in the non-heating group, which had a median age of 29 ( interquartile range, 4 ) years and had a mean gestational age of ( 276.82±0.66 ) days. The total concentration of PAHs in the blood of pregnant women [15.71 (4.30) vs. 12.98 (5.49) μg/L; P<0.05 ], the total concentration of PAHs in neonatal blood [ 14.29 (4.25) vs. 11.24 (5.09) μg/L; P<0.05 ] and the placental mtDNA copy number [4.67 (1.18) vs. 4.51 (0.62); P<0.05] were all higher in the heating group than in the non-heating group. Spearman rank correlation analysis revealed that the total concentration of PAHs in the blood of pregnant women and neonates was positively correlated with placental mtDNA copy number ( rs=0.240, P=0.001; rs=0.273, P<0.001 ), and the total concentration of PAHs in the blood of pregnant women was positively correlated with the placental mtDNA copy number in the heating group ( rs=0.245, P=0.014 ), while the PAHs concentration in the neonatal blood was positively correlated with the placental mtDNA copy number in the non-heating group ( rs=0.292, P=0.003 ).
Conclusions
Exposure to PAHs positively correlates with placental mtDNA copy number among pregnant women, and there is a correlation between maternal exposure to PAHs and neonatal oxidative stress.
2.Effects of Tongxin Plaster on Serum Macrophage Colony Stimulating Factor, Homocysteine and Granules Membrane Protein-140 in Patients with Coronary Heart Disease

Objective To study the effects of Tongxin Plaster on serum macrophage colony stimulating factor (M-CSF), homocysteine (HCY), and granules membrane protein-140 (GMP-140) of patients with angina pectoris of coronary heart disease. Methods Eighty CHD patients were randomly divided into two groups. Forty-one cases in treatment group were given acupoint application of Tongxin Plaster on acupoint Tanzhong (CV 17) and Xinshu (BL 15) 12 hours a day. Thirty-nine cases in control group were given orally Shexiangbaoxin Pill, 2 pills twice a day. The treatment course of the two groups was 3 weeks. The serum levels of M-CSF, HCY and GMP-140 were detected respectively. Results After treatment, the levels of M-CSF, HCY, GMP-140 in both groups were all decreased significantly (P 0.05). Conclusion Tongxin Plaster has good effects on antagonizing inflammatory response of coronary artery, inhibiting activation of platelets and degrading the level of homocysteine, which may be one of the mechanisms for Tongxin Plaster in treating CHD.
3.Effect of lipopolysaccharide on toll like receptor 7 and toll like receptor 9 in AR42J cell lines

Objective To investigate the roles of toll like receptor7 (TLR7) and toll like receptor 9 (TLR9) in the pathogenesis of acute pancreatitis.Methods AR42J cells were treated by lipopolysaccharide at different dosages (0,1,10,100 mg/L),and cell model of acute pancreatitis in vitro was established.AR42J cells without lipopolysaccharide treatment were as control.Cells and culture supernatant were collected after 24 hours cultivation.TLR7,TLR9 mRNA and protein expressions were detected by RT-PCR and Western Blot,and levels of TNF-α,IL-10 in culture supernatant were measured by ELISA.Results The TLR 7 mRNA expression levels in control group,1,10,100 mg/L lipopolysaccharide group were 0.12 ± 0.09,0.28 ± 0.06,0.49 ± 0.04,0.78 ± 0.04,and the TLR9 mRNA expression levels were 0.06 ± 0.02,0.32 ± 0.03,0.56 ± 0.14,0.84 ± 0.12; the TLR7 protein expression levels were 0.04 ± 0.01,0.26 ± 0.05,0.49 ±0.04,0.77 ±0.16,and the TLR9 protein expression levels were 0.10 ±0.14,0.62 ±0.23,1.21 ± 0.26,1.75 ± 0.13 ; the TNF-α levels in culture supernatant were (8.01 ± 5.32),(25.64 ± 8.71),(49.06 ± 10.23),(75.83 ± 6.65) ng/L,and the IL-10 levels were (155.54 ± 25.47),(105.16 ± 10.49),(69.36 ± 8.19),(14.07 ± 9.06)ng/L.The expression levels of TLR7 and TLR9's mRNA,protein in cell,as well as the levels of TNF-α in culture supernatant increased with the lipopolysaccharide concentration,while the levels of IL-10 in culture supernatant decreased with the lipopolysaccharide concentration,and the difference among these groups was statistically significant (P < 0.01).Conclusions The expressions of TLR7 and TLR9 in AR42J cells treated by using lipolysaccharide are obviously up-regulated,and it suggests that TLR7 and TLR9 may be vital in the pathogenesis of acute pancreatitis.
4.Affect of living quality from indapamide vs nifedipine in treating mild to moderate hypertension

AIM: To study the effects of indapamide and nifedipine in treating the patients with mild to moderate hypertension and the affect to the living quality. METHODS: One hundred and twenty seven patients with mild to moderate hypertension were divided into two groups, indapamide group 67 patients (M 30, F 37; age 63 a± s 12 a) received indapamide 2.5 mg, po, tid ×2 mo; nifedipine group 60 patients (M 30, F 30; age 61 a±12 a) received nifedipine 10 mg, po, tid× 2 mo. RESULTS: Both above medicines could reduce blood pressure effectively (P>0.01), and there was no obvious affect to the lipid, renal function, uric acid, electrolyte. Indapamide group had the results of getting living quality improved; while nifedipine group had that of increasing some self testing sonse symptoms and ling quality reduced. CONCLUSION: Both indapamide and nifedipine have definite effects in treating hypertension, while no obvious bad affect on blood lipid, renal function, et al. But the indapamide is less adverse reaction, and superior to nifedipine on the aspect of improving living quality.

6.Assistant Effect of Therapy of Strengthening Qi,Warming Yang,Activating Blood to Promote Diuresis for Congestive Heart Failure

【Objective】 To investigate the synergistic action of therapy of strengthening Qi,warming yang,activating blood to promote diuresis on routine western medicine for congestive heart failure(CHF) and to observe its effect on neuroendocrine factors.【Methods】Seventy CHF patients with cardiac function of grade Ⅱ~Ⅳ which were classified into Qi deficiency and blood stasis complicated with fluid retention,were randomized into groups A(n=42) and B(n=28).Group B received routine treatment with digoxin,diuretics,angiotensin converting enzyme inhibitors and ? receptor antagonists.Group A additionally received oral use of decoction of herbal medicines of Radix Astragali,Herba Epimedii,Herba Leonuri,Rhizoma Alismatis,etc.,which has the actions of strengthening Qi,warming yang,activating blood to promote diuresis.Four weeks constituted one treatment course.The therapeutic effect as well as the changes of cardiac function and neuroendocrine factors before and after treatment was observed.【Results】In group A,24 were markedly effective,16 effective,2 ineffective and the total effective rate was 95.24%;in group B,19 were markedly effective,4 effective,5 ineffective and the total effective rate was 82.14%.The therapeutic effect in group A was superior to group B(P
7.Relationship between Fractional Anisotropy of Corticospinal Tract and Motor Function in Stroke Patients Using Diffusion Tensor Imaging

Objective To explore the relationship between fractional anisotropy (FA) of corticospinal tract (CST) and motor function in stroke patients using diffusion tensor imaging (DTI). Methods From January, 2013 to October, 2016, 36 stroke patients were evaluated with Fugl-Meyer Assessment (FMA), Japan Upper Limb Function Test and DTI before and eight weeks after rehabilitation. FA in posterior limb of internal capsule, cerebral peduncle and pons were obtained, including ipsilesional and contralesional CST. Results The FA values within three sections of CST were significantly lower in the ipsilesional side than in the contralesional side both before and after rehabilitation (t>5.330, P<0.001). In posterior limb of internal capsule, the FA value increased after rehabilitation (t=-3.415, P<0.05), and no significant dif-ference was found in the other two sections (t<1.653, P>0.05) in the ipsilesional side, as well as in all the sections in the contralesional side (t<1.021, P>0.05). The FA values were positively correlated with the scores of FMA and Japan Upper Limb Function Test in all the sections both before and after rehabilitation (r>0.43, P<0.05), especially the relationship between the FA value and the score of Japan Upper Limb Function Test in the posterior limb of internal capsule (r=0.67). Conclusion DTI can be used to evaluate CST injury and the relationship be-tween CST injury and motor function.
8.Establishment and effect of oncological training base for professional nurse

Objective To investigate the establishment and effect of the oncological training base for professional nurses in Xinjiang Uygur Autonomous Region.Methods The oncological training base for professional nurses in Xinjiang was established to train 179 nursing specialists.Result One hundred and seventy-nine participants passed the qualification examinations,achieving an average score of (77.1±5.8)in the theoretical examination and(78.0±8.9)in the skills examination.Conclusions The establishment of oncological training base for professional nurses is effective in improving the professional knowledge and operational levels of professional nurses on oncology and promoting the development of oncological nursing.
9.Effect of Tongmai Liquid on ET,IL-1 8 and M-CSF in patients with coronary heart disease

AIM:To observe the effect ofTongmai Liquid on endothelin(ET),interlenkin-18(IL-18),macrophage colony stimulating factor(M-CSF)in patients with coronary heart disease(CHD).METHODS:Eighty patients with coronary heart disease accompanied by heart and spleen qi deficiency and qi stagnanty+phlegm stasis and were randomly assigned into test group and control groups.Forty patients in test group were treated with Tongmai Liquid,30 mL,thrice per day,Forty patients in control group were treated with Shexiang Baoxin Pills,2 pills every time,thrice per day.The therapeutic course for both groups was 30 days.The blood-serum ET,IL-18 and M-CSF had been tested before and after the treatment.RESULTS:The levels of blood-serum ET,IL-18 and MCSF in the two groups decreased significantly after treatment(P<0.05).There's significant difference in two groups'blood-serum of ET(P<0.05),There're no significant diffference in two groups'blood-serum level IL-18 and of M-CSF(P>0.05).CONCLUSION:Tongmai Liquid may decrease blood-serum level of ET,IL-18 and M-CSF in patients with coronary heart disease without side effect.
10.Expression of adrenomedullin in hepatic tissue of rats with acute necrotizing pancreatitis associated hepatic injury

Objective To observe the expression of adrenomedullin (ADM) mRNA in hepatic tissue of rats with acute necrotizing pancreatitis (ANP) complicated with hepatic injury.Methods Sixty-four SD rats were randomly divided into control group and ANP group with 32 rats in each group.In ANP group,ANP model was induced by retrograde injection of 5% sodium taurocholate into biliopancreatic duct of rats.Rats in control group only received sham operation and pancreas manipulation.All the rats were sacrificed at 3,6,12,24 h after the operation.The serum levels of amylase,alanine aminotransferase (ALT),aspartate aminotransferase (AST) and ADM were detected.Pathological changes in pancreatic and hepatic tissue were examined.The expressions of ADM mRNA in hepatic tissue were evaluated by fluorescence quantitative PCR.Results The serum concentrations of amylase,ALT,AST were (7229 ±968),(174.2 ±28.0),(657.7 ± 139.0) U/L,which were significantly higher than those in control group [(2036 ± 292),(104.3 ± 22.1),(419.7 ± 86.3) U/L],and the difference between the two groups was statistically significant (P < 0.05 or P <0.01).Pathological injury of pancreas and liver tissue in ANP gradually aggravated with time,and the pathological scores at 12 h were (11.60 ± 1.51),(2.60 ± 0.89),which were significantly higher than those in control group (1.20 ± 0.77,0),and the difference between the two groups was statistically significant (P<0.01).The serum concentrations of ADM in ANP group increased at 3 h after ANP induction,and reached (38.53 ± 6.25)pg/ml at 12 h,which was significantly higher than that in control group [(28.99 ±3.92)pg/ml] ; the concentrations of ADM in liver tissue increased at 3 h after ANP induction,and reached (3.00 ± 1.49) at 6 h,which was significantly higher than that in control group (1.04 ± 0.20),and the difference between the two groups was statistically significant (P<0.05 or P<0.01).Conclusions The expression of ADM mRNA in rat 's hepatic tissue increases in the early stage of ANP,and the serum concentration of ADM also increases.
