1.Safety management for tissue engineering laboratories in universities

BACKGROUND:Tissue engineering is an interdisciplinary field and its experimental safety covers a wide scope along with the increase in accidents of tissue engineering laboratories, the safety management in tissue engineering laboratories in universities has become an important public health problem. OBJECTIVE:To enhance the safety management strategies for tissue engineering laboratories in chinese universities and improve the safety management system. METHODS:PubMed, Elseveir, WanFang and CNKI databases were retrieved for the literatures concerning the biosafety, safety management of hazardous chemicals, waste, medical information, instrument operation and human body mechanics in tissue engineering laboratories, and then the status of laboratory management was analyzed. RESULTS AND CONCLUSION:The status of safety management in tissue engineering laboratories in China is analyzed based on the technique characters and regulations on safety management. Management strategies for laboratory safety have been proposed according to the differences in relevant laboratory safety management between China and the US, such as regular training on laboratory safety, improving the safety consciousness of experimental staff, constructing the strict system access to laboratory, improving safety facilities, standardizing the experimental safety manual and appointing specialized safety management committee. Comprehensive and systematical regulations on laboratory safety management will ensure the smooth running of tissue engineering laboratories in universities, thus accelerating the development of tissue engineering.
2.Correlation of Immune Infertility with Chronic Genital Tract Inflammation and Intervention of Chinese Medicine

【Objective】To explore the correlation of antisperm immune infertility(AII) in females with chronic genital tract inflammation and to observe the therapeutic effect of Chinese medicine of Qingshi Huayu Mixture(QHM) and Yishen Huoxie Prescription Ⅰ(YHPⅠ) for AII.【Methods】Fifty-eight AII patients were screened from 200 infertility patients,and were randomized into three groups.Group A(N=18) was treated with YHPⅠ,group B(N=20) with QHM and group C(N=20) with western medicine.Three months constituted one treatment course.The incidence of positive antisperm antibody(AsAb+)combined with chronic genital tract inflammation,therapeutic effect,pregnancy rate,the content of serum AsAb,as well as the changes of lymphocyte subtypes,natural killer(NK) and interleukin 6(IL-6) were observed.The correlation of the above indexes with serum AsAb content was also analyzed.【Results】(1) The incidence of positive antisperm antibody(AsAb+)combined with chronic genital tract inflammation: There were 96(48%)with AsAb+ in 200 patients,and 62(64.58%) with AsAb+ in 122 combined with chronic genital tract inflammation;there were 34(35.42%)AsAb+ patients in 78 without inflammation,the difference of AsAb+ incidence was significant between the inflammation patients and the non-inflammation patients(P0.05).There were 2 in 5 cured patients of group A,2 in 7 of group B and 1 in 7 of group C being pregnancy.(3) Content of serum AsAb: serum AsAb content was decreased in the three groups after treatment,the difference being significant as compared with that before treatment(P0.05).(4) The changes of lymphocyte subtypes,NK and IL-6: CD8+ was increased,CD4+/CD8+ and NK were decreased in groups A and B,the differences being significant as compared with those before treatment and those in group C after treatment(P
3.Immune enhancing effect of arginine on patients with chronic obstructive pulmonary disease complicated with malnutrition.

Objective To study the immune enhancing effect of arginine on patients with chronic obstructive pulmonary disease (COPD) complicated with malnutrition.Methods Thirty cases with COPD accompanied by malnutrition received nutritional support of total calorie intake of 1 5 times of resting energy expenditure (REE).Group A(n=12) received oral administration of 30g L-arginine per day as supplementation;Group B(n=18) were control subjects.Results The nutritional parameters such as TSF(triceps skinfold) thickness,PAlb(prealbumin),CHI(creatine height index) were markedly improved in Group A than Group B,TNF-?,CD 8 decreased and IL-2,NO(nitric oxide),IgG and CD 4 increased significantly in Group A(P
4.Study on the Thermic Effect of Food of Patients With Chronic Obstructive Pulmonary Disease

Objective To study on the thermic effect of food (TEF) of patients with chronic obstructive pulmonary disease(COPD).Methods Basal energy expenditure (BEE) of 12 male cases and resting energy expenditure (REE) of 11 male cases of COPD were detected by indirect calorimetry with metabolic cart. There were no significant difference in sex, age, height and body weight between two groups' e.g:TEF=REE-BEE.Results BEE and REE were 1114?208kcal/d,1433?136kcal/d respectively, the latter was higher 28% than BEE.Conclusion TEF increases significantly which may contributes to malnutrition and non-response to nutritional support in patients with COPD.
5.High-risk human papillomavirus infection among rural women of Jing'an County in Jiangxi Province:a cross-sectional study

Objective:To investigate the prevalence of cervical infection with high-risk human papillomavirus (hr-HPV) among the rural women of Jing-an County in Jiangxi Province. Methods:A population-based cross-sectional study was conducted among rural women of ages 35 years to 59 years in Jing-an County between May 2012 and November 2012. A total of 4 673 women were enrolled. Hybrid capture2 (hc2) was detected in 2 491 women, whereas HybriMax was detected in 2 182 women to understand the different hr-HPV types. Results:The overall hr-HPV prevalence was 11.9%. Multiple-type infections accounted for 41.5%. The most commonly seen hr-HPV type was HPV52 (2.8%), followed by HPV58, HPV16, and HPV39. No significant difference was observed in the preva-lence of hr-HPV infection between women in the mountainous and the plain areas in Jing-an County. However, the age distribution of HPV infection significantly differed (P<0.001). Conclusion:Hr-HPV infection rate in Jing-an County was close to that of other areas in China. HPV52 and HPV58 appeared to be the most prevalent HPV types and should be employed as a basis for HPV prevention in this area.
6.Effect of anterior cruciate ligament rupture on the strain of medial collateral ligament

Objective To explore the effect of anterior cruciate ligament (ACL) rupture on the biomechanics of medial collateral ligament (MCL). Methods Six fresh human knee joints were fixed on the self-made fixture and flexed 90°. ACL was divided into a normal group, an anteromedi-al bundle(AMB) break group, and an ACL complete break group. 800N burden was loaded, and the MCL strain data were compared among the 3 groups. Results The MCL strain was significantly increased in the AMB break group and the ACL complete break group than that in the normal group (P<0.05). There was no significant difference in the MCL strain between the AMB break group and the ACL complete break group (P>0.05). Conclusion ACL rupture may cause significant changes in the MCL biomechanics, but the effect of AMB break and ACL complete break on MCL has no significant difference.
7.Effect of electroacupuncturing Shuigou and Baihui on activation of microglia in brain of ischemia-reperfusion rats

BACKGROUND: Microglia is the immune effect cell in central nervous system, if it is activated, it will release neuron poisoning factors and inflammatory factors, and bring fatal injury.OBJECTIVE: To investigated the effect of focal cerebral reperfusion on activation of microglia and the effect of the regulation of electroacupuncturing Shuigou and Baihui.DESIGN: A randomized and controlled study.SETTING: The department of Physioltherapy, the 458 Hospital of Chinese PLA; Dongfang Hospital Affiliated to Shanghai Tongji University; The department of Histology-embryology, Tongji Medical College of Huazhong University of Science and Technology.MATERIALS: This experiment was processed at Histology-embryology Center of Tongji Medical College of Huazhong University of Science and Technology in March 2002. Total 80 Wistar rats were randomly divided into 8 groups with 10 in each group.METHODS: ① Normal control group: The rats were not given any treatment and killed next day. ② Sham operation group: Only artery was separated, no suture inserted, killed next day. ③ 6, 12 and 24-hour reperfusion groups: The rats' right middle cerebral artery (MCA) ischemia-reperfusion model was made by embolus suture technique, then the rat was killed after 30 minutes' occlusion at MCA and reperfusing respectively 6, 12, 24hours. ④ 6-hour reperfusion + electroacupuncture group: Electroacupuncture was carried out immediately after the model was made, then the rat was killedafter 30 minutes' occlusion at MCA and reperfusion 6 hours (The frequency was 4 Hz-16 Hz. Stimulus intensity increased 1 V per 10minutes and the final intensity was 3V. The stimulus was lasted 30 minutes). ⑤ 12-hour reperfusion + electroacupuncture B group: Electroacupuncture was carried out immediately after the model was made and 8 hours later, then the rat was killed after 30 minutes' occlusion at MCA and reperfusion 12 hours. ⑥ 24-hour reperfusion + electroacupuncture group: Electroacupuncture was carried out immediately after the model was made and 8, 16 hours later, then the rat was killed after 30 minutes' occlusion at MCA and reperfusion 24 hours.MAIN OUTCOME MEASURES: To calculate numbers of microglia and to observe its morphological changes.RESULTS: Data of totally 67 rats was entered the results analysis. No microglia was seen in the normal and sham operation groups. Large quantity of microglia was activated at the border of ischemic area in 6, 12 and 24reperfusion group, their quantity was largely increased, reaching apex at reperfusion 12 hours [(35.38±1.77), (54.25±1.67), (49.29±2.21)/200sights]. The quantity in every 6, 12 and 24 reperfusion + electroacupuncture group was less than that in model group [(32.11±2.80), (50.88±2.64),(45.45±3.95)/200 sights, P < 0.05].CONCLUSION: The microglia in brain is activated after the cerebral ischemia-reperfusion and induces toxic effect on neurons. The electroacupuncture can decrease activation of microglia so as to protect the neurons.

10.Role of bone marrow mesenchymalstem cells in recoveryprocess of hepatocyte injury

Objective To study the role of bone marrow mesenchymalstem cells in recovery process of hepatocyteinjury and to explore the possible mechanism. Methods BMSCs and primary generation of hepatocytes were cultured and identified. A model of hepatocyte injure was established. Liver cell proliferation at different conditionswas detected by MTT method. Hepatocytes were added with BMSCs,baxinhibitor and mixture of BMSCs and baxinhibitor for co?cultured.Expressions of TGF?β1,bcl?2,and baxwere detected by Westernblot and real?time PCR. Results BMSCs and primary generation of hepatocyte were successfully cultured and identified, and a model of hepatocyte injury was successfully established. MTT tests revealed that the OD value of the mixture wassignificant higher in BMSCs and HGF co?culture group than in BMSCs co?culture group or HGF co?culture group;there was no significant difference between BMSCs co?culture group and HGF co?culture group,while both groups were significant higher than control group. The MTT tests also revealed that the OD value of the mixture of BMSCs and Baxinhibitor co?culture group had no significant difference as compared with BMSCs co?culture group or Baxinhibitor co?culture group. There was no significant difference between BMSCs co?culture group and Baxinhibitor co?culture group,while the OD value was significant higher in both groups than in the control group. Westernb lot and real?time PCR results revealed that the value of TGF?β1 and bax in the mixture of BMSCs and Baxinhibitor co?culture group had no significant difference as compared with BMSCs co?culture group or Baxinhibitor co?culture group. There was no significant difference between BMSCs co?culture group and Baxinhibitor co?culture group,while both groups had a significant higher value than the control group. The value of Bcl?2 in the mixture of BMSCs and Baxinhibitor co?culture group had no significant difference as compared with BMSCs co?culture group or Baxinhibitor co?culture group. There was no significant difference between BMSCs co?culture group and Baxinhibitor co?culture group,while both groups had a significant lower value than the control group. Conclusions BMSCs can promote hepatocyte proliferation during the recovery process of hepatocyte injury,BMSCs and HGF promote liver cell proliferation have synergy effect,the effect of BMSCs probably through regulate TGF?β1/bcl?2(bax)correlation pathway.
